Journal: MWF of the corpus callosum is a robust measure of remyelination: Results from the ReBUILD trial. (Pubmed Central) - May 12, 2023 An increase in myelin water fraction was documented in the normal-appearing white matter of the corpus callosum, in correspondence with the administration of the remyelinating treatment clemastine...Moreover, our work strongly suggests that significant myelin repair occurs outside of lesions. We therefore propose myelin water fraction within the normal-appearing white matter of the corpus callosum as a biomarker for clinical trials looking at remyelination.

Journal: Disturbed Oligodendroglial Maturation Causes Cognitive Dysfunction in Schizophrenia: A New Hypothesis. (Pubmed Central) - May 10, 2023 We therefore propose myelin water fraction within the normal-appearing white matter of the corpus callosum as a biomarker for clinical trials looking at remyelination. Based on these findings, we propose that pro-myelinating drugs (e.g., the histamine blocker clemastine) combined with aerobic exercise training may foster the regeneration of myelin plasticity as a basis for restoring frontotemporal connectivity and cognition in schizophrenia.

Pro-Myelinating Drugs Promote Functional Recovery in an ASD Mouse Model of Pitt-Hopkins Syndrome (Sapphire AB) - Apr 29, 2023 - Abstract #SOBP2023SOBP_427; Results In vitro, clemastine treatment reduced excess oligodendrocyte precursor cells (OPCs) and normalized OL density...Conclusions In this study, we show that two different pro-myelinating compounds with independent modes of action, are effective at restoring myelination defects in a PTHS mouse model. Together, these results provide preclinical evidence that pro-myelinating therapies may be beneficial in PTHS and potentially other neurodevelopmental disorders characterized by hypomyelination.

Preclinical, Journal: Deficient deposition of new myelin impairs adult optic nerve function in a murine model of diabetes. (Pubmed Central) - Mar 21, 2023 In addition, clemastine, an antimuscarinic compound that enhances myelination, significantly restored oligodendroglia, and promoted the functional recovery of the optic nerve in diabetic mice. Together, our results point to the role of new myelin deposition in optic neuropathy under diabetic insult and provide a promising therapeutic target for restoring visual function.

Journal: Insufficient oligodendrocyte turnover in optic nerve contributes to age-related axon loss and visual deficits. (Pubmed Central) - Mar 20, 2023 To test whether enhancing oligodendrogenesis can prevent age-related visual deficits, 12-month-old mice were treated with clemastine, a pro-myelination drug, or induced deletion of the muscarinic receptor 1 in oligodendrocyte precursor cells...Conversely, pro-myelination approaches significantly increased new myelin generation in aging ONs, and consequently preserved retinal integrity and visual function. Our findings indicate that promoting OL generation might be a promising strategy to preserve visual function from age-related decline.

Preclinical, Journal: Pharmacological treatment promoting remyelination enhances motor function after internal capsule demyelination in mice. (Pubmed Central) - Mar 14, 2023 Moreover, clemastine-treated mice showed increased mature oligodendrocyte density, reduced axonal injury, an increased number of myelinated axons and thicker myelin in the IC lesions compared with control (PBS-treated) mice. These results suggest that the lysophosphatidylcholine-induced IC demyelination model is useful for evaluating changes in motor functions following pharmacological treatments that promote remyelination.

(Pubmed Central) - Dec 12, 2022 Collectively, we first demonstrated the bi-directional influence of early sevoflurane exposures on oligodendrocyte maturation and proliferation, which contributes to the cognitive impairment induced by general anesthesia. These findings illustrated the dynamic changes in oligodendrocytes in the developing brain following anesthetic exposures, as well as possible therapeutic strategies for multiple general anesthesia associated cognitive impairment.

Review, Journal: Drug repurposing: Clemastine fumarate and neurodegeneration. (Pubmed Central) - Nov 15, 2022 Hence, whether clemastine fumarate would make a protective impact on neurodegenerative diseases and what contribution fumarate probably makes are intriguing to us. In this review, we summarize the potential mechanism surrounding clemastine fumarate in current literature, and try to distinguish independent or synergistic effect between clemastine and fumarate, aiming to find worthwhile research direction for neurodegeneration diseases.

CN045 is a novel small molecule lead for remyelinating therapies in multiple sclerosis (ePoster Area) - Oct 18, 2022 - Abstract #ECTRIMS2022ECTRIMS_1799; We have generated novel and patentable analogs of CN045 that retain OPC differentiation with greater potency than CN045, T3, Clemastine and Beztropine. A remyelination therapy, in combination with the current anti-inflammatory therapies, will promote myelin repair and halt neurological decline in MS patients.

Repurposing clemastine to suppress glioblastoma stem cells (West/Central Hall) - Sep 28, 2022 - Abstract #SNO2022SNO_1090; Finally, we demonstrated that a mouse neural stem cell-derived glioma model was similarly susceptible to clemastine treatment. Taken together, our study identifies pathways essential for the perpetuation of stemness in GBM, and implicates a non-oncology drug with a well-established safety profile that can be repurposed to mitigate the stem-like properties of GBM.

Journal: The Neurotrophic Receptor Tyrosine Kinase in MEC-mPFC Neurons Contributes to Remote Memory Consolidation. (Pubmed Central) - Jul 24, 2022 Notably, TrkB deletion seriously inhibited experience-dependent maturation of oligodendroglia in the PFC, resulting in defects in remote recall that were rescued by clemastine administration...We evaluated the contextual fear memory and labeled cells, which revealed deletion of TrkB interrupts newborn oligodendrocyte and reactivation of cells in mPFC at remote recall. Our data provide the implication that remote memory is relevant to neurotrophic receptor signaling as well as its influence on non-neuronal cells.

(Pubmed Central) - Jun 10, 2022 Furthermore, clemastine does not improve VEP latency following demyelinating injury when administered to a transgenic animal incapable of forming new myelin. Therefore, using the capacity for therapeutic enhancement and biological loss of function we demonstrate conclusively that VEP measures myelin status and is thereby a validated tool for preclinical verification of remyelination.
