Clinical, Journal:  Prevalence of Escherichia Coli and Its Antimicrobial Susceptibility Profiles among Patients with UTI at Mulago Hospital, Kampala, Uganda. (Pubmed Central) -  Feb 23, 2020
Escherichia coli, the predominant uropathogen, showed significant multidrug resistance to antibiotics commonly prescribed for the management of UTIs. These findings should form a basis for preliminary decision making on the appropriate line of treatment for UTIs.Escherichia coli isolates were highly susceptible to cefotaxime/clavulanic acid (100%) and nitrofurantoin (70%) but showed high resistance to cefuroxime (100%), ceftazidime (100%), nalidixic acid (90%), and ciprofloxacin (90%).

Clinical, Journal:  Pseudotumour cerebri associated with mycoplasma pneumoniae infection and treatment with levofloxacin: a case report. (Pubmed Central) -  Feb 22, 2020
PTCS pathophysiology may be multifactorial and its specific features and severity may be a consequence of both constitutional and acquired factors interacting synergistically. It may be useful for paediatricians to know that some antibiotics may have the potential to precipitate PTCS in patients who already have an increased CSF pressure due to a transitory imbalanced CSF circulation caused by infections such as Mycoplasma pneumoniae, with headache being the first and most sensitive, but also the least specific, symptom.

Journal:  A slurry microcosm study on the interaction between antibiotics and soil bacterial community. (Pubmed Central) -  Feb 15, 2020
Beta-lactams (ampicillin and ceftriaxone) experienced fast biodegradation...Nalidixic acid was stable throughout the experimental period (164 h)...According to the PICRUSt prediction of metagenome, influence of antibiotics on overall metabolic function of the bacterial community is rather limited. This study has provided valuable information, from a phylogenetic viewpoint, about the influence of high concentration of antibiotics on soil bacterial community.

Review, Journal:  Drug-Induced Photosensitivity-An Update: Culprit Drugs, Prevention and Management. (Pubmed Central) -  Feb 14, 2020
We review the medical literature regarding the evidence for the culpability of each drug, including the results of phototesting, photopatch testing and rechallenge testing. Amiodarone, chlorpromazine, doxycycline, hydrochlorothiazide, nalidixic acid, naproxen, piroxicam, tetracycline, thioridazine, vemurafenib and voriconazole are among the most consistently implicated and warrant the most precaution by both the physician and patient.

Journal:  Tolerance to benzalkonium chloride and antimicrobial activity of Butia odorata Barb. Rodr. extract in Salmonella spp. isolates from food and food environments. (Pubmed Central) -  Feb 7, 2020
Resistance to ampicillin, streptomycin, nalidixic acid, sulfonamide, trimethoprim/sulfamethoxazole, trimethoprim, tetracycline, and chloramphenicol was observed...All isolates carried at least one virulence gene and produced biofilms on stainless steel surfaces at 10 and 22 °C. On the other hand, the B. odorata extract showed activity against Salmonella spp., and it has the potential to be used as a natural antimicrobial to control this important foodborne pathogen, despite its virulence potential and antimicrobial resistance profile.

Preclinical, Journal:  Characterization of Plasmid-Mediated Quinolone Resistance in Gram-Negative Bacterial Strains from Animals and Humans in China. (Pubmed Central) -  Feb 7, 2020
High rates of resistance to tetracycline (93.4%), nalidixic acid (81.5%), and norfloxacin (65.8%) were observed...In contrast, the coexistence of qnrD and bla was more prominent (p < 0.001) in P. mirabilis than in the other two species. Particularly, oqxAB and mcr-1 had an obvious preference for E. coli than K. pneumonia and P. mirabilis (p < 0.001), which had not been reported in previous studies on the prevalence of PMQRs.

Journal:  DksA-RNA polymerase interactions support new origin formation and DNA repair in Escherichia coli. (Pubmed Central) -  Jan 31, 2020
The requirement for DksA in repair of nalidixic acid (Nal)-induced DSBs or for the formation of new origins is not suppressed by a greA deletion mutation, indicating an active role of DksA rather than competition with GreA for insertion into the RNAP secondary channel...The roles of DksA in DSB repair and cSDR differ; certain DksA and RNAP mutants are able to support the first process, but not the latter. We suggest that new origin formation and DNA repair of protein adducts with DSBs may both involve removal of RNAP without destruction of the RNA:DNA hybrid.

Clinical, Journal:  Antibiotic Resistance of Campylobacter spp. in a Pediatric Cohort Study. (Pubmed Central) -  Jan 29, 2020
Personal macrolide exposure did not affect the risk of macrolide resistant Campylobacter Amoxicillin and clavulanic acid (94.0%) was one of the antibiotics with the highest rates of susceptibility.ConclusionThere is a high incidence of quinolone and macrolide resistant Campylobacter infections in infants under 24 months of age. Given the lack of association between personal exposure to macrolides and a subsequent Campylobacter infection resistant to macrolides, there is a need to evaluate the source of MDR Campylobacter This study provides compelling evidence to propose amoxicillin/clavulanic acid as a treatment for Campylobacteriosis.

Journal:  Surveillance of antibiotic resistance in Escherichia coli isolated from wild cranes on the Izumi plain in Kagoshima prefecture, Japan. (Pubmed Central) -  Jan 28, 2020
Since the previous surveillance in 2007, isolation rates of antibiotic-resistant E. coli recovered from wild cranes have remained at significantly low levels compared with those in Japanese livestock. Our results indicate that surveillance of antibiotic-resistant E. coli from wild cranes wintering in the Izumi plain could be a useful strategy to indicate natural environmental pollution by antibiotic-resistant bacteria in the environment.

Clinical, Journal:  Longitudinal Assessment of Antibiotic Resistance in Fecal Escherichia coli in Tanzanian Children. (Pubmed Central) -  Jan 27, 2020
Resistance to amoxicillin/clavulanate (361/836; 43.2%), ampicillin/sulbactam (178/819; 21.7%), nalidixic acid (131/831; 15.8%), and azithromycin (115/837; 13.7%) was also seen...In this setting, the carriage of antibiotic-resistant E. coli was common. Other than recent antibiotic exposure and high income, individual risk factors for the acquisition and carriage of resistance could not be identified, suggesting that population-level interventions are needed.

Journal:  Antibiotic-Resistant Escherichia coli in Drinking Water Samples from Rural Andean Households in Cajamarca, Peru. (Pubmed Central) -  Jan 25, 2020
The E. coli antibiotic resistance profile showed highest resistance against tetracycline (37.6%), ampicillin (34.2%), sulfamethoxazole-trimethoprim (21.4%), and nalidixic acid (13%)...The high prevalence of fecal contamination in drinking water highlights the importance of household water treatment methods. Likewise, the high levels of antibiotic resistance found indicate a need for further research to identify the origins of potential environmental contamination, misuse, or inadequate disposal of antibiotics.

Clinical, Journal:  Screening of tropical estuarine water in south-west coast of India reveals emergence of ARGs-harboring hypervirulent Escherichia coli of global significance. (Pubmed Central) -  Jan 17, 2020
The highest prevalence of antibiotic resistance was observed for ampicillin, followed by tetracycline, and nalidixic acid...ERIC-PCR (enterobacterial repetitive intergenic consensus sequence polymerase chain reaction) analysis revealed a high degree of genetic diversity among the ARGs-harboring E. coli isolates. The results demonstrate a high prevalence of ARGs-harboring E. coli in estuarine water and confirm the need for a better wastewater treatment facility and proper control measures to reduce the discharge of sewage and wastewater into the aquatic environments.

Journal:  Effect of crude oil-induced water repellency on transport of Escherichia coli and bromide through repacked and physically-weathered soil columns. (Pubmed Central) -  Jan 16, 2020
The present study aims to investigate the effect of crude oil-mediated water repellency on the movement of nalidixic acid-resistant Escherichia coli strain (E. coli NAR) and bromide (Br) as an inert tracer in two soil types...This finding was attributed to low specific surface area, continuity of the pores and water repellency-mediated preferential pathways in the weathered SL soil columns. Our findings implied that shallow groundwater resources could be very sensitive to microbial contamination particularly in the oil-mediated water-repellent soils.

Journal:  Novel nalidixic acid derivatives targeting topoisomerase II enzyme; Design, synthesis, anticancer activity and effect on cell cycle profile. (Pubmed Central) -  Jan 10, 2020
Cell cycle analysis indicated that compound 6a induced cell cycle arrest at G2-M phase leading to inhibition of cell proliferation and apoptosis. Molecular modeling demonstrated that the potent topoisomerase inhibitory activity of 6a was due to the interaction with the topoisomerase II enzyme through coordinate bonding with the magnesium ion Mg, hydrogen bonding with Asp 545 and arene cation interaction with His 759.

Journal:  Nalidixic acid surrogate test for susceptibility to ciprofloxacin in Salmonella. Revisiting the question. (Pubmed Central) -  Dec 28, 2019
However, there were differences among the serogroups in terms of sensitivity (P<0.001) and positive predictive values (P=0.013). NA is a reliable marker for serogroup D, but not for serogroups B or C. According to these findings, NA susceptibility measured with the MicroScan system can be used as a marker of ciprofloxacin resistance in some serogroups in our setting.

Journal:  Assessment of the genotoxicity of antibiotics and chromium in primary sludge and compost using Vicia faba micronucleus test. (Pubmed Central) -  Dec 27, 2019
Although the final compost product showed a significant decrease of the genotoxicity, almost 50% of the micronucleus frequency still remained, which could be explained by the persistence of several recalcitrant compounds such as chromium and some antibiotics. Overall, the presence of antibiotics and chromium showed that some specific combination of contaminants represent an ecological risk for soil health and ecosystems even at environmentally negligible concentrations.

Journal:  Phenotypic screening for quinolone resistance in Escherichia coli. (Pubmed Central) -  Dec 23, 2019
We suggest that in situations where low-level quinolone resistance might be of importance, such as when screening for quinolone resistance in fecal samples pre-TRUS-B, a pefloxacin (S ≥ 24 mm) or nalidixic acid (S ≥ 19 mm) disk, or a combination of the two, should be used. In a setting where plasmid-mediated resistance is prevalent, pefloxacin might perform better than nalidixic acid.

Journal:  Growth of Salmonella on Inoculated Inhull Pistachios during Postharvest Handling. (Pubmed Central) -  Dec 21, 2019
Similar growth curves were observed at the low and high inoculum levels; throughout the 30 h of incubation, Salmonella populations were consistently ∼1 to 2 log CFU/g lower on pistachios inoculated at the low inoculum level. Managing the time between harvesting and hulling will reduce the potential for growth of Salmonella on pistachios during postharvest handling.

Journal:  Transduction as a Potential Dissemination Mechanism of a Clonal qnrB19-Carrying Plasmid Isolated From Salmonella of Multiple Serotypes and Isolation Sources. (Pubmed Central) -  Dec 5, 2019
Altogether our results demonstrate that plasmid-mediated quinolone resistance (PMQR) is widespread in Salmonella enterica of different serotypes isolated from human clinical samples, wild and domestic animals, and food in Chile and suggest that transduction could be a plausible mechanism for its dissemination. The occurrence of these antimicrobial resistance elements in Salmonella in a widespread area is of public health and food safety concern, and it indicates the need for increased surveillance for the presence of these plasmids in Salmonella strains and to assess their actual impact in the rise and spread of quinolone resistance.
