Journal: Hyperuricemia drives intestinal barrier dysfunction by regulating gut microbiota. (Pubmed Central) - Sep 3, 2024 Allopurinol was used to decrease uric acid level, and antibiotics were administered to mice to deplete gut microbiota...However, removing gut microbiota aggravated lipid metabolic dysfunction. Gut microbiota is a direct and specific cause for intestinal barrier dysfunction.

This opinion article analyses the causes of HUA, offers dietary and exercise guidance with the aim of furnishing a point of reference for individuals with HUA or fitness enthusiasts.

Journal: Identification of a novel xanthine oxidoreductase inhibitor for hyperuricemia treatment with high efficacy and safety profile. (Pubmed Central) - Aug 24, 2024 It also showed favorable dose-dependent uric acid-lowering effects in two different XOR substrate-induced hyperuricemic mouse models, which was significantly superior than the current first-line drug, allopurinol...Notably, there was no obvious inhibition of CC15009 against cardiac ion channels, including hERG, Nav1.5, Cav1.2 at the concentration of 30??M, indicating its lower cardiovascular risk. Taken together, our results supported CC15009 as a candidate of high efficacy and safety profile to treat hyperuricemia through direct XOR inhibition.

(Pubmed Central) - Aug 23, 2024 Xanthine, typically enzymatically converted to uric acid, can accumulate when xanthine oxidases, such as allopurinol, are administered during TLS management. Whether measurement of xanthine is clinically useful to optimize the use of allopurinol or rasburicase remains to be determined.

Home medications included allopurinol, gabapentin, olmesartan, and amlodipine...As o ur patient also improved after valsartan was substituted for olmesartan, OIE rose to the top of our differential diagnosis...Other characteristic findings include increase in intra-epithelial lymphocytes, noted on her outpatient endoscopy but not on repeat inpatient endoscopy (likely due to initiation of budesonide and mesalamine, lowering lymphocytic infiltrate biopsy yield). Clinical suspicion and a broad differential are key to diagnosing OIE; it often presents a diagnostic dilemma and improves if olmesartan is suspended.

- |||||||||| Retrospective data, Journal: The Effectiveness of Measuring Thiopurine Metabolites in the Treatment of Autoimmune Hepatitis Patients. (Pubmed Central) - Aug 8, 2024
The thiopurine drugs-azathioprine and mercaptopurine-are purine antimetabolites used for the treatment of autoimmune hepatitis...In 11 cases, levels of 6-thioguanine nucleotide were low; the dose was not changed in 3 of these, and the dose was increased in the remaining 8...In the remaining 3, the dose was decreased, and allopurinol was added...We showed that, in cases of suboptimal response to thiopurine treatment, measuring thiopurine metabolites had an important role in optimizing therapy. In most patients, changing the dose led to a significant improvement with no need to switch to secondline therapies.
