Preclinical, Journal: Evidence of Functional Ryanodine Receptors in Rat Mesenteric Collecting Lymphatic Vessels. (Pubmed Central) - Mar 24, 2020 Responses to SP were characterized in lymphatic vessels in the absence or presence of pretreatment with nifedipine to block L-type Ca channels, caffeine to block normal release and uptake of Ca from the sarcoplasmic reticulum, ryanodine to block all RyR isoforms, or dantrolene to more selectively block RyR1 and RyR3...RyR2 and RyR3 protein are found predominantly in the collecting lymphatic smooth muscle layer. Collectively, the data suggest that SP-induced tonic contraction requires both extracellular Ca plus Ca release from internal stores, and that RyRs play a role in the normal contractions and responsiveness to SP of rat mesenteric collecting lymphatics.

Emergence from Vegetative State Following Intrathecal Baclofen Pump Placement () - Feb 11, 2020 - Abstract #ISPRM2020ISPRM_888; Additionally, three weeks after placement, the patient emerged from a vegetative state. Placement of an intrathecal baclofen pump allows the patient to be weaned off of sedating medications and reduce autonomic instability promoting emergence.Conclusions : Implantation of an intrathecal baclofen pump reduces autonomic storming, dystonic posturing, and may help with emergence from a vegetative state.

As more information is accrued regarding anti-NMDA receptor encephalitis, it is important to make physicians caring for this population aware of this possible clinical presentation.

The patient received dantrolene sodium approximately 120 minutes after induction of anesthesia...Intraoperative vigilance in the monitoring of vital signs cannot be overemphasized. An increase in end-tidal carbon dioxide values, in addition to other clinical signs that cannot be easily attributed to other causes, should increase the clinical index of suspicion for a diagnosis.

(Pubmed Central) - Jan 4, 2020 Because inhibition of glutamatergic excitatory neurotransmission is a possible mechanism involved in neuroprotection, we investigated the effect of piperine on the 4-aminopyridine (4-AP)-evoked release of glutamate from rat hippocampal synaptosomes...In the presence of ω-conotoxin MVIIC, an N- and P/Q-type channel blocker, the piperine-mediated inhibition of 4-AP-evoked glutamate release was markedly reduced; however, dantrolene and CGP37157, which are intracellular Ca2+-release inhibitors, did not alter the piperine effect...The glutamate release-inhibiting effect of piperine was discovered to be prevented by the 5-HT1A receptor antagonist WAY100635 and the G protein βγ subunit inhibitor gallein; however, it was unaffected by the adenylate cyclase inhibitor SQ22536 or the protein kinase A inhibitor PKI622. These results suggest that piperine inhibits glutamate release from rat hippocampal nerve terminals by reducing Ca2+ influx through N- and P/Q-type Ca2+ channels and that the activation of presynaptic 5-HT1A receptors and the G protein βγ subunit is involved in this effect.

We investigated the role of four chemical analogs, commonly prescribed β-blockers (atenolol, metoprolol, propranolol, and sotalol), on Ca release and whole-cell currents in mammalian cancer cells (PC3 prostate cancer and MCF7 breast cancer cell lines)...The propranolol-induced Ca release was significantly inhibited by the chelation of extracellular calcium with ethylene glycol tetraacetic acid (EGTA) and by dantrolene, an inhibitor of the endoplasmic reticulum (ER) ryanodine receptor channels, and it was completely abolished by 2-aminoethoxydiphenyl borate, an inhibitor of the ER inositol-1,4,5-trisphosphate (IP) receptor channels...Finally, pre-incubation of cells with sotalol or timolol, non-selective blockers of β-adrenoceptors, also reduced the Ca release activated by propranolol. Our results show that different β-blockers have differential effects on whole-cell currents and free Ca release and that propranolol activates store-operated Ca release via a mechanism that involves calcium-induced calcium release and putative downstream transducers such as IP The differential action of class II antiarrhythmics on Ca release may have implications on the pharmacology of these drugs.

Furthermore, our simulation results suggest that these arrhythmogenic triggers can be potentially suppressed by inhibiting sarcoplasmic reticulum (SR) calcium leak and reversing remodeled I. More importantly, this study has clinically significant implications on the drugs used for maintaining SR calcium homeostasis, whereby drugs such as dantrolene may confer significant improvement for the treatment of AF patients with TBX5/PITX2 insufficiency.
