These data should be considered in decisions regarding SDRM list updates and TDR monitoring.

Clinical, Journal: Liver stiffness change with HCV cure in HIV-infected patients on non-nucleoside analogues. (Pubmed Central) - Oct 30, 2021 The biological mechanisms underlying the differential effects of switching ART agents on weight and associated clinical implications require further study. Treatment with NNRTI plus 2 NRTI combinations is associated with a higher LS decline than other ART combinations in HIV/HCV-coinfected patients receiving DAA-based therapy.

(Pubmed Central) - Oct 16, 2021 They displayed up to single-digit nanomolar activity against wild-type (WT) and rilpivirine-associated resistant mutant E138K viruses, as well as potent inhibitory ability toward the RT enzyme...The structure-activity relationships (SARs) of the newly obtained DAPYs were also investigated. Molecular docking analysis elucidated the biological activity and offered a structural insight for follow-up research.

Preclinical, Journal: Development of a novel in vitro assay to screen for neuroprotective drugs against iatrogenic neurite shortening. (Pubmed Central) - Oct 14, 2021 Additionally, two hits originated from a previous primary screening aimed to detect inhibitors of hyperexcitability to inflammatory mediators in DRG neurons (nitrendipine and felodipine) also prevented neurite shortening in our model. In summary, in this work we developed a novel secondary assay for identifying hits with neuroprotective effect against iatrogenic neurite shortening, consistent with the anti-hyperexcitability action previously tested: highlighting nitrendipine and felodipine against iatrogenic damage in DRG neurons.

Journal: Rilpivirine inhibits SARS-CoV-2 protein targets: A potential multi-target drug. (Pubmed Central) - Oct 14, 2021 As a result of our in silico molecular docking study, we suggest that rilpivirine is a compound that could act as a powerful inhibitor against SARS-CoV-2 targets. Although in vitro and in vivo experiments are needed to verify this prediction we believe that this antiviral drug may be used in preclinical trials to fight against SARS coronavirus.

(Pubmed Central) - Oct 3, 2021 Treatment-related adverse events occurred in 51 (15%) patients (all Grade 1-2) and led to 8 discontinuations.In this real-world cohort, switching to B/F/TAF was associated with maintenance of virologic control, and avoidance of DDIs in a large proportion of patients. These data support use of B/F/TAF as a treatment option in older PLWH.

Journal: Structure-Based Non-Nucleoside Inhibitor Design: Developing Inhibitors that Are Effective Against Resistant Mutants. (Pubmed Central) - Sep 25, 2021 The best of the new compounds, 12, was a more potent inhibitor of the NNRTI-resistant mutants we tested than either doravirine or efavirenz but was inferior to RPV. We describe the limitations on the modifications that can be appended to the "upper right side" of the RPV core and the effects of substituting other cores for the central pyrimidine core of RPV and make suggestions about how this information can be used in NNRTI design.
