Journal:  Bicyclomycin Activity against Multidrug-Resistant Gram-Negative Pathogens. (Pubmed Central) -  Dec 20, 2022
More significant is the synergistic growth-inhibitory effect and the enhancement of killing by the additional presence of doxycycline since this increases the in vivo efficacy. These data demonstrate that bicyclomycin-containing regimens have potential as new treatment options for MDR Gram-negative infections such as those caused by CRE, E. coli, and K. pneumoniae.

Journal:  The impact of Post-Transplant doxycycline in AL amyloidosis - updated results after Long-Term follow up. (Pubmed Central) -  Dec 20, 2022
There was a minimal trend towards improved survival with doxycycline among patients with ≥ very good partial response and among patients with organ response that was not statistically significant. After long-term follow-up, there is no clear evidence to support benefit of doxycycline in the post-transplant setting.

Clinical, Clinical data, Review, Journal:  Does prophylactic application of doxycycline at the implant-abutment interface improve clinical outcomes of dental implants? (Pubmed Central) -  Dec 18, 2022
Marginal bone levels and pocket probing depths after 6 and 12 months on the test side were less compared to the control side and these differences were statistically significant. The test side also showed fewer implants with bleeding on probing and lower risk of peri-implant mucositis after 3, 6 and 12 months. Conclusion Prophylactic placement of doxycycline hyclate may reduce peri-implant bone loss and pocket probing depths, and may also reduce the risk of peri-implant mucositis.

Journal:  Three-Fluid Nozzle Spray Drying Strategy for Efficient Fabrication of Functional Colloidosomes. (Pubmed Central) -  Dec 16, 2022
Doxycycline hydrochloride (DH) was encapsulated in colloidosomes for controllable release, and the sustained release time is up to 100 h. The release rate can be adjusted by varying the glass transition temperature (T) and size of latex particles...The magnetic response intensity of colloidosomes can be modulated by varying the amount of FeO NPs. The anticancer drug encapsulation and loading of other functional particles were also explored to expand applications.

Journal:  Normocomplementemic urticarial vasculitis secondary to Lyme disease: A rare association with challenging treatment. (Pubmed Central) -  Dec 16, 2022
Response to cyclosporine was satisfactory but with side effects, and only methotrexate showed substantial and consistent improvement...Possible triggers for this disease must be sought, even if rarely described, such as LD. Normocomplementemic UV frequently presents a therapeutic challenge, but methotrexate can be a particularly effective therapy in this setting.
  • ||||||||||  Journal:  Ocular surface squamous neoplasia in a patient following oral steroids for contralateral necrotising scleritis. (Pubmed Central) -  Dec 16, 2022
His ocular history was significant for pterygium excision with mitomycin C 3 months prior in the affected eye, open-angle glaucoma, nuclear sclerosis cataract and previous sixth cranial nerve palsy...The patient was treated for necrotising scleritis with oral doxycycline, vitamin C, ranitidine and prednisone with gradual taper over 45 days...He was treated with topical interferon alpha-2b 1 million IU qid and achieved complete resolution after 4 months. The case highlights both the acute precipitation of OSSN following oral steroids and a sight-threatening complication following pterygium excision.

Preclinical, Journal, PD(L)-1 Biomarker, IO biomarker:  Human stem cell derived beta-like cells engineered to present PD-L1 improve transplant survival in NOD mice carrying human HLA class I. (Pubmed Central) -  Dec 13, 2022
NOD.HLA-A2.1 mice, which lack classical murine MHC I and instead express human HLA-A*02:01, underwent transplantation of 1,000 human HLA-A*02:01 sBCs under the kidney capsule and were separated into HLA-A2 positive iP-sBC and HLA-class I negative iP-BKO sBC groups, each with +/- doxycycline (DOX) induced PD-L1 expression...These results show that T cells rapidly infiltrate and attack sBC grafts in this model but that significant numbers of PD-L1 expressing sBCs manage to survive in this harsh immunological environment. This investigation represents one of the first in vivo studies recapitulating key aspects of human autoimmune diabetes to test immune tolerance approaches with renewable sources of beta cells.
  • ||||||||||  Review, Journal:  Antibiotic Resistance in Patients with Peri-Implantitis: A Systematic Scoping Review. (Pubmed Central) -  Dec 12, 2022
Other microorganisms such as Tannerella forsythia, Parvimonas micra, and Prevotella intermedia/nigrescens also presented significant levels of resistance to other antibiotics including amoxicillin, azithromycin, and moxifloxacin...Although the management of adjunctive antimicrobials in the therapy of PI is controversial, in this review, the resistance of relevant microorganisms to antibiotics used to treat PI, and usually prescribed in dentistry, was observed. Clinicians should consider the antibiotic resistance demonstrated in the treatment of PI patients and its public health consequences.

Journal, BRCA Biomarker:  FOXO3a Mediates Homologous Recombination Repair (HRR) via Transcriptional Activation of MRE11, BRCA1, BRIP1, and RAD50. (Pubmed Central) -  Dec 12, 2022
The mRNA levels of the recombined GFP increased after doxycycline treatment in MEF but not in wt-MEF, and increased in young HDF comparing to old HDF, indicating that FOXO3a activates HRR. Overall, these results demonstrate that MRE11, BRCA1, BRIP1, and RAD50 are transcriptional target genes for FOXO3a, and HRR activity is increased via transcriptional activation of MRE11, BRCA1, BRIP1, and RAD50 by FOXO3a.

Journal:  Yap governs a lineage-specific neuregulin1 pathway-driven adaptive resistance to RAF kinase inhibitors. (Pubmed Central) -  Dec 9, 2022
YAP governs adaptive resistance to RAF kinase inhibitors and induces a gene expression program in BRAF-mutant cells encompassing effectors in the NRG1 signaling pathway, which play a central role in the insensitivity to MAPK inhibitors in a lineage-dependent manner. HIPPO pathway inactivation serves as a lineage-dependent rheostat controlling the magnitude of the adaptive relief of feedback responses to MAPK inhibitors in BRAF- cancers.

Review, Journal:  Bullous pemphigoid in a patient with a neuropsychological disorder and a possible novel drug trigger: A case report and review of the literature. (Pubmed Central) -  Dec 9, 2022
Nevertheless, new blisters continued developing, hence dapsone 50 mg was introduced, with significant improvement...Furthermore, as there was an association of empagliflozin initiation and BP onset, this further complicates the aetiology and presents a potential novel drug cause of BP. This case emphasises the neuropsychological issues associated with managing complex BP cases, a possible novel cause of drug-induced BP and highlights the likelihood of these issues becoming increasingly prevalent for the future.

Journal:  Routine Donor and Recipient Screening for Mycoplasma hominis and Ureaplasma Species in Lung Transplant Recipients. (Pubmed Central) -  Dec 6, 2022
Pretransplant recipient urine screening had a low yield and was not correlated with post-transplant Mollicute infection, likely because most M. hominis and U. parvum/urealyticum infections in lung transplant recipients are donor-derived. Routine donor bronchus swab PCR for M. hominis, U. urealyticum, and U. parvum followed by preemptive therapy did not obviously impact the overall incidence of Mollicute infection or HS in this cohort.

Preclinical, Journal:  Generation of a doxycycline-inducible ETV2 expression cell line using PiggyBac transposase system. (Pubmed Central) -  Dec 5, 2022
The Dox-ETV2-H9 cells maintain normal morphology, karyotype as well as high expression of pluripotent markers. In comparison with previous studies that employing costly ETV2 modRNA to improve differentiation from ESCs into endothelial cells (ECs), the tet-on system in Dox-ETV2-H9 cells could precisely manipulate ETV2 expression, which enables ECs differentiation process more efficient and controllable upon addition of doxycycline.

Preclinical, Journal:  Highly efficient reprogrammable mouse lines with integrated reporters to track the route to pluripotency. (Pubmed Central) -  Dec 2, 2022
These systems were derived by exposing primary reprogramming cells containing doxycycline-inducible Yamanaka factor expression to a transient interruption in transgene expression, resulting in selection for a subset of clones with robust transgene response...Notably, somatic cells derived from various fetal and adult tissues from these 2° mouse lines gave rise to highly efficient and rapid reprogramming, with transgene-independent iPSC colonies emerging as early as 1 wk after induction. These mouse lines serve as a powerful tool to explore sources of variability in reprogramming and the mechanistic underpinnings of efficient reprogramming systems.

Preclinical, Journal:  Novel allosteric modulator SRI-32743 reverses HIV-1 Tat-induced increase in dopamine release in the caudate putamen of inducible Tat transgenic mice. (Pubmed Central) -  Dec 2, 2022
We found that 7- or 14-day doxycycline (Dox)-induced Tat expression in iTat-tg mice resulted in a two-fold increase in phasic but not tonic stimulated baseline dopamine release relative to saline control mice...Understanding the mechanisms underlying HIV-1 Tat protein-induced alteration of extracellular dopamine levels will provide insights into the development of molecules that can attenuate Tat interaction with targets in the dopaminergic system. Here, we determined whether Tat alters dopamine release and how the novel DAT allosteric modulator, SRI-32743, impacts dopamine neurotransmission to attenuate Tat-induced effects on extracellular dopamine dynamics.

Review, Journal:  Dermatological manifestations of tick-borne viral infections found in the United States. (Pubmed Central) -  Nov 30, 2022
Doxycycline is often empirically prescribed by clinicians to treat symptomatic patients following tick bites due to suspicions of bacterial TBDs such as Rocky Mountain spotted fever, anaplasmosis, and ehrlichiosis...KEY POINTS: Viral tick-borne diseases have increased in prevalence over the last decade and often have similar clinical manifestations to other tick-borne diseases, including bacterial infections. Here, we review the dermatologic manifestations of Heartland Virus (HRTV), Bourbon Virus (BRBV), Powassan Virus (POWV), Deer Tick Virus (DTV) and Colorado Tick Fever Virus (CTFV) that are important for clinicians.
