We measured GI motility with and without loperamide, an antidiarrheal medication, by intestinal transit using an orally administered dye, a C-octanoic acid breath test, and ultrasonography...In accordance with these results, quantifiable locomotion activity was also significantly decreased. In conclusion, ultrasonography is an effective in vivo method to quantify small intestinal motility in mice.

In an analysis of reports on eluxadoline submitted to the Federal Adverse Event Reporting System, we confirmed a previously reported risk of pancreatitis associated with eluxadoline. The need for hospitalization in at least half of these instances and a recent report of 2 fatalities should prompt reassessments of the agent's risk-benefit ratio.

Taken together, the results of the present study provide the first strong evidence that SpiA improves chronic constipation through muscarinic cholinergic regulation of the enteric nerve in a Lop-induced constipation ICR mice model.

Inhibition of autophagy induction either chemically (by 3-methyladenine) or genetically (by Atg5 silencing) did not reverse the synergistic apoptotic effect, indicating that the apoptotic cell death was not mediated by autophagy induction. Conclusion Our results indicated that loperamide could be repurposed to combine with gefitinib to produce synergistic anti-cancer effect in KRAS mutant NSCLC cell lines by further inducing synergistic apoptosis, which is independent of autophagy induction.

As a result, CYP3A4.6, .17, .20 and .30 showed extremely low activity or no activity and the rest of CYP3A4 variants presented varying degrees of decrements in catalytical activities when compared with CYP3A4.1. As the first study to identify the properties of these CYP3A4 variants toward loperamide metabolism, our investigation may establish the genotype-phenotype relationship for loperamide, predict an individual's capability in response to loperamide, and provide some guidance of clinical medication and treatment for loperamide.

GIT ratio and loperamide-induced serum metabolic parameters, such as gastrin (GAS), motilin (MTL), and somatostatin (SS) significantly changed after supplementation with taurine in loperamide-induced constipated rats. We suggest that taurine had a potent effect against loperamide-induced constipation in part by increasing gastrointestinal motility.

At the optimized μ-EME conditions (250 V, 15 min, 300 rpm, acidic donor) and the optimized ratio of the sample to the FLM volume (500:14 μL), extraction recoveries between 49 and 100% and enrichment factors up to 35.7 were achieved. Quantitative analyses of the basic drugs in the resulting FLMs (diluted with methanol) were performed by capillary electrophoresis with ultraviolet detection and demonstrated excellent repeatability (RSD ≤ 4.9%) and linearity (r ≥ 0.9997), and low limits of detection (5-28 ng/mL) of the method.

However,rhubarb showed obvious hepatic and colon toxicities at the dosage of 2 and 8 times of the high limit set forth in the Chinese Pharmacopoeia,and the combination with licorice could significantly reduce its toxicity. It shows that licorice has a " mediating" effect on rhubarb by alleviating the purgation property and reducing the toxicity.
