Review, Journal: Optimal Management of Status Epilepticus in Children in the Emergency Setting: A Review of Recent Advances. (Pubmed Central) - Sep 29, 2022 Up until 2019, phenobarbital and phenytoin (or fosphenytoin) were the preferred second-line treatments but with no good supporting RCT evidence...Lacosamide is an alternative second-line treatment but with no supporting RCT evidence...Pragmatically, levetiracetam or sodium valproate are preferred to phenytoin (or fosphenytoin) because of their ease of administration and lack of serious adverse side-effects, including potentially fatal cardiac arrhythmias. Sodium valproate must be used with caution in children aged three and under because of the rare risk of hepatotoxicity and particularly if there is an underlying mitochondrial disorder.

Journal: Lacosamide decreases neonatal seizures without increasing apoptosis. (Pubmed Central) - Sep 29, 2022 Lacosamide reduces neocortical seizure-like activity in neonatal mice in vitro and in vivo without an acute increase in apoptosis. Our results support the use of lacosamide to treat neonatal seizures, with the advantage of not increasing apoptosis acutely.

Retrospective data, Journal: Status epilepticus management and mortality risk factors: a retrospective study. (Pubmed Central) - Sep 10, 2022 Patients with these two factors should be carefully checked for skin rashes. To improve SE management and reduce mortality rates, training activities targeting emergency department physicians should be implemented, together with elective intensive care admission for patients with multiple mortality risk factors (eg, absence of history of epilepsy, acute stroke, or cardiovascular complications).

(Pubmed Central) - Sep 7, 2022 P2 For these reasons, IMI2-PainCare-BioPain-RCT2 hypothesizes that spinal and brainstem neurophysiological measures can serve as biomarkers of target engagement of analgesic drugs for future Phase 1 clinical trials. Phase 2 and 3 clinical trials could also benefit from these tools for patient stratification.

Efficiency of Third-Generation Antiseizure Medications in Spain for Drug-Resistant Epilepsy: A Cost-per-Number Needed to Treat Analysis (In-person; Virtual) - Sep 1, 2022 - Abstract #ISPOREU2022ISPOR_EU_1623; The findings show a clear link between the efficacy of third-generation ASMs available in Spain for the treatment of FOS in patients with DRE and their efficiency, facilitating determination of drug’s value contribution and decision-making in Spain. Cenobamate presents the lowest cost per NNT relative to seizure freedom, proving to be the most efficient option compared to third-generation alternatives.
- |||||||||| Review, Journal: Neurosyphilis presenting as autoimmune limbic encephalitis: A case report and literature review. (Pubmed Central) - Aug 28, 2022
The present case was diagnosed with neurosyphilis presenting as ALE, but meanwhile, in most case, neurosyphilis presenting as LE developed at a slower progressive rate, and it had a broader or restricted involvement on brain MRI than ALE based on the literature review. Therefore, an appropriate differential diagnosis of LE can be obtained by identifying clinical differences between the 2 conditions.

Therefore, an appropriate differential diagnosis of LE can be obtained by identifying clinical differences between the 2 conditions. The anticonvulsants are probably safer, but the risk of developing PCOS associated with the antiepileptics cannot be established, since there are insufficient studies.

Journal: Reponse of second-line treatment in focal status epilepticus: A tertiary hospital experience. (Pubmed Central) - Aug 24, 2022 Our findings further support the notion that early termination of SE with a first or second ASM confers a better functional outcome. The large difference in response between the first ASM and consecutive ones suggests that the sum of different ASMs might be the key to resolving focal SE.

A systematic method for the forced degradation studies on anti-epileptic drugs such as "Topiramate, Vigabatrin, Lacosamide, Tiagabine, Levetiracetam and Zonisamide" is discussed. This review helps researchers to get an idea about stability-indicating methods of development and validation for newer antiepileptic drugs and the characteristics of drug products that degrade under specific degradation conditions.

Although little information is available on the adverse effects or benefits on movement disorders of newer antiseizure drugs (such as brivaracetam, cenobamate, eslicarbazepine, lacosamide, and rufinamide), the evidence collected in this review should guide the choice of antiseizure drugs in patients with concomitant epilepsy and movement disorders. Finally, these notions can lead to a better understanding of the mechanisms involved in the pathophysiology and treatments of movement disorders.

Preclinical, Journal: An Investigation of the Effects of Chronic Zonisamide, Sultiam, Lacosamide, Clobazam, and Rufinamide Antiseizure Drugs on Foliculogenesis in Ovarian Tissue in Prepubertal Non-Epileptic Rats. (Pubmed Central) - Aug 5, 2022 HSCORE, immunohistochemical EGF, IGF-1 and GDF-9 staining, a very strong immunoreaction was observed in the ovarian multilaminar primary follicle granulosa cells and oocytes in the control group (p<0.001), and an immunoreaction ranging from weak to medium was observed in the antiseizure drug groups. Long-term anti-seizure drug therapy with zonisamide, sultiam, lacosamide, clobazam, and rufinamide from prepubertal to adulthood causes apoptosis and disruption of folliculogenesis in the ovarian follicles of nonepileptic rats.

The ASM most robustly associated with increased appetite and/or weight gain is valproic acid; however, both pregabalin and perampanel may also lead to modest weight gain or increased appetite in children. Certain ASMs may impact both appetite and weight, which may lead to increased morbidity of the underlying disease and impaired adherence to the treatment regimen.
