  • ||||||||||  Preclinical, Journal:  Hippocampal LASP1 ameliorates chronic stress-mediated behavioral responses in a mouse model of unpredictable chronic mild stress. (Pubmed Central) -  Nov 28, 2020
Here, using an unpredictable chronic mild stress (UCMS) paradigm, we found that the mRNA and protein levels of LASP1 were decreased in the hippocampus of stressed mice and that UCMS-induced down-regulation of LASP1 was abolished by chronic administration of fluoxetine...It also restored the dendritic spine density, elevated the levels of AKT (a serine/threonine protein kinase), phosphorylated-AKT, insulin-like growth factor 2, and postsynaptic density protein 95. These findings suggest that LASP1 alleviates UCMS-provoked behavioral defects, which may be mediated by an enhanced dendritic spine density and more activated AKT-dependent LASP1 signaling, pointing to the antidepressant role of LASP1.

Journal:  Widespread monitoring of chiral pharmaceuticals in urban rivers reveals stereospecific occurrence and transformation. (Pubmed Central) -  Nov 22, 2020
Due to the considerable contribution by TPs (64% in present study) as well as the unexpected impacts from enantiomeric existence, the stereoselectivity of chiral pollutants during environmental process should be taken into account in future study. To the best of the authors' knowledge, it is the first comprehensive evaluation of chiral pharmaceuticals and transformation products at enantiomeric level in aquatic environment in China, which would facilitate better understanding of their environmental fate.

Journal:  Short- and Long-Term Repeated Forced Swim Stress Induce Depressive-Like Phenotype in Mice: Effectiveness of 3-[(4-Chlorophenyl)Selanyl]-1-Methyl-1H-Indole. (Pubmed Central) -  Nov 18, 2020
In protocol 1, CMI [1 and 10 mg/kg, intragastric (i.g.) route] or fluoxetine, a positive control (10 mg/kg, i.g. route), were administered 30 min before of sections of repeated FSS in both days of stress...Mice subjected to repeated FSS had an increase in the corticosterone levels and CMI regulated the levels of this glucocorticoid. These findings demonstrate that CMI was effective to abolish the depressive-like behavior induced by repeated FSS, which was accompanied by changes in the corticosterone levels and oxidative stress of prefrontal cortices and hippocampi of mice.

1960-1980), and the so-called "Prozac revolution" emerging at the end of the twentieth century...During this period, concordance between patient perceptions of postnatal depression and clinical understandings of the condition has been variable. Highlighting the detriment to therapeutic relationship when discordance is present, the narrators have demonstrated the need to destigmatize illness and facilitate cooperation between physician and patient and remind clinicians of the importance of placing patient experience at the center of care.

Journal:  Psychotropic Drugs Show Anticancer Activity by Disrupting Mitochondrial and Lysosomal Function. (Pubmed Central) -  Nov 18, 2020
The cytotoxicity of psychotropic drugs with cationic amphiphilic structures relied on simultaneous mitochondrial and lysosomal disruption and induction of cell death that not necessarily requires apoptosis. Since dual targeting of lysosomes and mitochondria constitutes a new promising therapeutic approach for cancer, particularly those in which the apoptotic machinery is defective, these data further support their clinical development for cancer therapy.

The current available literature has not shown any differences in efficacy between haloperidol and second-generation antipsychotics in patients with delirium...While suvorexant administered nightly to elderly patients in acute care settings may lower the incidence of delirium, larger studies are needed to confirm this finding...While the evidence supporting trazodone's use is limited, it is generally well tolerated and is an option as a PRN for irritability and agitation in patients with Alzheimer's and mixed dementia...It is important to note that memantine can cause or exacerbate agitation and may be the cause of behavioral dysregulation...Escitalopram and citalopram do have a unique pharmacokinetic properties in the sense that they have been found to have 50-56% plasma protein binding, compared to sertraline, fluoxetine, and paroxetine (95% or more)...Depression can be fatal as the rates of suicide are higher in later life than in any other age group. Consultation can improve the management of elderly patients and prevent unnecessary nursing home placement.

Journal:  Long-Term Pharmaceutical Contamination and Temperature Stress Disrupt Fish Behavior. (Pubmed Central) -  Nov 14, 2020
Accordingly, we investigated effects of long-term exposure to the pervasive pharmaceutical contaminant fluoxetine (Prozac) and acute temperature stress on reproductive behaviors and activity levels in the guppy (Poecilia reticulata)...Under cold-temperature stress, both sexes were less active and males exhibited less frequent reproductive behaviors. Our results demonstrate that long-term exposure to a common pharmaceutical pollutant and acute temperature stress alter fundamental fitness-related behaviors in fish, potentially shifting population dynamics in contaminated ecosystems.

Journal:  Perinatal fluoxetine exposure disrupts the circadian response to a phase-shifting challenge in female rats. (Pubmed Central) -  Nov 12, 2020
Our results demonstrate that long-term exposure to a common pharmaceutical pollutant and acute temperature stress alter fundamental fitness-related behaviors in fish, potentially shifting population dynamics in contaminated ecosystems. Perinatal FLX exposure altered the response to a phase-shifting challenge in female rats, whether this may pose health risks remains to be investigated.
  • ||||||||||  Journal:  Characteristics of drowning deaths in an inner city river. (Pubmed Central) -  Nov 12, 2020
Although the numbers of cases were not high, the urban portion of the River Torrens had a much higher number of drowning events per kilometre compared to other inland waterways in South Australia such as the Murray River. This is most likely due to the vulnerability that exists for intoxicated individuals in the city from falls into the water and to the availability of the river as a means of suicide to members of the adjacent urban population.

Review, Journal:  The Current Recommended Drugs and Strategies for the Treatment of Coronavirus Disease (COVID-19). (Pubmed Central) -  Nov 1, 2020
Co-administration of fluoxetine in the treatment of COVID19 could be considered due to the possibility of its interaction with ACE2 receptors, immune-modulatory function, and a proper immune response at the right time. Fluoxetine plays a beneficial role in reducing stress due to fear of infecting by COVID19 or worsening the disease and psychological support for the affected patients.

Journal:  Phasic and tonic serotonin modulate alarm reactions and post-exposure behavior in zebrafish. (Pubmed Central) -  Oct 30, 2020
Acute fluoxetine dose-dependently decreased fear-like behavior, but increased post-exposure freezing...Finally, CAS was shown to decrease the activity of monoamine oxidase in the zebrafish brain after exposure. These results suggest that phasic and tonic serotonin encode an aversive expectation value, switching behavior towards cautious exploration/risk assessment/anxiety when the aversive stimulus is no longer present.

Journal:  Hexosomes as Efficient Platforms for Possible Fluoxetine Hydrochloride Repurposing with Improved Cytotoxicity against HepG2 Cells. (Pubmed Central) -  Oct 30, 2020
The anionic OFH-HEX with a particle size of 145.5 ± 2.5 nm and drug EE% of 45.4 ± 1.2% was able to prolong the in vitro FH release, where only 19.5 ± 2.3% released in phosphate-buffered saline (PBS) pH 7.4 after 24 h. Contrarily, HEX rapidly released FH in acetate buffer pH 5.5 and achieved a 90.5 ± 4.7% release after 24 h. The obtained HEX showed an improved cellular internalization in a time-dependent manner and enhanced the cytotoxicity (2-fold higher than FH solution). The current study suggests the potential of FH-HEX as a possible anticancer agent against hepatocellular carcinoma.

Preclinical, Journal:  Involvement of mTOR-related signaling in antidepressant effects of Sophoraflavanone G on chronically stressed mice. (Pubmed Central) -  Oct 27, 2020
Chronic Unpredictable Mild Stress (CUMS) mice were treated with fluoxetine and SG...The blockade of mammalian target of rapamycin signaling blunted the antidepressant effect and reversed the up-regulation of BDNF expression caused by SG. These findings suggested that SG treatment alleviated depressive-like symptoms via mTOR-mediated BDNF/Trkb signaling.
