  • ||||||||||  Preclinical, Journal:  Serial passage of PDCoV in cell culture reduces its pathogenicity and its damage of gut microbiota homeostasis in piglets. (Pubmed Central) -  Feb 13, 2024   
    Here, the PDCoV virulent HNZK-02 strain was successfully attenuated by serial passage on cell cultures, and the pathogenesis and effects on the gut microbiota composition and metabolic function of the PDCoV HNZK-02-P5 and P150 strains were investigated in piglets. We also found the genetic changes in the S protein during passage in vitro and the gut microbiota may contribute to the pathogenesis of PDCoV, while their interaction molecular mechanism would need to be explored further.

    In addition, liver-specific expression of APNTri by AAV delivery increases the circulatory APN levels and improves memory functions in AD mice. AAV-APNTri treatment reduces microglia activation and IL1b levels in the brain of 5xFAD in which NLRP3, ASC and caspase-1 levels are also reduced in microglia.  Conclusions Taken altogether, our study demonstrates the circulatory APN inhibits IL1β secretion from microglia through NLRP3 inhibition, suggesting that increasing periphery trimeric APN can be a potential AD therapy.
