  • ||||||||||  Retrospective data, Review, Journal:  Syphilis Treatment: Systematic Review and Meta-Analysis Investigating Nonpenicillin Therapeutic Strategies. (Pubmed Central) -  Apr 10, 2024
    The studies were considerably homogeneous, and stratified analyses considering each alternative treatment separately revealed that penicillin monotherapy did not outperform ceftriaxone (pooled odds ratio, 1.66 [95% confidence interval, .97-2.84]; I2 = 0%), azithromycin (0.92; [.73-1.18]; I2 = 0%), or doxycycline (0.82 [.61-1.10]; I2 = 1%) monotherapies with respect to serological conversion. Alternative treatment strategies have serological cure rates equivalent to penicillin, potentially reducing global dependence on this antibiotic.

    Similarly, a significant phenotype-genotype correlation (p < 0.001) was observed between chloramphenicol and the tetB, and colistin with blaTEM and resistance gene of quinolone (qnrA)...We deciphered the complex nature of AMR in APEC populations. This information of molecular surveillance is useful to combat AMR in APEC and to contribute to manage APEC associated diseases and develop policies and guidelines to enhance the commercial chicken production.

    Journal:  Generation of Connexin-Expressing Stable Cell Pools. (Pubmed Central) -  Apr 9, 2024
    Taking advantage of the ability of lentivirus (LV) to promote transgene delivery, and genomic integration and expression in both dividing and nondividing cells, a doxycycline-inducible transfer vector expressing a bicistronic transgene was developed to study the function of connexins in HeLa DH cells...We further present step-by-step immunolabeling protocols to characterize the transgene protein expression by confocal microscopy using an antibody that targets an extracellular domain epitope of Cx32 in living cells, and in fixed permeabilized cells using high affinity anti-Cx32 antibodies. Using common molecular biology laboratory techniques, this protocol can be adapted to generate stable pools expressing any transgene of interest, for both in vitro and in vivo functional assays, including molecular, immune, and optical assays.

    Journal:  Elucidating doxycycline biotransformation mechanism by Chryseobacterium sp. WX1: Multi-omics insights. (Pubmed Central) -  Apr 8, 2024
    The elucidated degradation pathway encompassed several key biological processes, such as the microbial transmembrane transport of DOX and its intermediates, the orchestration of enzyme synthesis for transformation, energy metabolism, and other gene-regulated biological directives. This study provides the first insight into the adaptive biotransformation strategies of Chryseobacterium under DOX-induced stress, highlighting the potential applications of this strain to augment DOX removal in wastewater treatment systems containing high concentrations of DOX.

    Retrospective data, Journal:  A Retrospective Study of Covid-19 Patients: A Real-Life Experience in a Secondary Care Hospital, Jashore, Bangladesh. (Pubmed Central) -  Apr 5, 2024
    Therefore, we can conclude that the disease was well managed with highly satisfactory outcomes even in the low resource health care setting in the early stage of Covid-19 pandemic era. Hence, the early case detection, identification of the high-risk groups and proper management may mitigate to develop complications and fatalities of Covid-19.

  • ||||||||||  Journal:  Prevalence and antimicrobial resistance of Enterobacteriaceae in the north of Kazakhstan. (Pubmed Central) -  Apr 2, 2024
    The most common genes encoding antimicrobial resistance were as follows: BlaTEM and OXA in 41 and 28 isolates, respectively, encoding resistance to beta-lactams; StrA and StrB in 45 and 48 isolates encoding aminoglycosides; and tetA and tetB in 43 and 28 isolates encoding tetracyclines. Obtained data demonstrate that uncontrolled and frequent use of beta-lactam and tetracycline antibacterials, in cats and dogs, results in the spread of genotypic resistance among micro-organisms of the family Enterobacteriaceae.

    Journal:  Antimicrobial Indole-3-Carboxamido-Polyamine Conjugates Target Bacterial Membranes and Are Antibiotic Potentiators. (Pubmed Central) -  Apr 1, 2024
    The same analogue also restored the action of doxycycline toward P. aeruginosa with a 21-fold enhancement, while the corresponding 5-bromo-indole-3-carboxamide-PA3-7-3 analogue was able to enhance the action of both doxycycline and erythromycin toward P. aeruginosa and Escherichia coli, respectively. The analogue 13b was capable of disrupting the bacterial membrane of both S. aureus and methicillin-resistant S. aureus (MRSA) and the outer membrane of P. aeruginosa, suggesting that membrane perturbation could be a mechanism of action of both intrinsic antimicrobial activities and antibiotic potentiation.

Unfavorable outcomes have been noticed to be minimal in our cohort of patients with osteoarticular brucellosis, treated mainly with a three-drug regimen and a longer duration of therapy.

    Intricate mechanisms trigger scattered crypt IEC apoptosis mediated necrotizing enterocolitis pathogenesis in neonatal mice (Exhibit Hall F1; Poster Board Number: B216) -  Mar 29, 2024 - Abstract #IMMUNOLOGY2024IMMUNOLOGY_1859;
    We developed a novel triple-transgenic mouse model, namely, 3xTg-iAPcIEC (inducible apoptosis phenotype in crypt-IEC) using doxycycline (Dox)-inducible tetO-rtTA system and vil-cre technology, for inducing tissue-specific overexpression of Fasl in IECs...Notably, we observed that blocking any of these events protects NEC development in 3xTg-iAPcIEC mouse pups, underlining their central roles in the NEC pathogenesis. Our findings may advance knowledge in preventing or treating this devastating disorder in humans.

    Using doxycycline inducible HIV-1 Tat transgenic (iTat) mice, we and others have shown that Tat induction is associated with HIV-sensory neuropathy (SN)-like behaviors...Time course study showed consistent changes in Tollip expression at the protein level to that at the RNA level. Future work will identify potential sex-dependent Tollip expression and investigate the role of Tollip in Tat-associated HIV-SN.

    Learning points for clinical practice: Whipple's disease is a very rare disease with often a significant delay in diagnosis. Persistent arthritis and biological inflammatory parameters in other condition with articular involvement, such as typical rheumatoid purpura, should lead clinicians to consider Whipple's disease and perform PCR in saliva and stool.

    Observational data, Journal, Adherence:  Adherence to doxycycline for uncomplicated genitourinary chlamydia: A prospective observational study. (Pubmed Central) -  Mar 25, 2024
    The CDC recommendation to consider azithromycin when nonadherence is a "substantial concern" should be applied broadly in these settings by routinely discussing barriers to adherence when selecting the optimal antimicrobial regimen. The benefit of dispensing the doxycycline regimen on-site compared to prescribing it could not be determined given differences in baseline characteristics between the two groups.
