    That is, baseline serotonin firing rate rate returns to normal not because of 5-HT1A desensitization but rather despite ongoing feedback inhibition. Thus while 5-HT1A receptors remain important for emotional behavior, it may be other slow-adaptive changes triggered by antidepressants that allow for therapeutic effects, such as those involving glutamatergic synaptic plasticity.

    During this period, mice received silymarin (200 mg/kg, p.o.) per se or in its nanoparticle form or fluoxetine (10 mg/kg, p.o.)...Silymarin nanoparticles were superior to silymarin in certain parameters probably due to significantly higher brain silybinin (the major active component of silymarin) concentration by 12.46 fold in the group administered silymarin nanoparticles compared with the mice which were administered silymarin per se. The antidepressant-like effect of silymarin can be attributed to its antioxidant and anti-inflammatory effects as well as increased neurogenesis in the prefrontal cortex and hippocampus, which delineates silymarin, especially in nanoparticle form, as a promising strategy for treatment of depression.

    Moreover 5-HT fibers innervating the cortex showed proper density and no overt morphological disorganization, indicating that the reported fluoxetine-induced rearrangements were hippocampus specific. On the whole, these data suggest that 5-HT fibers are shaped in response to subtle changes of 5-HT homeostasis and may provide a structural basis by which antidepressants exert their therapeutic effect.

    Some of these processes, such as immune system, replicated previous findings in the literature, whereas, neurogenesis, a mechanism proposed to be involved in fluoxetine response, had been identified for first time using peripheral tissues. In conclusion, our study identifies several biological processes in relation to fluoxetine treatment in peripheral blood, offer new candidate genes for pharmacogenetic studies and valuable markers for peripheral moderator biomarkers discovery.

    Journal:  Development of electrospun lignin nanofibers for the adsorption of pharmaceutical contaminants in wastewater. (Pubmed Central) -  Jun 8, 2020   
    In this study, nanofibers of alkali lignin and a co-polymer, poly(vinyl alcohol), were developed and tested on the adsorption of a pharmaceutical contaminant (fluoxetine) in an aqueous solution. Results showed that the lignin nanofibers, of 156 nm in diameter, adsorbed 70% of fluoxetine in solution which corresponds to 32 ppm of contaminants removed in water.

    It was observed that commercially available GAC performed much better than biochar for all the compounds considered. Even at 1% of obtained capacity, biochar amendment to soils where reclaimed water is used for irrigation can reduce the uptake of these compounds by plants.

    Western blot showed that cofilin 1 expression is dynamically regulated during postnatal life, reaching a peak at the critical period, and decreasing at adult stage, and that it increases in fluoxetine-treated vs. untreated adult mice. In summary, by using a 2D gel electrophoresis differential approach on basic proteins followed by mass spectrometry and immunoblot analysis, we identified cofilin 1 as a potential candidate controlling plasticity levels of the mouse visual cortex.

    Long-term in vivo treatment with fluoxetine and sertraline promoted: (a) an enhancement of rat distal cauda spontaneous contractions; (b) a potentiation of phenylephrine-induced contractions; (c) a decreased in serum testosterone levels; and (d) a diminished daily sperm production, sperm reserves trough epididymis and sperm transit time in rat cauda epididymis. In conclusion, the alteration in the motor activity of epididymis could be associated to the low sperm count in this organ and accelerated transit time trough epididymal cauda of rats.

    Importantly, targeting this newly identified AR-mediated miR-204-5p/BDNF/AKT/MAPK signaling with small molecules including 7,8-DHF and fluoxetine, all led to alter the depressive-like behavior in AR knockout mice under CMS exposure. Together, results from these preclinical studies conclude that decreased AR may accelerate the stress-induced MDD via altering miR-204-5p/BDNF/AKT/MAPK signaling, and targeting this newly identified signaling may help in the development of better therapeutic approaches to reduce the development of MDD.

    Starting from T1, a significant decrease in 5-HT levels was observed. Our data suggest that fluoxetine (1.5 mg/kg/day) when associated with behavior treatment is effective in controlling canine aggression over a six-month period, and that, in dogs norfluoxetine levels seem reliable in predicting clinical efficacy.

    In the present study, we characterised pulmonary and hippocampal dysfunction by researching the interaction between inflammasome-regulated cytokines and glucocorticoid receptor (GR) signalling by investigating the role of fluoxetine (FLU), one of the most widely used antidepressants in clinical practice...These alterations were reversed by chronic FLU treatment, indicating that FLU-mediated GR signalling was involved in the COPD induced inflammasome activation. Our research explored the underlying molecular mechanism of comorbid COPD/depression and provided in vivo evidence that glucocorticoid resistance occurred during CS-induced central nervous system inflammation, a potential mechanism underlying the cross talk between the lung and brain.

    Introduction According to evidence from the literature, valproate and opioid substitution therapy may be the first line in the treatment of patients with bipolar I disorder (BDI) and opioid use disorder (OUD), while quetiapine and lithium may be not effective in this population [1,2]...Results The first patient, 34-year old, male, diagnosed with BDI, was receiving olanzapine and sodium valproate, for a severe major depressive episode with psychotic features...The third patient, 32-year old, male, diagnosed with BDI, received treatment with quetiapine and fluoxetine for 8 weeks, for a major depressive episode, severe, without psychotic features...After 3 months, one patient was abstinent, while the other two admitted periods of naltrexone discontinuation and opioid use. Conclusion Naltrexone could be combined with current BDI treatment in order to control OUD in patients with dual diagnosis, but the therapeutic adherence is an important obstacle in reaching the opioid abstinence.

    These variables are considered necessary for the early detection of possible reasons for non-adherence related to poor tolerability, and monthly evaluation is recommended during the entire duration of the treatment. Conclusion The use of psychometric instruments and biological variables should accompany the regular clinical interview focused on adverse events in patients with BD undergoing pharmacological treatment, in order to increase their adherence.
