Journal:  Raman and fluorescence imaging of phospholipidosis induced by cationic amphiphilic drugs in endothelial cells. (Pubmed Central) -  Feb 19, 2022
Interestingly, lysosomal volume expansion, occurrence of DIPL, and activation of autophagy induced by selected CADs all depended on drug-accumulation in acidic pH of lysosome cellular compartments whereas reduced endothelial viability did not, and was attributed to mitochondrial mechanisms as evidenced by a decreased mitochondrial transmembrane potential. In conclusion, drug-induced phospholipidosis in the endothelium did not reduce endothelial viability per se and can be efficiently assayed by Raman imaging.

Preclinical, Journal:  Change of hypothalamic adult neurogenesis in mice by chronic treatment of fluoxetine. (Pubmed Central) -  Feb 19, 2022
Interestingly, FLX treatment did not affect the total number of newly generated cells in the hypothalamus, comparing that in controls. These results suggest that FLX treatment influence hypothalamic adult neurogenesis and shift the balance between the numbers of neurons and astrocytes under studied conditions.

Journal:  Proteomics of sea bass skin-scales exposed to the emerging pollutant fluoxetine compared to estradiol. (Pubmed Central) -  Feb 17, 2022
A large number (103) of FLX-specifically regulated proteins indicated numerous actions independent of estrogen signalling. This study provides the first quantitative proteome of the fish skin-scale barrier, elucidates routes of action and biochemical and molecular signatures of E2 or FLX-exposure and identifies potential physiological consequences and candidate biomarkers of pollutant exposure, for monitoring and risk assessment.

Clinical, Journal:  Targeting glioblastoma signaling and metabolism with a re-purposed brain-penetrant drug. (Pubmed Central) -  Feb 16, 2022
Incorporation of real-world evidence from electronic medical records from insurance databases reveals significantly increased survival in GBM patients treated with fluoxetine, which was not seen in patients treated with other selective serotonin reuptake inhibitor (SSRI) antidepressants. These results nominate the repurposing of fluoxetine as a potentially safe and promising therapy for patients with GBM and suggest prospective randomized clinical trials.

Journal:  The Polysite Pharmacology of TREK K Channels. (Pubmed Central) -  Feb 16, 2022
This polysite pharmacology within a relatively small (~70 kDa) ion channel comprises four structurally defined modulator binding sites that occur above (Keystone inhibitor site), at the level of (K modulator pocket), and below (Fenestration and Modulatory lipid sites) the C-type selectivity filter gate that is at the heart of K function. Uncovering this rich structural landscape provides the framework for understanding and developing subtype-selective modulators to probe K function that may provide leads for drugs for anesthesia, pain, arrhythmia, ischemia, and migraine.
  • ||||||||||  Clinical, PK/PD data, Review, Journal:  A Review on Pharmacokinetic and Pharmacodynamic Drug Interactions of Adrenergic β-blockers with Clinically Relevant Drugs-An Overview. (Pubmed Central) -  Feb 15, 2022
The majority of the β-blockers including Propranolol, Metoprolol, Acebutolol, Alprenolol, Betaxolol, Carvedilol, Nebivolol and Oxprenolol are metabolised majorly by CYP2D6, and Bisoprolol is primarily metabolised by CYP3A4 enzymes...The plasma concentrations of Propranolol might be elevated by the concomitant use of drugs, such as SSRIs (Fluoxetine, Paroxetine), SNRIs (Duloxetine) and Cimetidine, while the plasma concentrations of Metoprolol increased by the concurrent use of SSRIs (Fluoxetine, Paroxetine), Amiodarone, Celecoxib, Cimetidine, Terbinafine, and Diphenhydramine...In addition to pharmacokinetics and pharmacodynamics of the drug interactions, recommendations for clinical practice are highlighted. The prescribers and the pharmacists are needed to be aware of the drugs interacting with β-blockers to prevent possible adverse drug interactions.

Journal:  Baicalin Ameliorates Chronic Unpredictable Mild Stress-Induced Depression through the BDNF/ERK/CREB Signaling Pathway. (Pubmed Central) -  Feb 15, 2022
There were decreases in fluoxetine (FLU) and BA groups (p < 0.05, p < 0.01)...The IOD value of the p-ERK and p-CREB in the CUMS group was decreased versus the CON group (p <  0.01), and these changes were improved via BA and FLU treatment (p <  0.05, p <  0.01). This study indicated that BA can improve cognitive functions and has antidepressant effects in mice, which may be associated with activation of the BDNF/ERK/CREB signaling pathway in the hippocampus.

Preclinical, Journal:  Behavioral and neurochemical effects of folic acid in a mouse model of depression induced by TNF-α. (Pubmed Central) -  Feb 15, 2022
Additionally, ERK1 phosphorylation was increased in mice treated with TNF-α + folic acid (1 mg/kg), but no effects on ERK2 or JNK1/2/3 phosphorylation were found in any group. The results indicate the efficacy of folic acid to counteract the depressive-like behavior induced by a pro-inflammatory cytokine, an effect that might be associated with the activation of monoaminergic systems, inhibition of N-methyl-D-aspartate (NMDA) receptors and nitric oxide (NO) synthesis, as well as Akt modulation.

Journal:  The Influence of CYP2D6 and CYP2C19 Genetic Variation on Diabetes Mellitus Risk in People Taking Antidepressants and Antipsychotics. (Pubmed Central) -  Feb 15, 2022
Among participants with diabetes who were taking fluoxetine, CYP2D6 intermediate metabolizers and decreased HbA1c, compared to normal metabolizers (mean difference -7.74 mmol/mol; p = 0.017)...Our results indicate that the impact of genetic variation in CYP2D6 differs depending on diabetes status. Although our findings support existing clinical guidelines, further research is essential to inform pharmacogenetic testing for people taking antidepressants and antipsychotics.

Journal:  Applications of the SR4G Transgenic Zebrafish Line for Biomonitoring of Stress-Disrupting Compounds: A Proof-of-Concept Study. (Pubmed Central) -  Feb 15, 2022
We employed zebrafish larva (transgenic SR4G line) with a cortisol-inducible green fluorescence protein reporter (eGFP) as a model to detect stress responses upon exposure to compounds with environmental impact, including bisphenol A (BPA), vinclozolin (VIN), and fluoxetine (FLX)...Hydrocortisone (CORT)and dexamethasone (DEX) were used as positive and negative controls, respectively...Our findings support the use of SR4G transgenic larvae as an in vivo biomonitoring model to screen chemicals for their stress-disrupting potentials. This is important because there is increasing evidence that brief exposures to environmental pollutants modify the stress response and critical coping behaviors for several generations.

Clinical, Review, Journal:  The pediatric psychopharmacology of autism spectrum disorder: A systematic review - Part I: The past and the present. (Pubmed Central) -  Feb 12, 2022
Clonidine and guanfacine display some efficacy on hyperactivity and stereotypic behaviors...Research in the pediatric psychopharmacology of ASD is still faced with at least two major hurdles: (a) Great interindividual variability in clinical response and side effect sensitivity is observed in the ASD population. This low level of predictability would benefit from symptom-specific treatment algorithms and from biomarkers to support drug choice; (b) To this date, no psychoactive drug appears to directly ameliorate core autism symptoms, although some indirect improvement has been reported with several drugs, once the comorbid target symptom is abated.

Clinical, Journal:  Effects of aqueous fluoxetine exposure on gut microbiome of adult Pimephales promelas. (Pubmed Central) -  Feb 9, 2022
These changes were associated with a decrease in relative proportions of commensal bacteria, which can be important for health of fish including bacteria essential for fatty acid oxidation, and an increase in relative proportions of pathogenic bacteria associated with inflammation. Results demonstrate, for the first time, how antidepressants found in some aquatic environments can influence gut microbiota of fishes.

Journal:  Multifunctional amino acids empowering bifunctional biosensing platform for depression study. (Pubmed Central) -  Feb 9, 2022
The obtained biosensor exhibited a remarkably low limit of detection (0.283 pg mL) and could thus sensitively identify the GRα differences in healthy and depressive rats with and without fluoxetine. The electrochemical biosensor developed herein was not only outstandingly sensitive but also simple to use and labor-saving, making it a promising all-in-one platform for depression diagnosis and drug discovery.

Preclinical, Journal:  Prenatal fluoxetine impairs non-hippocampal but not hippocampal memory in adult male rat offspring. (Pubmed Central) -  Feb 8, 2022
These results suggest that in utero exposure to fluoxetine induces detrimental effects on non-hippocampal memory and in remote retention of hippocampal-dependent memory, which is believed to be stored in the temporal cortex, possibly due to changes in cortical NMDA receptor subunits. The present results warrant the need for studies on potential remote memory deficits in human offspring exposed to fluoxetine in utero.

Preclinical, Journal:  Effects of social stress and fluoxetine treatment on fracture healing in a rat femur fracture model. (Pubmed Central) -  Feb 8, 2022
Although fluoxetine had no detrimental effect on fracture healing in non-depressive rats, impaired fracture healing was reversed and better radiological and histological findings were obtained in depressive rats treated with fluoxetine. Our findings indicate that fluoxetine, which minimizes the negative effects of social stress on bone healing, can be used safely in the treatment of depressive disorders in patients with fractures.
