Journal:  Chemicals of emerging concern in coastal aquifers: Assessment along the land-ocean interface. (Pubmed Central) -  Feb 5, 2023
Among them, the perfluoroalkyl and polyfluoroalkyl substances (PFAS), along with the pharmaceuticals carbamazepine and topiramate, were revealed to be good markers for tracing anthropogenic contamination in ground- and seawater, in concrete situations (e.g., highly contaminated sites). Additionally, non-target analysis expanded the number of potential tracers, making it a promising tool for identifying both the source and the fate of pollutants.

Review, Journal:  The impact of N-nitrosamine impurities on clinical drug development. (Pubmed Central) -  Jan 29, 2023
Consequent interruption or discontinuation of the manufacturing and distribution of several marketed drugs has culminated into shortages of marketed drugs, including the antidiabetic drug metformin and the potentially life-saving drug rifampin for the treatment of tuberculosis...In particular, rifampin is a key clinical index drug employed in drug-drug interaction (DDI) studies, and as a result of nitrosamine impurities regulatory bodies no longer accept the administration of rifampin in DDI studies involving healthy subjects. Drug developers are now forced to look at alternative approaches for commonly employed perpetrators, which will be discussed in this review.

Clinical, Journal:  Cabotegravir: a novel HIV integrase inhibitor combined with rilpivirine as the first long-acting injectable program for the treatment of HIV infection. (Pubmed Central) -  Jan 19, 2023
Cabotegravir, a novel HIV integrase inhibitor, shares structural similarity with dolutegravir and bictegravir...Drug interactions are minimal, although oral doses require spacing from polyvalent cations, and potent uridine glucuronosyltransferase induction (e.g., rifampin, carbamazepine) requires avoidance due to anticipated subtherapeutic cabotegravir exposure through extended intervals...A key component of the first U.S. Food and Drug Administration (FDA)-approved injectable maintenance treatment program for HIV, injectable cabotegravir heralds a new era in HIV treatment innovation. Here we provide a detailed review of the clinical pharmacology, administration and available formulations of the novel HIV integrase inhibitor cabotegravir with in-depth analysis of the clinical trial data, safety, satisfaction and viral resistance development when combined with rilpivirine as the first long-acting injectable program for the treatment of HIV infection.

Journal:  Lignin nanoparticle-decorated nanocellulose cryogels as adsorbents for pharmaceutical pollutants. (Pubmed Central) -  Jan 8, 2023
The resistance of the cryogels towards disintegration with exposure to water also improved with crosslinking, which eventually enabled the cryogels, especially the TCNF-based one, to be regenerated and reused for five cycles of adsorption-desorption experiment for the model pharmaceutical MPL. Thus, this study opened new opportunities to utilize LNPs in providing nanocellulose-based adsorbents with additional functional groups, which were otherwise often achieved by rigorous chemical modifications, at the same time, crosslinking the nanocellulose network.

Journal:  Removal of pharmaceutically active compounds (PhACs) by zero-valent iron: quantification of removal mechanisms consisting of degradation, adsorption and co-precipitation. (Pubmed Central) -  Jan 1, 2023
The removal mechanisms of carbamazepine (CBZ), which is one of pharmaceutically active compounds, using zero-valent iron (ZVI) were quantified by defining three fractions, namely "degradation", "adsorption", and "co-precipitation"...A novel kinetic model for removal of CBZ by ZVI was developed to simulate the dynamic concentration profiles of CBZ, TOC, total Fe ions, and dissolved oxygen linked closely with each other and the contributions of degradation, adsorption, and co-precipitation in TOC removal of CBZ. Reasonable agreement between experimental data and model predictions suggests the applicability of the proposed kinetic model to quantitatively analyze the mechanisms of CBZ removal by ZVI.
