    Journal:  Modern aspects of antiepileptic therapy during pregnancy (Pubmed Central) -  Jun 15, 2019
    In addition to classic AEDs, new AEDs (vigabatrin, gabapentin, topiramate, tiagabine, oxcarbazepine, levetiracetam, pregabalin) were investigated...It has been found that the risk of birth defects in children is associated with high doses of drugs and polytherapy compared to monotherapy. New data of recent studies showed the effect of psychotropic drugs on the fetus allowing to assess the 'risk/benefit' ratio and develop recommendations on rational pharmacotherapy of epilepsy in pregnancy.

    Clinical, Journal:  Clinical features and treatment outcomes of Juvenile myoclonic epilepsy patients. (Pubmed Central) -  Jun 7, 2019
    There is a trend of lower rates of seizure freedom in patients treated with LEV than in those treated with VPA after the first-year treatment period. Our data suggest that in JME, seizure control is linked to seizure type, possibly allowing a more individualized approach when counseling JME patients.
  • ||||||||||  Brabafen (low-dose fenfluramine) / Zogenix
    Review, Journal:  Treatment Strategies for Dravet Syndrome. (Pubmed Central) -  May 31, 2019
    Status epilepticus is a recurring problem for patients with DS, particularly in their early childhood years. All patients should be prescribed a home rescue therapy (usually a benzodiazepine) but should also have a written seizure action plan that outlines when rescue should be given and further steps to take in the local hospital if the seizure persists despite home rescue therapy.

    Journal:  Drug-drug interactions between antiepileptics and cannabinoids. (Pubmed Central) -  May 16, 2019
    Cannabidiol proved effective in clinical trials, reducing seizure frequency and the most frequently observed adverse effects were diarrhea, somnolence, and poor appetite. The retention rate was within 14-24% (12-14 weeks - 1 year) but it reached the level of 35% after 2 years.
  • ||||||||||  Preclinical, Journal:  The anti-epileptic drugs valproate, carbamazepine and levetiracetam cause bone loss and modulate Wnt inhibitors in normal and ovariectomised rats. (Pubmed Central) -  May 15, 2019
    Further, our results demonstrate for the first time that these effects are more pronounced in ovariectomised rats as compared to normal rats and that this could be related to estrogen deficiency which in turn enhances bone resorption via increased RANKL and reduces bone formation via increased sclerostin and reduced P1NP. Finally, our study demonstrated for the first time that AED treatment displayed changes in the serum levels of wnt inhibitors and hence modulation of wnt inhibitors might be partly involved in their adverse effects on bone.
  • ||||||||||  Trileptal (oxcarbazepine) / Novartis
    Journal:  Reliability of information about the use of antiepileptic drugs during pregnancy from three major web search engines in China. (Pubmed Central) -  May 14, 2019
    Finally, our study demonstrated for the first time that AED treatment displayed changes in the serum levels of wnt inhibitors and hence modulation of wnt inhibitors might be partly involved in their adverse effects on bone. Accurate and professional online information for female patients with epilepsy should be provided through major efforts by the government, search engine companies, professional websites and epilepsy physicians.
  • ||||||||||  Seizalam (midazolam intramuscular) / Pfizer
    Review, Journal:  Focal status epilepticus: a review of pharmacological treatment. (Pubmed Central) -  May 12, 2019
    The available scientific evidence is insufficient to claim that pharmacological treatment of focal SE should be different from treatment for generalised SE. More studies with a greater number of patients are needed.

    Clinical, Review, Journal:  Strategies of using the new antiepileptic drugs for epilepsy in adults (Pubmed Central) -  May 11, 2019
    The new antiepileptic drugs can succesfully used for the treatment of special patients' groups as for the post stroke, poszttraumatic epilepsies, for the epilepsies accompanied with brain tumours as well as for epilepsies in the elderly. The new drugs are advantageous for the treatment of such patients who have psychiatric symptoms or signs of cognitive decline and high risk of these symptoms respectively.
  • ||||||||||  Briviact (brivaracetam) / UCB
    Retrospective data, Journal:  BRIVA-LIFE - a multicentre retrospective study of the long-term use of brivaracetam in clinical practice. (Pubmed Central) -  May 9, 2019
    The new drugs are advantageous for the treatment of such patients who have psychiatric symptoms or signs of cognitive decline and high risk of these symptoms respectively. In a large population of patients with predominantly drug-resistant epilepsy, brivaracetam was effective and well-tolerated; no unexpected AEs occurred over 1 year, and the incidence of PAEs was lower compared with levetiracetam.
  • ||||||||||  Cerebyx (fosphenytoin) / Pfizer, Nobelpharma
    Journal:  Levetiracetam in toxic seizures. (Pubmed Central) -  May 7, 2019
    Levetiracetam used as a second-line agent was associated with control of drug-induced seizures and prevention of seizure recurrence without obvious adverse effects. A prospective study is needed to confirm these results.
  • ||||||||||  pentylenetetrazole (BTD-001) / Balance Therap
    Journal:  Function of synaptic vesicle protein 2A (SV2A) as a novel therapeutic target for epilepsy (Pubmed Central) -  May 7, 2019
    In addition, the Sv2a mutation specifically impaired depolarization-induced GABA, but not glutamate, release in the hippocampus and amygdala. All this evidence indicates that the SV2A-GABAergic system plays a crucial role in modulating epileptogenesis and encourages discovery research into the novel antiepileptic agents which enhance the function of the SV2A-GABA system.

    Clinical, Journal:  Consensus Protocol for the Treatment of Super-Refractory Status Epilepticus. (Pubmed Central) -  May 7, 2019
    We propose a treatment protocol based on a sequential combination of anaesthetics, anti-epileptic drugs and alternative therapies. Strategies to evaluate treatment response and to wean drugs based on clinical results are also proposed.
  • ||||||||||  Radicava (edaravone) / Mitsubishi Tanabe
    Clinical, Journal:  Atypical case of posterior reversible encephalopathy syndrome related to late onset postpartum eclampsia: A case report. (Pubmed Central) -  May 5, 2019
    Her clinical course was uneventful and she fully recovered without neurological complications LESSONS:: The possible diagnosis of late onset postpartum eclampsia, even weeks post-delivery, should be considered, since initiation of early treatment averts severe complications and decreases mortality. Sharing our experience may increase awareness of PRES induced by late-onset postpartum eclampsia.
  • ||||||||||  levetiracetam / generics
    Retrospective data, Review, Journal:  A meta-analysis of levetiracetam for randomized placebo-controlled trials in patients with refractory epilepsy. (Pubmed Central) -  May 4, 2019
    Regarding safety profile, patients treated with levetiracetam had significantly higher occurrence than placebo for somnolence, asthenia, dizziness, infection, nasopharyngitis, anxiety, and irritability; however, most studies reported that these adverse events were mild and transient. Levetiracetam is an effective anti-epileptic drug for both adults and children with generalized or partial-onset refractory seizures at 1,000-3,000 or 60 mg/kg/day, with a favorable adverse event profile.
  • ||||||||||  levetiracetam / generics
    Journal:  Levetiracetam circulating concentrations and response in status epilepticus. (Pubmed Central) -  May 2, 2019
    Loading doses of 30 mg/kg seem, however, appropriate to quickly reach the target exposure level. The short LEV half-life makes standardized sampling measurement necessary to obtain directly interpretable LEV levels.

    PK/PD data, Journal:  Influence of Sex, Age and Weight on Levetiracetam Pharmacokinetics. (Pubmed Central) -  Apr 30, 2019
    It is confirmed that LEV dosage per body weight is not required, and prescribing higher doses of LEV in obese patients is not justified. These data suggest that routine LEV therapeutic drug monitoring (TDM) in the elderly patients, patients with renal dysfunction and obese patients is indicated.
