Journal:  A portable test strip fabricated of luminescent lanthanide-functionalized metal-organic frameworks for rapid and visual detection of tetracycline antibiotics. (Pubmed Central) -  Aug 29, 2023
Upon exposure to TCs, significant luminescence quenching of ATUC excited at 255 nm was observed due to the inner filter effect (IFE) and photo-induced electron transfer (PET), and the established strategy has a detection limit (LOD) of 11.0, 20.1, 9.1, and 22.5 nM for TC, CTC, OTC, and DOX, respectively. More importantly, given its portability and conspicuous luminescence color gradation variation, a portable test strip based on ATUC was manufactured and the results could be distinguished immediately by the naked eye and smartphone analysis, allowing for on-site rapid quantitative assay of TCs, not only in the laboratory but also in a point-of-care setting.

Journal:  Cytomegalovirus cyclin-dependent kinase ortholog vCDK/pUL97 undergoes regulatory interaction with human cyclin H and CDK7 to codetermine viral replication efficiency. (Pubmed Central) -  Aug 29, 2023
Here we provide a variety of novel results, as generated on a methodologically extended basis, and present a concept for the codetermination of viral replication efficiency through these kinase activities: (i) cyclin H expression, in various human cell types, is substantially upregulated by strains of HCMV including the clinically relevant HCMV Merlin; (ii) vCDK/pUL97 interacts with human cyclin H in both HCMV-infected and plasmid-transfected cell systems; (iii) a doxycycline-inducible shRNA-dependent knock-down (KD) of cyclin H significantly reduces pUL97 activity (qSox in vitro kinase assay); (iv) accordingly, pUL97 in vitro kinase activity is seen significantly increased upon addition of recombinant cyclin H; (v) as a point of specific importance, human CDK7 activity shows an increase by vCDK/pUL97-mediated trans-stimulation (whereas pUL97 is not stimulated by CDK7); (vi) phosphosite-specific antibodies indicate an upregulated CDK7 phosphorylation upon HCMV infection, as mediated through a pUL97-specific modulatory effect (i.e. shown by pUL97 inhibitor treatment or pUL97-deficient viral mutant); (vii) finally, an efficient KD of cyclin H in primary fibroblasts generally results in an impaired HCMV replication efficiency as measured on protein and genomic levels. These results show evidence for the codetermination of viral replication by vCDK/pUL97, cyclin H and CDK7, thus supporting the specific importance of cyclin H as a central regulatory factor, and suggesting novel targeting options for antiviral drugs.

Journal:  Context-dependent effects of CCN2 on ?-cell mass expansion and indicators of cell stress in the setting of acute and chronic stress. (Pubmed Central) -  Aug 28, 2023
CCN2 induction in isolated islets treated with thapsigargin ex vivo resulted in upregulation of the gene encoding the Nrf2 transcription factor, a master regulator of antioxidant genes, suggesting that CCN2 further activates this pathway in the presence of cell stress. These studies indicate that the potential of CCN2 to induce ?-cell mass expansion is context-dependent and that the presence of ?-cell stress does not ensure ?-cell proliferation in response to CCN2.

Journal, Combination therapy:  Efficacy of supramolecular salicylic acid in combination with doxycycline in the treatment of acne. (Pubmed Central) -  Aug 21, 2023
The overall incidence of adverse reactions was significantly lower in the experimental group versus control group (5.71% vs. 17.14%, P<0.05). Supramolecular salicylic acid in combination with doxycycline in the treatment of patients with acne is an optimal option, as it could better promote acne regression, reduce the level of depression and reduce the patient's self-perceived burden.

Journal:  Imaging telomerase reverse transcriptase expression in oligodendrogliomas using hyperpolarized ?-[1-C]-gluconolactone. (Pubmed Central) -  Aug 21, 2023
Doxycycline-inducible TERT silencing abrogated 6-PG production from hyperpolarized ?-[1-C]-gluconolactone in oligodendroglioma cells, consistent with the loss of G6PD activity...These results indicate that hyperpolarized ?-[1-C]-gluconolactone reports on TERT expression and early response to therapy in oligodendrogliomas. Our studies identify a novel agent for imaging tumor proliferation and treatment response in oligodendroglioma patients.
