All together, these results suggest that 5-HT2ARs mediate the head twitch response associated with hallucinogenic activity, but are detached from the BDNF/TRKB-dependent actions of psychedelics on plasticity and antidepressant-like behavioral responses.

    Consistent with this, we found increased expression of the phosphoinositide PIP3(phosphatidylinositol-(3,4,5)-trisphosphate) in the soma of tectal neurons following exposure to fluoxetine, and inhibiting PIP3 synthesis using the PI3-K inhibitor LY294002 abolished the potentiating effect of fluoxetine on retinotectal transmission. Given that PIP3 has been shown to be important for postsynaptic AMPA receptor clustering and long-term potentiation, our data indicate that endogenous serotonin plays a role in the maturation of retinotectal synapses through PI3-K signaling.

    Given that PIP3 has been shown to be important for postsynaptic AMPA receptor clustering and long-term potentiation, our data indicate that endogenous serotonin plays a role in the maturation of retinotectal synapses through PI3-K signaling. These data indicate that the opioid system is involved in the antidepressant effects of fluoxetine, and this effect may be mediated through the upregulation of proenkephalin in a subpopulation of mature granule cells.

    Further, using immunohistochemistry, we found that sash mice show reduced serotonin+ cells in the dorsal raphe, and preliminary data suggest that the anxiety-like behavior in sash mice can be reduced with only a single IP injection of either 1mg/Kg ketamine (NMDAR antagonist) or 20mg/Kg fluoxetine (serotonin transporter inhibitor). Together, these results suggest that brain MCs play a fundamental role in preventing exaggerated anxiety responses through modulation of serotonergic/BDNF signaling, providing a novel mechanism underlying anxiety-like responses to stress.

    All experiments were repeated 4-6 times in triplicate.Antidepressants, psychedelics and ketamine all show an antidepressant biosignature, revealing amobilization of Gsα from lipid rafts and an increased signaling of GPCRs working through Gsα.Previous studies in postmortem brain and in platelets showed that this biomarker was enriched in lipidrafts from depressed subjects and reverted toward control values after effective treatment (as indicated byHAM-D). The similarities and differences in the actions of these drugs at the cellular level may lead to abetter molecular understanding of the distinct kinetic features of their actions.

    We are currently exploring changes in gut microbiota composition in male and female BALB/c mice after chronic treatment with fluoxetine or citalopram followed by microbiome profiling using 16s rRNA sequencing from fecal sample, including analysis of alpha (within community diversity) and beta (between communities) diversity across three-time points: baseline, midpoint, and after 28 days of SSRI administration to then compare between groups. The results of these experiments may help shed light on the possible mechanism of action of these SSRIs in the gut microbiome and the potential differences between males and females in therapy outcomes.

    The ketamine-like glutamatergic antagonist RO-25-6981 exerts both rapid and sustained antidepressant-like activity...To assess antidepressant-like behaviors, the tail suspension test (TST) and locomotor activity (LA) tests were performed using RO-25-6981 and its analogs with traditional antidepressant drugs, including the serotonin selective reuptake inhibitor fluoxetine and the tricyclic antidepressant desipramine, as positive controls for monoaminergic reuptake activity...Taken together, these data suggest that the antidepressant-like activity of RO-25-6981 and its analogs does not correlate with the degree of NMDA receptor antagonism. Furthermore, these data point to serotonergic reuptake inhibition contributing to the overall antidepressant-like activity of RO-25-6981 in animal models of mood.

    These findings suggest that acute treatment with ketamine or inhibition of mGlu2/3 can normalize frontal low- and high-frequency EEG activity, suggesting that EEG activity may be used as a biomarker for treatment response. Further, inhibition of mGlu2/3 is as effective as ketamine in reversing reward learning impairments commonly observed in MDD and other psychiatric disorders characterized by anhedonia, but with fewer psychotomimetic effects, making it a potentially safer alternative treatment for anhedonia.

In sum, this work implicates histone serotonylation in DRN as an important mediator of stress vulnerability via alterations in serotonergic signaling dynamics and related gene expression programs.

    The method was fully validated with an acceptable accuracy of 75.63-108.21%, matrix effect of 80.41-117.71% except for fenofibrate (76.07% at low concentration levels), and precision of 0.32-13.12%. Owing to the advantages of simple operation, high accuracy and sensitivity, this method is suitable for the rapid and simultaneous detection of 12 anti-obesity drugs in human plasma, providing support for clinically monitoring the development of adverse reactions and guiding the rational and appropriate use of weight-loss drugs for obese people.

    Our results showed fluoxetine attenuated PPI deficit and the decrease of cerebral BDNF expression in the MK-801-injected mice. These results suggest that fluoxetine can be used to treat sensorimotor gating deficit in a neurodevelopmental mouse model of schizophrenia, and the attenuating effect of fluoxetine on sensorimotor gating deficit may be related to fluoxetine's neuroprotective effect targeting on the modulation of cerebral BDNF.

Our findings suggest that BTVCs can alleviate depressive-like behaviours, restore damage to the hippocampus in CUMS rats and regulate serum metabolism, which may be related to endothelin-1 signalling or CREB signalling in neurons.

    Exposure to exogenous serotonin re-established the phototactism and locomotor activity of TRH- individuals to those of the wild type but did not affect phototactic responses to fish kairomones. Unexpectedly, fluoxetine was able to modify locomotor activity and phototactic behavior against fish kairomones in TRH- individuals lacking serotonin, and also it increased the concentrations of acethylcholine and GABA in exposed animals, which support the argument that fluoxetine may also affect other neurological pathways.

    The concentrations of the psychoactive drugs diphenhydramine, fluoxetine, doxepin, imipramine, sulpiride, zolpidem, carbamazepine, and flunitrazepam in the wastewater were determined...We found associations between the use of the three drugs and age, occupation, and obesity, which are risk factors for mental disorders. The results showed that the monitoring of psychotropic drug using WBE has a certain reference value for public health care and for improving the understanding of mental disorders.
