    Human induced pluripotent stem cells (iPSCs) with a stable genomic insertion of the neurogenin-2 transcription factor under the control of the TetOn promoter can be differentiated into excitatory human neurons (i3Neurons) within 3 days of exposure to doxycycline...The assay is rapid, with results obtained after a one-week-long i3Neuron culture or one month if neurons are co-cultured with astrocytes. This live-cell, mitochondrial phenotypic assay can be used for high-throughput screening or as an orthogonal assay for compounds obtained via other high-throughput screening campaigns.

    Using Drosophila white as the initial model, we demonstrated that introduction of a single insertion by CRISPR/Cas9 created a null mutation, a tagged rescue construct, which could be induced with doxycycline, and which allowed assessment of protein expression...Thus, this system avoids the need to perform genetic crosses to introduce an inducible rescue transgene in a mutant background, or to combine driver and reporter lines to examine expression of the targeted protein. We propose that DREaMR provides a system that can be applied to additional mosquito vectors as well as other organisms in which CRISPR/Cas9 is effective.

    Here, we comprehensively tested prime editing in hPSCs by generating a doxycycline-inducible prime editing platform...Finally, whole-genome sequencing showed that, unlike the cytidine deaminase domain of cytosine base editors, the reverse transcriptase domain of a prime editor does not lead to guide RNA-independent off-target mutations in the genome. Our results demonstrate that prime editing in hPSCs has great potential for complementing previously developed CRISPR genome editing tools.

    We compared the development of narcoleptic symptomatology in male vs. female orexin-tTA; TetO-DTA mice, a model in which Hcrt neuron degeneration can be initiated by removal of doxycycline (DOX) from the diet...During degeneration, both sexes also exhibited the "Delta State", characterized by sudden cessation of activity, high delta activity in the EEG, maintenance of muscle tone and posture, and the absence of phasic EMG activity. Since the phenotypes of the two sexes were indistinguishable by Week 6, we conclude that both sexes can be safely combined in future studies to reduce cost and animal use.

    Oral enterococci carry virulence genes and express resistance to some antibiotics commonly used in dentistry, such as ciprofloxacin and erythromycin. Specific subgingival taxa are associated with oral enterococci, suggesting they may interact with species of the dysbiotic periodontitis biofilm, constituting a potential source of factors to tissue destruction, antibiotic resistance dissemination and poor response to periodontal therapy.

    In the current study, we used doxycycline-inducible astrocyte-specific HIV Tat transgenic mice (iTat), a surrogate HAND model, and determined effects of PNU-125096, a positive allosteric modulator of α7 nicotinic acetylcholine receptor (α7 nAChR) on Tat-induced behavioural impairments and neuropathologies...Lastly, we used primary mouse cortical individual cultures and neuron-astrocytes co-cultures and in vivo staining of iTat mouse brain tissues and showed that glial activation was directly involved in the interplay among Tat neurotoxicity, α7 nAChR activation, and p38 MAPK signaling pathway. Taken together, these findings demonstrated for the first time that α7 nAChR activation led to protection against HAND and suggest that α7 nAChR modulator PNU-125096 hold significant promise for development of therapeutics for HAND.

Our case highlights an uncommon tumor type in the setting of progressive respiratory symptoms refractory to medical therapy, demonstrates successful endobronchial management, and suggests watchful waiting for optimization prior to surgical resection can be effectively performed in this low grade tumor.

    Etiology is familial children but secondary in adulthood, where triggers include viral infection, cancers.Case presentation: A 60-year-old immunocompetent female with past medical history of hypertension, asthma, deep vein thrombosis on warfarin, hypothyroidism, and hyperlipidemia who presented due to fatigue and encephalopathy...She completed two separate month treatments with doxycycline for Lyme disease (despite negative serology) but had continued intermittent symptoms...She was empirically treated with antimicrobials, Dexamethasone 10 mg/m2 body surface area and etoposide...EBV serology was completed and was positive with viral load exceeding 5 million, thus rituximab was empirically started.She continued to have mucosal bleeding with increasing amounts of frank blood being passed from her ET tube...Upon removal of the bronchoscope, she promptly arrested, with no attempt at resuscitation made on the second arrest.Summary: Described is a rare case of HLH that was secondary to severe EBV infection masquerading as Lyme disease in an immunocompetent individual. Even with rapid initiation of empiric treatment and supportive measures, she expired in the setting of severe coagulopathy leading to respiratory compromise secondary to endobronchial hemorrhage.Figure 1: Bone marrow aspirate.

Symptomatic improvement in such patients is quite impressive with steroids with complete recovery in most of the patients.

    Herein we present a patient with complete collapse of the right lung complicated with post-obstructive necrotizing pneumonia and septic shock secondary to an endobronchial carcinoid tumor causing central airway obstruction.CASE DESCRIPTION: A 19-year-old male with a history of asthma managed with albuterol and montelukast presented with fever, acute hypoxic respiratory failure, and septic shock to a local emergency room...He was started on IV fluids, vasopressors, oxygen supplementation, and empiric vancomycin, piperacillin-tazobactam, and doxycycline...Bronchoscopic biopsies are usually diagnostic, and imaging modalities such as CT scan can aid in establishing the diagnosis. As both asthma and endobronchial tumors present with obstructive physiology, the diagnosis of such neoplasms can be delayed in patients with history of asthma.

While historically considered a contaminant, corynebacteria may present a serious infectious threat and should be approached accordingly.

This case brings to light that is extremely important to treat empirically if the diagnosis is unknown to save lives!

    GOLD guidelines allow for various antibiotic choices (e.g., aminopenicillin with clavulanic acid, macrolide or tetracycline) and hospitalized patients with AECOPD receive a variety of antibiotics...Of the 31 patients who received azithromycin during their index admission, 51.6% had 1 or more readmissions within 60 days compared with 55.5% of 45 patients who received levofloxacin and 59.5% of 42 patients who received doxycycline (p=0.80 by Chi-square)...Patients received the same antibiotics when readmitted within 60 days, occasionally over multiple readmissions. Antibiotic selection did not influence outcomes in patient’s frequently hospitalized for AECOPD, but specific guidelines for antibiotic usage are needed for these patients.

    Here, we employ a doxycycline-inducible PRL-3 mouse strain to show that aberrant PRL-3 expression within a non-cancerous background leads to the death of Lgr5+ ISCs and to Paneth cell expansion...The impaired intestinal organoid formation was rescued by a PRL inhibitor, providing a functional link to the observed phenotypes. These results demonstrate that elevated PRL-3 phosphatase activity in healthy intestinal epithelium impairs intestinal cell homeostasis, which correlates this cellular mechanism of tumor onset with PRL-3-mediated higher susceptibility to tumor formation upon inflammatory or mutational events.Key messages• Transgenic mice homozygous for PRL-3 overexpression die early.• PRL-3 heterozygous mice display disrupted intestinal self-renewal capacity.• PRL-3 overexpression alone does not induce tumorigenesis in the mouse intestine.• PRL-3 activity leads to the death of Lgr5+ ISCs and Paneth cell expansion.• Impairment of cell homeostasis correlates PRL-3 action with tumor onset mechanisms.
