    Clinical, Journal:  Switching to a Healthy Diet Prevents the Detrimental Effects of Western Diet in a Colitis-Associated Colorectal Cancer Model. (Pubmed Central) -  Jul 29, 2020
    We hypothesized that Western-style diet (WD) aggravates azoxymethane (AOM)/dextran sulfate sodium salt (DSS)-induced colitis-associated tumorigenesis and that switching to the standard AIN93G diet will ameliorate disease symptoms even after cancer initiation...The switch to the AIN93G diet ameliorated this effect, reflected by longer colons, fewer (p < 0.05) and smaller (p < 0.01) aberrant colonic crypt foci, comparable with the AIN group. Our results show that switching to a healthy diet, even after cancer initiation is able to revert the deleterious effect of the WD and could be an effective preventive strategy to reduce colitis symptoms and prevent tumorigenesis.

    Journal:  Effect of Sishen Pill on Memory T Cells From Experimental Colitis Induced by Dextran Sulfate Sodium. (Pubmed Central) -  Jul 28, 2020
    Furthermore, SSP markedly inhibited activation of PI3K, Akt, phospho-Akt, Id2, T-bet, forkhead box O3a, Noxa, and C-myc proteins in the PI3K/Akt signaling pathway and activated Rictor, Raptor, tuberous sclerosis complex (TSC)1, TSC2, phospho-AMP-activated kinase (AMPK)-α, AMPK-α, eukaryotic translation initiation factor 4E-binding protein 2, kinesin family member 2a, and 70-kDa ribosomal protein S6 kinase. These results indicate that SSP effectively controls Tem cells in the peripheral blood to relieve experimental colitis induced by DSS, which were potentially related with inhibiting the PI3K/Akt signaling pathway.

    Journal:  5-Hydroxytryptamine (5-HT)-exacerbated DSS-induced colitis is associated with elevated NADPH oxidase expression in the colon. (Pubmed Central) -  Jul 21, 2020
    These results showed the multiple effects of β-sitosterol against pathogenic bacteria for a novel approach to the treatment of colonic inflammation. As 5-HT induced NOX1 and DUOX2 gene and protein expression in colon epithelial and HT-29 cells, NOX2 and NOX4 in the infiltrating leukocyte in mouse colon and in U-937 cells, the exacerbate colitis induced by combined 5-HT and DSS treatment might be relevant to increased NOX expression in mice colons.

    Preclinical, Journal:  Endogenous murine microbiota member Faecalibaculum rodentium and its human homologue protect from intestinal tumour growth. (Pubmed Central) -  Jul 21, 2020
    Reconstitution of Apc or azoxymethane- and dextran sulfate sodium-treated mice with an isolate of F. rodentium (F...Both F. PB1 and H. biformis produced short-chain fatty acids that contributed to control protein acetylation and tumour cell proliferation by inhibiting calcineurin and NFATc3 activation in mouse and human settings. We have thus identified endogenous anti-tumourigenic bacterial strains with strong diagnostic, therapeutic and translational potential.

    Preclinical, Journal:  Effects of sDR5-Fc fusion protein on infant mice with ulcerative colitis via the TRAIL-DR5 pathway. (Pubmed Central) -  Jul 19, 2020
    To explore effects of the sDR5-Fc fusion protein on ulcerative colitis of infant mice via the TRAIL-DR5 pathway, 50 female mice were randomly divided into 5 groups, i.e., control group (group A), dextran sulfate sodium group (group B), hIgG group (group C), 10 mg/kg sDR5-Fc group (group D), and 20 mg/ kg sDR5-Fc group (group E)...Thus, sDR5-Fc could inhibit the activation of NLRP3 inflammasomes induced by TRAIL, thereby decreasing the apoptosis of NCM460 cells. In conclusion, the sDR5-Fc fusion protein could block the TRAIL-DR5 pathway to reduce the expression of NLRP3 inflammasomes, thereby improving ulcerative colitis.

    [VIRTUAL] Scavenger Receptor-Mediated Uptake of DNAZyme-Gold Nanoparticle Conjugates in Alveolar Macrophages Using Two Photon Microscopy Analysis (ATS 2020 Virtual) -  Jul 6, 2020 - Abstract #ATSI2020ATS-I_3660;
    Methods : Using two-photon photoluminescence microscopy, we measured the emission intensity of DzNPs in murine alveolar macrophages (MH-S) treated for 24 hours with increasing doses of DzNP with and without increasing doses of the SR inhibitor dextran sulfate sodium (DSS)...DzNP uptake in alveolar macrophages is dose dependent with a Poisson distribution and is mediated by scavenger receptors. With spectral resolution around 170 nm, spacial localization of DzNP within the cell is possible with relative precision without the need for fluorescent labeling, thus offering greater optical versatility when fluorescent labeling is necessary to visualize other proteins or organelles of interest.
