  • ||||||||||  Review, Journal:  Human Borrelia miyamotoi Infection in North America. (Pubmed Central) -  Apr 28, 2023
    Antibiotics are effective in clearing infection and are the same as those used for Lyme disease, including doxycycline, tetracycline, erythromycin, penicillin, and ceftriaxone. Preventive measures include avoiding areas where B. miyamotoi-infected ticks are found, landscape management, and personal protective strategies such as protective clothing, use of acaricides, and tick checks with rapid removal of embedded ticks.

    Journal:  Caveats of Using Overexpression Approaches to Screen Cellular Host IFITM Proteins for Antiviral Activity. (Pubmed Central) -  Apr 28, 2023
    Previously, we used a doxycycline-inducible overexpression system, in conjunction with approaches to modulate the expression of endogenous protein, to demonstrate the antiviral activity of IFITM1, IFITM2, and IFITM3 against influenza A virus (IAV) but not parainfluenza virus-3 (PIV-3) in A549 cells...We propose that exceedingly high levels of overexpressed IFITMs may not accurately reflect the true function of endogenous protein, thus contributing to discrepancies when attributing the antiviral activity of individual IFITM proteins against different viruses. Our findings clearly highlight the caveats associated with overexpression approaches used to screen cellular host proteins for antiviral activity.

    Journal:  Role of Doxycycline as an Osteoarthritis Disease-Modifying Drug. (Pubmed Central) -  Apr 28, 2023
    Current evidence does not favor the regular use of doxycycline for the treatment of osteoarthritis as an individual treatment option or in combination with others. However, multicenter large cohort studies are warranted to determine the long-term benefits of doxycycline.

    Journal:  Assessment of Heavy Metals Contamination and Antimicrobial Drugs Residue in Broiler Edible Tissues in Bangladesh. (Pubmed Central) -  Apr 28, 2023
    Irrespective of the source, broiler meat was found to contain antimicrobial drugs and heavy metals residues below the MRL, except for lead, suggesting that broiler meat is safe for human consumption. Therefore, raising public awareness regarding misconceptions about broiler meat consumption among consumers would be warranted.

    LentiVEX Packaging Cell Line v2.0 Yields Comparable LVV Titres to the 4-plasmid Process (Board No. 794) -  Apr 21, 2023 - Abstract #ASGCT2023ASGCT_1368;
    It only requires transfection of the LV genome plasmid with doxycycline induction to achieve LVV yields of 2 x 108 TU/mL (with eGFP as Gene of Interest), comparable to the four-plasmid process with WXATUS0028...Scale-up vector production and process development with the packaging cell line are ongoing, with the Master Cell Bank (MCB) being generated for GMP-standard vector manufacturing. With a reduced plasmid cost and a simplified transfection process, the LentiVEX Packaging Cell Line v2.0 is a flexible platform to support both early-phase discovery studies with easy screening of multiple GOIs and large-scale LVV manufacture.

    Inhibition of c-MYC in Hepatocellular Carcinoma via AAV8 Mediated Gene Transfer (Board No. 600) -  Apr 21, 2023 - Abstract #ASGCT2023ASGCT_1132;
    Expression of Omomyc in HCC cell lines in vitro by transient transfection and doxycycline induced expression induces an 8-20 fold inhibition of proliferation depending on cell type, including G2 cell cycle arrest (about 50%) in the high c-MYC expressing cell line Hep3B...Our preliminary data suggests that this could be due to induction of autophagic cell death, with mitigation of the effect seen when treated with Bafilomycin A1 or Chloroquine...We have also demonstrated a potential synergistic effect of combining autophagy inducing compounds like sorafenib and mTOR inhibitors being able to increase transduction efficiency by approximately 50-60% in our HCC cells lines. Taken together, our data suggest that our novel AAV8 delivery of Omomyc has the ability to inhibit cellular proliferation, induce autophagy, and induce cell death in HCC which may prove to be a novel therapeutic approach for the treatment of HCC, especially when combined with conventional therapeutics.

    Preclinical, Journal:  Acute generation of reactive oxygen species that induced by doxycycline pretreatment results in rapid cell death in polyphyllin G-treated osteosarcoma cell lines. (Pubmed Central) -  Apr 18, 2023
    Co-administration of the ROS inhibitor NAC reversed the suppressed cell viability and colony-forming ability, and abolished the increased level of ?-H2A.X in the cells with the combined treatment, indicating that the enhanced ROS was involved in the anti-proliferative effect of the combined treatment. Overall, the results demonstrated that doxycycline may function as chemosensitizers by inducing an acute and lethal ROS production to enhance cytotoxic of polyphyllin G in osteosarcoma cell lines, and the combined use of drugs may provide an alternative thinking for the development of new therapeutic agents.

    Antigen Binding Domain Influences CD47-CAR T Cell Function in Both Constitutively Active and Inducible Systems (Board No. 614) -  Apr 6, 2023 - Abstract #ASGCT2023ASGCT_595;
    Despite limited expansion, B6H12-CAR T cells exert antitumor activity in vitro and, consistent with CV1-CAR T cells, lack antitumor activity in vivo likely due to macrophage mediated elimination.Given that multiple constitutively active CD47-CAR T cell products downregulate CD47 and fail to function in vivo, we generated CD47-CARs under control of a doxycycline inducible (Tet-On) promoter that constitutively express ZsGreen (ZsG) as a detection marker (indB6H12- and indCV1-CAR)...Overall, these results demonstrate the CAR binding domain influences CD47-CAR T cell functional characteristics in both constitutive and inducible systems.While studies are ongoing, we demonstrate that 1) similar to CV1-CAR T cells, B6H12-CAR T cells downregulate CD47 surface expression and lack antitumor activity in vivo and 2) CD47-CAR binding domain determines CAR surface expression upon activation in an inducible system. We speculate that CAR affinity or binding site on CD47 is responsible for differing CD47-CAR T cell functional phenotypes and studies are underway to elucidate mechanisms of these findings.
