    Furthermore, we revealed that the sensitivity of CaMKII to PP2A in the three species differs, with rat, C. elegans, and hydra being less dephosphorylated in that order. The evolutionarily acquired features of mammalian CaMKII?-specific structural arrangement and phosphatase tolerance may differentiate neuronal function between mammals and other species.

    Using constitutive and inducible models to delete Tbx21 specifically in B cells we showed that the rapid differentiation potential of Bmem is controlled by T-bet and that T-bet is required for the development and persistence of NP+ eBmem subsets in LN and lung. Thus T-bet not only marks Bmem with effector potential but also regulates the persistence and function of flu-induced NP+ Bmem that are transcriptionally poised to provide rapid humoral protection from infection.

    BTK-deficient mice have increased commensal diversity and enrichment of specific microbes. Based on these findings, we conclude that disruption of BTK-mediated BCR signaling alters IgA production and microbiome composition, with downstream effects on T1D pathogenesis.

    The surgical management of achalasia in children is most commonly addressed with a laparoscopic Heller myotomy; however, the advent of POEM and the advancement of this technology has presented other options in younger patients. This session will discuss “best practice” approaches to achalasia in children and adolescents including technical pointers related to laparoscopic Heller myotomy as well as the emerging role of POEM for adolescents.

    Journal:  Mutability of mononucleotide repeats, not oxidative stress, explains the discrepancy between laboratory-accumulated mutations and the natural allele-frequency spectrum in C. elegans. (Pubmed Central) -  Mar 11, 2022   
    Using genome sequence data from C. elegans MA lines carrying a mutation (mev-1) that increases the cellular titer of reactive oxygen species (ROS), leading to increased oxidative stress, we find the base-substitution spectrum is similar between mev-1, its wild-type progenitor (N2), and another set of MA lines derived from a different wild strain (PB306)...Further, the mutational properties of mononucleotide repeats in all strains are different from those of nonmononucleotide sequence, both for indels and base-substitutions, and whereas the nonmononucleotide spectra are fairly similar between MA lines and wild isolates, the mononucleotide spectra are very different, with a greater frequency of A:T → T:A transversions and an increased proportion of ±1-bp indels. The discrepancy in mutational spectra between laboratory MA experiments and natural variation is likely owing to a consistent (but unknown) effect of the laboratory environment that manifests itself via different modes of mutability and/or repair at mononucleotide loci.

    To achieve this, four industrially relevant epoxy resins (two based on bisphenol A-Epikote 828 (E828) and Dow Epoxy Resin 332 (DER 332)-and two based on bisphenol F-Dow Epoxy Resin 354 (DER 354) and Araldite PY306 (PY306)) and the isomerically pure para-para-diglycidyl ether of bisphenol F (ppDGEBF) were used to explore variation caused by epoxy monomer variation...Bisphenol F-based resins were found to outperform bisphenol A-based analogues, and chain extension within the epoxy component was found to be detrimental to performance. For amines, 1,3-substitution (vs 1,4) and aromaticity were both found to be beneficial to chemical performance.

    Methods : The human Arg306-region PKKTRNL was exchanged for the homologous ptFV sequence GNPDTLT, generating ptFV-h306 and hFV-pt306; recombinant removal of ptFV’s disulfide bond resulted in ptFV-h306-SS...Remarkably, upon proteolysis the structural integrity of ptFVa is maintained, indicating a completely novel role for specific A1 and A2-domain structural regions that have been uniquely adapted in ptFVa in order to prevent A2-domain dissociation. As such, these results challenge our current understanding by which strict regulatory mechanisms control FVa activity.

    Journal:  The nematode Caenorhabditis elegans and the terrestrial isopod Porcellio scaber likely interact opportunistically. (Pubmed Central) -  Sep 10, 2020   
    We show that two different strains-the wild isolate PB306 and the laboratory-adapted strain N2 -are not attracted to P. scaber during either the dauer or adult life stages. Our results indicate that C. elegans was not attracted to chemical compounds or volatile odorants from P. scaber, providing valuable empirical evidence to suggest that any associations between these two species are likely opportunistic rather than specific phoresy.

    The PTMs identified include phosphorylation at S185, S201, T315, S316, T317 and T324, acetylation at K218, S223, S227, K309, K434 and K439, formylation at K218, K309, K325, K341, K369, K434 and K439, methylation at S19, S166, K168 and T364, carboxylation at D108, K298, W307, K309, E323, K325 and K369, and hydroxylation at P22, Y87, P107, D108, D111, P269, P306, R310, N321, K325, Y334, R366, P392 and Y398...SLiMs are proposed to be preferential sites of phosphorylation. Although 6 of 7 phosphosites map to regions of predicted SLiMs, we find no support for the hypothesis that the individual S, T and Y residues of predicted SLiMs are phosphorylated more frequently than S, T and Y residues outside of predicted SLiMs.
