Journal: Impact of drug incorporation into micelle on reduced griseofulvin and meloxicam permeation across a hollow fiber membrane. (Pubmed Central) - Dec 22, 2024 Permeation from both free griseofulvin and micelle-bound griseofulvin successfully accommodated observed flux, where micelle permeability was about 5-fold lower than free drug permeability for HFM with 10KDa MWCO. Permeation with enhancement from micelle shuttling was not the preferred explanation, although the model accommodated flux data and provided aqueous boundary layer thicknesses similar to other setups.

Journal: Ionic resorcinarenes as drug solubilization agents in water. (Pubmed Central) - Oct 29, 2024 This article reports on the aqueous-solubility enhancing effect of an octa-sulfonated resorcinarene and a C-hydroxybenzyl ammonium resorcinarene chloride on three hydrophobic drugs: isoniazid, caffeine, and griseofulvin...Cytotoxicity studies suggest that resorcinarenes alone, or complexed with any of the drug compounds, do not exert cytotoxicity in mammalian cells HEK-293 up to 200 ?M. We herein propose a set of hydrophilic resorcinarene macrocycles as potential drug solubilizers.

Journal: In Situ Raman Spectroscopy as a Valuable Tool for Monitoring Crystallization Kinetics in Molecular Glasses. (Pubmed Central) - Oct 16, 2024 However, from the quantitative point of view, the reproducibility of the IRM measurements was found to be potentially significantly hindered due to inaccurate temperature recording and calibration, variability in the Raman spectra corresponding to the fully amorphous and crystalline phases, and an overly limited number of spectra possible to collect during acceptable experimental timescales because of the applied heating rates. Since theoretical simulations showed that, from the kinetics point of view, the constant density of collected data points per kinetic effect results in the smallest distortions, only the employment of the fast Raman mapping functions could advance the performance of IRM above that of calorimetric measurements.

Amphotericin B, itraconazole, posaconazole, voriconazole, terbinafine, amorolfine, and griseofulvin MICs were overall high (GM 12.79-29.49 ?g/mL)...Antifungal susceptibility of the three different A. egyptiacum genotypes was homogeneous. Overall, the high MICs recorded for all systemically administered drugs, and for some topical antifungals against the tested A. egyptiacum and other acremonium-like clinical isolates, justify the routine susceptibility testing of clinical isolates.

This study highlights that general expectations based on in vitro dissolution (i.e. that smaller particles dissolve faster than larger particles when surrounded by liquid) do not always translate to in vivo and demonstrates the importance of understanding the physicochemical properties of the drug, the characteristics of the formulations and the microphysiology at the delivery site.

Apart from requirement of prolonged treatment durations, relapse post successful treatment is a distressing and yet unexplained consequence of this "species-shift". Use of third generation azoles and combinations of systemic antifungals is unwarranted as both have not demonstrated clear superiority over ITZ given alone, and the former is an important class of drugs for invasive mycoses.

These results demonstrate the power of automated 3D ED for accurate phase identification and direct structure determination of complex, beam-sensitive crystallisation products, which is significant for drug development where solid form screening is crucial for the overall efficacy of the drug product.
