Cardiovascular studies in dogs and toxicology studies in rats and dogs revealed no adverse effects at doses (50 mg/kg) that achieved plasma concentrations >10-fold above the efficacious concentration. A-1574083 (ABBV-4083) shows potential as an anti-Wolbachia macrolide with an efficacy, pharmacology, and safety profile that is compatible with a short-term oral drug course for treating lymphatic filariasis and onchocerciasis.

Clinical, Journal: Safety of the Subconjunctival injection of Doxycycline in Rabbits. (Pubmed Central) - Feb 24, 2020
There was no visible sign of inflammation or necrosis, and also no histological change in both clinical and histopathological examinations. Subconjunctival injection of doxycycline with different dosages of 250 to 2000 ug in 0.1cc in rabbit eyes was safe and no clinical or histological changes were observed after one month.

A plaque reduction assay as an in vitro culture method was conducted to determine the MICs of antibacterial agents (4 types of tetracyclines: tetracycline, doxycycline, minocycline, and tigecycline; 3 types of quinolones: ciprofloxacin, ofloxacin, and levofloxacin; and 2 types of macrolides: azithromycin and clarythromycin) against R. japonica. R. japonica was sensitive to the antibacterial agents tested with MICs similar to those against other spotted fever rickettsia determined in previously described plaque reduction assays.

Aβ production is enhanced during increased neuronal activation, therefore, the excitation of DOX treated female CRFOE LC neurons projecting to the mPFC may exhibit more frequent co-localization with Aβ due to increased neuronal activity of noradrenergic neurons.

In summary, the adhesive antibacterial and conductive GT-DA/chitosan/CNT hydrogels showed great potential as multifunctional bioactive dressings for the treatment of infected wounds.

In the study, three configurations of two-stage hybrid CWs (horizontal subsurface flow-down-flow vertical subsurface flow CWs, HF-DVF; horizontal subsurface flow-up-flow vertical subsurface flow CWs, HF-UVF; down-flow vertical subsurface flow-up-flow vertical subsurface flow CWs, DVF-UVF) were operated to evaluate their ability to remove high-concentration antibiotics (tilmicosin-TMS and doxycycline-DOC), ARGs (seven tet genes and three erm genes), intI1, 16S rRNA, and nutrients from goose wastewater...The relatively high removal efficiencies of NH-N, NON, and NO-N were also observed from DVF-UVF, ranging from 87% to 95% (p > 0.05), indicating that anaerobic ammonium oxidation (anammox) might be established in the CWs. Our results demonstrate that the removal performances of antibiotics using two-stage hybrid CWs are not affected by the combined configuration, whereas the combination of DVF and UVF CWs perform better on the removal of ARGs and nutrients compared with HF-DVF and HF-UVF CWs.

Thus, c-Src-miR-129-1-3p circuits serve as critical triggers for tumor progression in many human cancers that harbor upregulation of c-Src.

A 76-year-old man with a history of giant cell arteritis (GCA) and polymyalgia rheumatica (PMR) approaching the end of a year-long prednisone taper, at a current dose of 4mg/day, presented to the emergency department with a one-week history of suspected flare of his GCA/PMR symptoms...Infectious disease was consulted, and he was treated with a two-week course of doxycycline...Differentiating features include scalp tenderness, jaw and limb claudication, and visual changes in GCA, versus erythema migrans, peripheral neuropathy, meningitis, Bell’s palsy, and cardiac manifestations including myocarditis, pericarditis, and AV nodal block in Lyme disease. In conclusion, in patients with a presentation suggestive of giant cell arteritis or polymyalgia rheumatica who have a history of possible exposure to Borrelia burgdorferi, Lyme disease should be considered as part of the differential in order to prevent morbidity.

All three kirromycin congeners depleted Wolbachia in LDW1 Drosophila cells in vitro with half-maximal inhibitory concentrations (IC) in nanomolar range, while doxycycline, a registered drug with anti- Wolbachia activity, showed lower activity with an IC of 152 ± 55 nM...No cytotoxicity against HEK293T and HepG2 mammalian cells was observed with 1-3 at the highest concentration (40 μM) used in the assay. These results suggest kirromycin is an effective lead scaffold, further exploration of which could potentially lead to the development of novel treatments for filarial nematode infections.

All the AR-VF isolates remained susceptible to doxycycline...The majority of the isolates were resistant to β-lactam antibiotics (bla [96%], bla [93%], and bla [68%]) and aminoglycoside actetyltransferase, conferring resistance to ciprofloxacin-modifying enzyme [aac(6')Ib-cr] (96%)...Resistance to azithromycin is encoded in the gene mph(A). Emerging azithromycin resistance in V. fluvialis is a major public health challenge, and future studies should be focused on identifying ways to prevent the dissemination of this antibiotic resistance gene.

MICs of ampicillin, tetracycline and doxycycline were higher for porcine H. suis isolates compared to primate isolates and in these porcine isolates SNPs were detected in genes encoding penicillin binding and ribosomal proteins. This study indicates that acquired resistance occasionally occurs in H. suis isolates and that zoonotically important porcine isolates may be intrinsically less susceptible to β-lactam antibiotics and tetracyclines than primate isolates.

Here we have developed a doxycycline-inducible strategy where Wnt7b is temporally induced in the bones of aged mice or during fracture healing...Induced expression of Wnt7b at the onset of fracture did not affect the initial cartilage formation but promoted mineralization of the subsequent bone callus. Thus, targeted delivery of Wnt7b to aged bones or fracture sites may be explored as a potential therapy.
