MLE treatment in adulthood reversed the deficit in prepulse inhibition (PPI) but it failed to attenuate the sensitivity to amphetamine and the deficit in novel object recognition (NOR) induced...In addition, MLE administration in adolescence reduced iNOS mediated inflammatory activation and it promoted the expression of biomarkers of compensatory antioxidant activity in the prefrontal cortex and hippocampus, as witnessed through the reduction of Keap1 and the accumulation of NRF2 and HO1. Together, these findings suggest that MLE might be an alternative therapeutic or preventive add-on strategy to improve the clinical expression of schizophrenia in adulthood, while also modifying the time course of this disease at earlier stages in populations at high-risk.

Olanzapine has a better safety profile in terms of SD, experience with quetiapine is insufficient.

The variables that showed a statistically significant association with the current use of psychotropic drugs were the child's age (OR = 1.25, 95% CI: 1.12, 1.40, p < 0.001), presence of comorbidities (OR = 7.75, 95% CI: 3.48, 17.24, p < 0.001), communication difficulties (OR = 1.79, 95% CI: 1.09, 2.95, p < 0.021), and anxiety symptoms (OR = 1.70, 95% CI: 1.00, 2.87, p < 0.049). Similarly, the child's age (OR = 1.23, 95% CI: 1.08, 1.40, p < 0.002) and presence of comorbidities (OR = 2.83, 95% CI: 1.16, 6.29, p < 0.022) showed statistically significant associations with previous use.

In the hERα-HeLa-9903 cells, estrogen receptor α agonistic activities were shown for fluoxetine, paroxetine, and sertraline (at 10 μM and 25 μM), and in the GH3.TRE-Luc cells, the same three CNS drugs showed antithyroid activities (ICs, 11.6, 11.9, 2.7 μM, respectively)...Little to no effects were seen toward any of these nuclear receptors for paliperidone and risperidone...These data elucidate the potential for adverse endocrine effects for some of these CNS drugs, which should therefore contribute to informed choice when prescribing them. However, long-term exposure to therapeutic concentrations of CNS drugs that have activities on the endocrine system should be explored further also in vivo.

Overall, 78% (n = 61/78), including all 20 registrars surveyed, expressed a wish to participate in a training course for prescribing antipsychotics. Our national survey demonstrates that there is a need to improve both exposure to psychocutaneous conditions; confidence in the prescription of antipsychotic medications across all grades, including consultants,; that there is a clear demand from trainees for participation in such courses, which would satisfy this training need, in addition to ensuring that consultants of the future are well equipped to provide care for patients with psychocutaneous conditions.
