Preclinical, Journal:  Mice with a Pax2 missense variant display impaired glomerular repair. (Pubmed Central) -  Mar 14, 2024
Lastly, Pax2A220G/+ mice have increased numbers of glomerular tuft cells expressing Ki67 and CC3 compared to wild type after Adriamycin injury, consistent with maladaptive responses to podocyte loss. Collectively, our results suggest that decreased glomerular numbers in Pax2A220G/+ mice are likely compounded with the inability of their mutated PECs to regenerate podocyte loss, and together these two mechanisms drive the worsened FSGS phenotype in these mice.

Journal:  Metabolic engineering of Streptomyces peucetius for biosynthesis of N,N-dimethylated anthracyclines. (Pubmed Central) -  Mar 14, 2024
DoxA is the key enzyme that determines the efficiency of the biosynthesis of N,N-dimethylated anthracyclines, and engineering of this enzyme will be a major step forwards towards the efficient production of more N,N-dimethylated anthracyclines, including N,N-dimethyldoxorubicin. This study provides valuable insights into the biosynthesis of clinically relevant daunorubicin derivatives, highlighting the importance of combinatorial biosynthesis.

Journal:  Injectable Chitosan Hydrogels Doped with 2D Peptide Nanosheet-Drug Conjugates for Glutathione-Responsive Sustained Drug Delivery. (Pubmed Central) -  Mar 13, 2024
Covalent attachment of thiol-modified doxorubicin (DOX) drugs to 2D PNSs via disulfide bonds results in the peptide-drug conjugates (PDCs), which is denoted as PNS-SS-DOX...Meanwhile, the covalently linked disulfide bond within PNS-SS-DOX is responsive to the intracellular glutathione (GSH) within tumor cells, enabling controlled drug release and significantly inhibiting cancer cell growth. This responsive peptide-drug conjugate based on a 2D peptide nanoplatform paves the way for the development of smart drug delivery systems and has bright prospects in the future biomedicine field.

Journal:  Anthracyclines induce cardiotoxicity through a shared gene expression response signature. (Pubmed Central) -  Mar 13, 2024
The most widely used TOP2i include anthracyclines (AC) Doxorubicin (DOX), Daunorubicin (DNR), Epirubicin (EPI), and the anthraquinone Mitoxantrone (MTX)...To investigate this, we studied the effects of treatment of DOX, DNR, EPI, MTX, and an unrelated monoclonal antibody Trastuzumab (TRZ) on iPSC-derived cardiomyocytes (iPSC-CMs) from six healthy females...Two thirds of the genes that respond to at least one TOP2i, respond to all ACs with the same direction of effect. Our data demonstrate that TOP2i induce thousands of shared gene expression changes in cardiomyocytes, including genes near SNPs associated with inter-individual variation in response to DOX treatment and AC-induced cardiotoxicity.

Journal:  Designing Molecularly Imprinted Polymer-Modified Boron-Doped Diamond Electrodes for Highly Selective Electrochemical Drug Sensors. (Pubmed Central) -  Mar 12, 2024
In this study, we selected the anticancer drug doxorubicin (DOX) as the template molecule...Other drugs, i.e., mitomycin C or clonazepam (CZP), also underwent a reduction reaction at a similar potential to that of DOX, when using the unmodified BDD, which rendered the accurate quantification of DOX in a mixture challenging...Selective DOX measurement using the MIP-BDD was also possible in human plasma, and the respective limits of detection of DOX in PBS and human plasma were 32.10 and 16.61 nM. The MIP-BDD was durable for use in six repeated measurements, and MIP-BDD may be applicable as an electrochemical sensor for application in therapeutic drug monitoring.

The high expression of E2F7, regulated by activated p53, confers chemoresistance to GBM cells by inhibiting drug uptake and DNA damage. These findings highlight the significant connection between sustained p53 activation and GBM chemoresistance, offering the potential for new strategies to overcome this resistance.

Super-soft hydrogels is prepared through co-assembly of DNA chains and magnesium pyrophosphate during rolling-circle amplification (RCA), followed by subsequent loading of antioxidant curcumin, and antibiotic ciprofloxacin through specific intermolecular interactions between DNA and drugs...By sequentially incorporating DNA hydrogel with immune checkpoint inhibitor aPDL1 and chemotherapeutic agent doxorubicin (DOX), a chemo-immunotherapeutic DNA hydrogel system is formed...DOX is first released to induce immunogenic cell death that intimately works together with the CpG ODN in gel scaffold for effectively recruiting and activating dendritic cells. The subsequently released aPDL1 antibody from the expanded gel matrix blocks the immune-inhibitory PD-L1 molecule on the tumor cell surface, thus sensitizing the tumor to antitumor T-cell immunity.
