Journal: Regulation of TGF-β1-Induced EMT by Autophagy-Dependent Energy Metabolism in Cancer Cells. (Pubmed Central) - Oct 15, 2022 Suppression of autophagy activity by knocking down of BECN1 or chloroquine (CQ) treatment inhibited mesenchymal protein expression...These results suggest that A549 non-small cell lung cancer required autophagy to maintain mitochondrial homeostasis during TGF-β1 induced EMT. In conclusion, blocking autophagy decreased energy production and down-regulated proteins synthesis inhibiting TGF-β1 induced EMT.

Journal: APOL1 variant-expressing endothelial cells exhibit autophagic dysfunction and mitochondrial stress. (Pubmed Central) - Oct 15, 2022 Upon blocking autophagy with non-selective lysosome inhibitor, hydroxychloroquine, autophagosome accumulation between 0RV HUVECs and untreated 2RV HUVECs was similar, implicating lysosomal dysfunction in the HRG-associated autophagy defect...Taken together, our data reveal that IFNɣ induces APOL1 expression, and that each additional RV associates with mitochondrial dysfunction and autophagy inhibition. IFNɣ amplifies this phenotype even in 1RV HUVECs, representing the first description of APOL1 pathobiology in variant heterozygous cell cultures.

(Pubmed Central) - Oct 15, 2022 Actinomycin D was used to determine the mRNA stability...Activation of autophagy enhanced the osteogenesis of hBMSCs and inhibition of autophagy reduced the osteogenesis through using autophagy regulators chloroquine and rapamycin...Knockdown of circHIPK3 released the binding sites of HUR to ATG16L1, which stabilized the mRNA expression of ATG16L1, resulting in the upregulation of ATG16L1 and autophagy activation. CircHIPK3 functions as an osteogenesis and autophagy regulator and has the potential for clinical application in the future.

Journal: Albendazole-induced autophagy blockade contributes to elevated apoptosis in cholangiocarcinoma cells through AMPK/mTOR activation. (Pubmed Central) - Oct 14, 2022 In addition, ABZ blocked autophagic flux by inhibiting the fusion of autophagosome-lysosome, which increased the apoptotic death of CCA cells. However, the apoptotic death of CCA cells induced by ABZ was reversed by 3-methyladenine (3-MA), an autophagosome formation inhibitor, but increased by chloroquine (CQ), an autophagosome-lysosome fusion inhibitor.Our work provides novel mechanisms for anti-tumor effects of ABZ on CCA, suggesting that ABZ may be used as a potent autophagy inhibitor in the treatment of CCA.

The Characteristics and Mechanism of Cell Senescence in Patients With Diabetic Kidney Disease (Exhibit Hall, Orange County Convention Center, West Building) - Oct 13, 2022 - Abstract #KIDNEYWEEK2022KIDNEY_WEEK_3574; Electronic microscope showed an increase in autophagosomes in the renal tubules of DKD rats.HK-2 cells treated with high glucose exhibited increased expression of p21, p53, p62 and LC3(p<0.05).CQ elevated p21 and p53 compared to the high glucose group, whereas opposite trend was observed in the rapamycin treatment (p<0.05). Conclusion Our study demonstrated a positive correlation between cell senescence and progression of DKD, and autophagy attenuated the high-glucose-mediated renal tubule senescence.

Conclusion We found dynamic autophagy dysregulation in the Pkd1 RC/RC mouse model of ADPKD, i.e., impaired cargo clearance in the early phase, and adaptive/maladaptive autophagy activation in the later phase, and a spatial- and temporal-dependent dysregulation of Tfeb. Our study suggests a time-dependent mechanism that needs to be considered when developing an autophagy-based therapy for ADPKD.

- |||||||||| Review, Journal: Molecular Mechanism of the Protective Effects of M2 Microglia on Neurons: A Review Focused on Exosomes and Secretory Proteins. (Pubmed Central) - Oct 13, 2022
M2 microglia have attracted increasing attention in recent years owing to their beneficial effects on various diseases and injuries of the CNS, such as traumatic brain injury, stroke, Alzheimer's disease and multiple sclerosis. They exert neuroprotective effects by various mechanisms, e.g., suppressing inflammation, promoting the degradation of misfolded and aggregated proteins, promoting neurite growth, enhancing neurogenesis, inhibiting autophagy and apoptosis, promoting myelination, maintaining blood-brain barrier integrity, and enhancing phagocytic activity.This review summarizes the molecular mechanisms by which M2 microglia exert protective effects on neurons and provides a reference for the selection of therapeutic targets for CNS diseases.

(Pubmed Central) - Oct 12, 2022 Inhibiting proteasome by Velcade was found to be highly toxic to Danon mice, suggesting that proteasome is activated to compensate for defective autophagy...Since Danon's is a lifelong disease, we suggest that lifestyle interventions to decrease cardiac stress may be useful to slow progression of Danon's cardiomyopathy. While Danon mice better tolerate high fat diet and sedentary lifestyle, the benefit regarding cardiomyopathy in humans needs to be balanced against other health consequences of such interventions.

Journal: Autophagy (but not metabolism) is a key event in mitoxantrone-induced cytotoxicity in differentiated AC16 cardiac cells. (Pubmed Central) - Oct 12, 2022 When focusing in autophagy, the mTOR inhibitor rapamycin and the autophagy inhibitor 3-methyladenine exacerbated the cytotoxicity caused by MTX and NAPHT, while the autophagy blocker, chloroquine, partially reduced the cytotoxicity of MTX...In conclusion, in our in vitro model, neither metabolism nor exogenously given NAPHT are major contributors to MTX toxicity as seen by the residual influence of metabolism modulators used on the observed cytotoxicity and by NAPHT's low cytotoxicity profile. Conversely, autophagy is involved in MTX-induced cytotoxicity and MTX seems to act as an autophagy inducer, possibly through p62/LC3-II involvement.

- |||||||||| Cytosolic ENC1 increases pathological tau accumulation by inhibiting autophagy flux (SDCC 1) - Oct 10, 2022 - Abstract #Neuroscience2022NEUROSCIENCE_816;
We also found that overexpression of ENC1 significantly increased DS9 tau seeding activity, while knockdown of ENC1 decreased tau seeding activity in the SH-SY5Y seeding model. Interestingly, overexpression of ENC1 increased the number of autophagosomes and reduced the number of autolysosomes, indicating ENC1 may function to inhibit autophagic flux, thereby disrupting the ALP and promoting the accumulation of tau aggregates.

Our study demonstrates that OL cells can acquire the "metastatic ability" from OL-SCs cells. - |||||||||| Journal: Autophagy is essential for neural stem cell proliferation promoted by hypoxia. (Pubmed Central) - Oct 9, 2022
Thus, we suppose that the interaction of BNIP3 with Rheb reduces the regulation of Rheb towards mTORC1 activity, which relieves the suppression of mTORC1 on autophagy, thereby promoting the rapid proliferation of NSCs. Altogether, this study identified a new HIF-1/BNIP3-Rheb/mTORC1 signaling axis, which regulates the NSC proliferation under hypoxia through induction of autophagy.
