    Journal:  Sex-specific effects of psychoactive pollution on behavioral individuality and plasticity in fish. (Pubmed Central) -  Nov 16, 2023   
    In sharp contrast, effects on behavioral plasticity were observed in females-but not in males-whereby exposure to even low levels of fluoxetine resulted in a substantial decrease (activity) and increase (freezing behavior) in the behavioral plasticity of females. Our evidence reveals that psychoactive pollution has sex-specific effects on the individual behavior of fish, suggesting that males and females might not be equally vulnerable to global pollutants.

We describe a novel association of de novo missense variants in KCNA3 with a human DEE, and provide evidence that fluoxetine might represent a potential targeted treatment for individuals carrying variants with significant GoF effects.

    In addition, altered levels of neurochemicals belonging to the serotonergic and dopaminergic systems (increased levels of tryptophan and norepinephrine) highlighted the sensitivity of early life stages of zebrafish to low concentrations of fluoxetine, inducing effects that may compromise larval survival. The obtained data support the necessity to test low concentrations of SSRIs in environmental risk assessment and the use of biomarkers at different levels of biological organization for a better understanding of modes of action.

Indeed, this evidence allows formulation of a mechanistic explanation of SSRI discontinuation which draws on parallels with the withdrawal states of other psychotropic drugs.

    Our results suggest that both rTMS and fluoxetine could benefit the CUMS-induced abnormal behaviors including depressive-like behaviors, and the beneficial effects of rTMS as well as fluoxetine on depression might be partly related to their neuroprotective effect on attenuating astroglial activation and BDNF decrease. Our findings may provide new insights into the cardiac and hepatic adverse effects of FLX.

    Collectively, our findings demonstrate that fluoxetine exerts potent anti-inflammatory effects, and suppresses the release of the endogenous itch mediator endothelins in human keratinocytes, most likely via indirect inhibition of PI3K. Thus, clinical studies are encouraged to explore whether the currently reported beneficial effects translate in vivo following its topical administration in inflammatory and pruritic dermatoses.

Moreover, our findings suggest that prior unsuccessful antidepressant treatment induces a "priming effect" that increases the likelihood of successful response to KET.

    Using SH-SY5Y neuroblastoma cells, U87MG astrocytoma cells, and pharmacologically relevant doses, we showed that atropine, azithromycin, fluoxetine, metoprolol, and tamoxifen (1) de-acidified endolysosomes, (2) decreased endolysosome Fe2+ levels (3) increased cytosolic and mitochondrial Fe2+ and ROS levels, (4) depolarized mitochondria membrane potential, and (5) induced cell death; effects all blocked by the endolysosome-iron chelator deferoxamine. These cell biology findings suggest that, in general, weak base pharmaceuticals can induce lysosome stress responses that might affect their safety profiles.

    One of the psychedelic compounds that was inactive in the NOG assay, psilocin, was active in the synaptogenesis assay on the other hand, indicating that the two assays in combination could be valuable tools to identify and elucidate the function of molecules with novel mechanisms.In conclusion, we have validated a high content NOG assay with excellent reproducibility and the ability to differentiate compounds based on their efficacy and potency. Using the two assays in combination will be a powerful approach to identify compounds with differential effects and understand their mechanism of action on neuroplasticity.
