    Journal:  Development and Evaluation of PLGA Nanoparticle-Loaded Organogel for the Transdermal Delivery of Risperidone. (Pubmed Central) -  Nov 16, 2022
    Statistical analysis confirmed that there was a significant difference (p < 0.05) between the nanoparticle-loaded PLO gel and conventional drug formulations in terms of drug release and ex vivo permeation across rat skin (three-fold). The results confirm enhanced drug release and permeation through the skin at 72 h. Hence, the investigated formulation could be a better alternative to the conventional route for improving patient compliance.

    Review, Journal:  Antipsychotics in the Management of Disruptive Behavior Disorders in Children and Adolescents: An Update and Critical Review. (Pubmed Central) -  Nov 16, 2022
    Overall, the data from these trials suggests that of all available antipsychotics, risperidone appears to be effective in the short-term management of DBDs...These results are discussed in the light of global trends towards increasing off-label prescription of antipsychotic medication in children and adolescents and of recent literature on the neuropharmacology of aggression in this patient population. The need for rational, short-term use of these drugs is highlighted, as well as the importance of post-marketing surveillance for long-term or severe adverse events.

    Journal:  Later onset of Childhood Disintegrative Disorder (CDD): a case report. (Pubmed Central) -  Nov 15, 2022
    Following treatment with lithium carbonate combined with risperidone, the patient experienced a reduction in irritability and aggression. CDD is a rare condition; therefore, the data presented may be useful to investigate its characteristics of the onset, to improve the understanding of the aspects of differentiation from the Autism Spectrum Disorder and finally to propose the possibility of treatment.

    Journal, Adverse events:  Physicochemical Insights on Terahertz Wave Diminished Side Effects of Drugs from Slow Dissociation. (Pubmed Central) -  Nov 15, 2022
    We have proven that a 4.0 THz wave could reduce the affinity by 71% between the D2R and a risperidone ligand and meanwhile expand the exit via conformation modulation, which promises an accelerated dissociation of risperidone...This work elaborates the terahertz wave-modulated noncovalent interactions critical in cell signaling pathways. Most importantly, it demonstrates the feasibility of terahertz technologies intervening in receptor-ligand complex regulated diseases such as neurodegenerative disorders, metabolic diseases, etc.
  • ||||||||||  Clinical, Review, Journal:  Pharmacologic treatment of delirium symptoms: A systematic review. (Pubmed Central) -  Nov 15, 2022
    Pharmacologic agents can reduce delirium symptoms (e.g., agitation) in some hospitalized patients. Additional double-blinded, randomized, placebo-controlled clinical trials are critically needed to investigate the efficacy of pharmacologic agents for diverse hospitalized populations (e.g., post-surgical patients, patients at the end-of-life, or in intensive care units).
  • ||||||||||  Use of Valproic Acid to Treat Refractory Delirium in a Pediatric Patient (Anaheim Convention Center, Hall A, Board # 098) -  Nov 10, 2022 - Abstract #AAPNCE2022AAP_NCE_2254;
    Day-night cycling was encouraged and weaning of sedation was attempted, however she continued to have significant agitation and required maintenance of continuous infusions of dexmedetomidine, hydromorphone, and midazolam as well as scheduled risperidone, lorazepam, methadone, and clonidine. Our experience indicates that that VPA is a promising agent for patients with protracted delirium and may provide an alternative strategy for successfully managing these patients.

    Journal:  Neuroleptic malignant syndrome caused by olanzapine, risperidone and haloperidol (Pubmed Central) -  Nov 9, 2022
    This is a case report of a 27-year-old man who developed neuroleptic malignant syndrome after administration of olanzapine, risperidone and haloperidol. He was treated with benzodiazepine, dantrolene, amantadine and electroconvulsive therapy with good recovery.

    Journal:  Effects of icariin on alleviating schizophrenia-like symptoms by regulating the miR-144-3p/ATP1B2/mTOR signalling pathway. (Pubmed Central) -  Nov 8, 2022
    A developmental schizophrenia rat model employing 2-week repeated MK-801 administration was established...Lastly, icariin can regulate the expression of miR-144-3p and ATP1B2, and enhance the phosphorylation of PI3K, Akt, and mTOR. In conclusion, this study revealed that icariin may have therapeutic effects on schizophrenia-like disorders via regulating the miR-144-3p/ATP1B2/mTOR signalling, suggesting that icariin has potential to become an antipsychotic drug.

    Biomarker, Journal:  Frontal lobe fALFF measured from resting-state fMRI as a prognostic biomarker in first-episode psychosis. (Pubmed Central) -  Nov 5, 2022
    At baseline, patients who would later meet strict criteria for clinical response (defined as two consecutive ratings of much or very much improved on the CGI, as well as a rating of ≤3 on psychosis-related items of the BPRS-A) demonstrated significantly greater baseline fALFF in bilateral orbitofrontal cortex compared to non-responders. Thus, spontaneous activity in orbitofrontal cortex may serve as a prognostic biomarker of antipsychotic treatment.

    Preclinical, Journal:  Homotaurine ameriolates the core ASD symptomatology in VPA rats through GABAergic signalling: Role of GAD67. (Pubmed Central) -  Nov 2, 2022
    Male pups from VPA-exposed mothers were further divided into five groups (n = 6 in each group): disease control (DC, no-further treatment), standard treatment (risperidone (RES) 2.5 mg/kg; i.p, consecutively from PND 23-43), HT (10, 25 and 50 mg/kg; i.p, consecutively from PND 23-43)...Compared to the DC, HT treatment (50mg/kg) was able to ameliorate the aberrant core behavioral deficits, decreased oxidative stress, decreased pro-inflammatory and increased anti-inflammatory cytokine profile with preservation of the Purkinje cell density in the cerebellum, decreased pyknosis in the prefrontal cortex and normalised the expression of GAD67. Thus, HT can be a useful therapeutic agent in ASD and requires further clinical evaluation.
  • ||||||||||  Plasma Free Fatty Acids After Six Weeks of Antipsychotics Treatment of Schizophrenia () -  Oct 31, 2022 - Abstract #ACNP2022ACNP_1004;
    Quetiapine did not change FFA levels similar to haloperidol and abilify [4,5]. Notably, the mean decrease of PANSS total scores was lower in patients treated by quetiapine than in patients treated by olanzapine and risperidone, although we did not observe correlations between FFA and PANSS scores [5].

    Pharmacological Interventions for the Treatment of Tourette’s Syndrome in Youth: A Systematic Review and Network Meta-Analysis (Grand Saguaro) -  Oct 31, 2022 - Abstract #ACNP2022ACNP_892;
    Because there were no differences in efficacy between any pairs of antipsychotics based on the currently available data, the choice of antipsychotic should be guided mainly by their tolerability. Nevertheless, whether antipsychotics should be used as first-line pharmacological interventions in the treatments of TS is debatable given they are potentially toxic and may lead to somatic and serious adverse events, particularly in vulnerable populations such as children and adolescents.

    Journal:  Patterns of Antipsychotic Dispensation to Long-Term Care Residents. (Pubmed Central) -  Oct 30, 2022
    Healthcare providers should educate their patients about the medicines that may cause weight gain. With 90% of antipsychotic dispensations in Nova Scotia being to LTC residents and 40% of LTC residents being dispensed antipsychotics in any year there is a need to address this level of antipsychotic dispensation to older adults.

    Journal:  Incidence of antipsychotic use among community dwellers with and without Parkinson's disease. (Pubmed Central) -  Oct 26, 2022
    Persons with PD have symptoms treated with antipsychotics both before and after diagnosis. Psychotic symptoms may be challenging to recognize as prodromal symptoms since they can occur years before the motor symptoms and thus, they cannot be clinically associated with the diagnosis of PD.
