    Preclinical, Journal:  Atypical Antipsychotic Drug Ziprasidone Protects against Rotenone-Induced Neurotoxicity: An In Vitro Study. (Pubmed Central) -  Apr 10, 2021   
    Interestingly, the use of serotonin 5-HT receptor antagonist 1-(2-methoxyphenyl)-4 (4-(2-phtalimido) butyl) piperazine (NAN-190) completely blocked the protective effect of ZPD against ROT-induced cell death. Our results demonstrate the neuroprotective effect of ZPD against ROT-induced neurotoxicity and suggest that ZPD may be a potential candidate for the prevention of mitochondrial dysfunction and oxidative stress in schizophrenia.

In addition, pharmacological treatments are discussed as well.

    Clinical, Journal:  An Emulation of Randomized Trials of Administrating Antipsychotics in PTSD Patients for Outcomes of Suicide-Related Events. (Pubmed Central) -  Apr 7, 2021   
    In addition, olanzapine was associated with higher SREs than quetiapine and risperidone, and ziprasidone was associated with higher SREs than risperidone. The results of this study suggest that certain antipsychotics may put individuals within the PTSD population at an increased risk of SREs, and that careful consideration may need to be taken when prescribed.

    The small-to-modest sample size, the lack of control groups and the open-label and short-term nature of most of the reviewed studies hinder definitive conclusions regarding either the short-term and long-term efficacy of antipsychotic and anticonvulsant medications or their potential long-term side effects. Some atypical antipsychotic and anticonvulsant medications may have a role in the treatment of some PD patients, mostly when more conventional approaches have not been successful, but the quality of supporting evidence is limited.

    In subproject 1, 1,800 first-episode patients with schizophrenia are randomized into six different antipsychotic monotherapy groups (olanzapine, risperidone, aripiprazole, ziprasidone, amisulpride, and haloperidol) for an 8-week treatment...In subproject 2, apart from verifying the prediction model established in subproject 1 based on an independent cohort of 1,800 first-episode patients with schizophrenia, we recruit patients from a verification cohort who did not get an effective response after an 8-week antipsychotic treatment into a randomized double-blind controlled trial with minocycline (200 mg per day) and sulforaphane (3 tables per day) to explore add-on treatment for patients with schizophrenia...In subproject 3, we tend to carry out one trial to construct an intervention strategy for metabolic syndrome induced by antipsychotic treatment and another one to build a prevention strategy for patients at a high risk of metabolic syndrome, which combines metformin and lifestyle intervention...Trial Registration: This trial has been registered on Clinicalrials.gov (NCT03451734).     Biomarker, Clinical, Journal:  Comparative Study of the Effects of Atypical Antipsychotic Drugs on Plasma and Urine Biomarkers of Oxidative Stress in Schizophrenic Patients. (Pubmed Central) -  Feb 26, 2021   
    A 4-week treatment with ADs caused the improvement of psychopathology symptoms estimated by Positive and Negative Syndrome Scale (PANSS) that was accompanied by decreased lipid peroxidation (F2-isoprostanes, TBARS; p=2.9x10, p=7.6x10, respectively) and an increase in total antioxidative capacity (FRAP) (p=5.16x10). Atypical antipsychotics especially clozapine, olanzapine and quetiapine demonstrate the effective outcome of antipsychotic treatment, beneficial antioxidative action by reducing lipid peroxidation and increased total plasma antioxidant activity.

Finally, a variety of useful strategies (such as the addition of sildenafil) are also described where the previous ones cannot be applied, although they may not yield as optimal results.

    The electronic medical record system will identify men and women 18 years of age and older over a 12-month period who were prescribed of one the following QTc prolonging medications: amitriptyline, citalopram, chlorpromazine, clozapine, dofetilide, doxepin, flecainide, haloperidone, lithium, nortriptyline, or ziprasidone...Data will be reviewed and used to calculate the percentage of patients who received orders to obtain an EKG while on prescribed QTc prolonging medications, as well as the change in msec on the EKG as a result of these medications. If accepted, the final results of this project will be presented at the 2020 ASHP Midyear Clinical Meeting.

    In the association analysis of LDL change, rs142100580 in KCNQ1 (p=4.1010-8) is significantly associated with the LDL change after treatment with olanzapine, and rs75684025 in ZFPM2 (p=3.8810-8) is associated with the treatment with ziprasidone. In the ROC analyses of all the developed model, the prediction accuracy of the genetic risk score was area under the curve 70%.
