  • ||||||||||  Clinical, Observational data, Journal:  Antibiotic resistant Cutibacterium acnes among acne patients in Jordan: a cross sectional study. (Pubmed Central) -  Jun 30, 2021
    The findings of this study demonstrate the distribution of antibiotic resistance of C. acnes towards used antibiotics and emphasizes the influence of antibiotic consumption on development of antibiotic resistance. The similar pattern of resistance between skin bacteria tested in this study highlights the genetic transfer of resistance between skin commensals including S. aureus and S. epidermidis hence promoting its circulation in the community.

    Journal:  Metallo-β-lactamase resistance in Enterobacteriaceae is an artefact of currently utilized antimicrobial susceptibility testing methods. (Pubmed Central) -  Jun 26, 2021
    Results indicate that meropenem in vivo efficacy is best represented by the pharmacodynamic profile generated using MICs determined in zinc-depleted media for MBL-producing Enterobacteriaceae. These translational data suggest that the use of conventional CAMHB for MBL susceptibility testing is inappropriate in distinguishing meaningful in vivo resistance given that zinc concentrations are supraphysiological in conventional CAMHB and negligible at infection sites.

    Journal:  Repurposing drugs for treatment of Mycobacterium abscessus: a view to a kill. (Pubmed Central) -  Jun 26, 2021
    The low MICs and efficacy at clinically achievable concentrations mean that tebipenem/avibactam, daunorubicin, omadacycline and bedaquiline give a view of components of a three-drug regimen likely to effectively kill Mab. We propose pharmacokinetic/pharmacodynamic studies to identify such a regimen and the doses to be combined.

    Journal:  Cigarette smoke extract induces the Pseudomonas aeruginosa nfxC drug-resistant phenotype. (Pubmed Central) -  Jun 26, 2021
    Overall, our results showed that CSE may induce antibiotic resistance in P. aeruginosa. The results of antimicrobial susceptibility tests, transcriptome sequencing and qRT-PCR showed that CSE induced P. aeruginosa to the nfxC drug-resistant phenotype.
  • ||||||||||  minocycline / Generic mfg.
    Journal:  The impact of serum protein binding on bacterial killing of minocycline. (Pubmed Central) -  Jun 24, 2021
    If validated, these results implied dose escalation might not the best approach to improve the clinical efficacy of minocycline for bacteremia. Future investigations will focus on the specificity and mechanism(s) of minocycline protein binding.

    Journal:  Mutation rate and efflux response of bacteria exposed to a novel antimicrobial iodo-thiocyanate complex. (Pubmed Central) -  Jun 24, 2021
    Our results show strong bactericidal potential of CHP against M.tuberculosis that warrant its immediate mechanistic, pharmacokinetic and pharmacodynamic studies. ITC usage most likely will not promote resistance development via increased mutation rates, and that efflux-mediated resistance emergence to it is less likely than for some other antimicrobials.

    Journal:  Levaquin gets a pass. (Pubmed Central) -  Jun 24, 2021
    We evaluated the effect of levofloxacin (LEVO), the most commonly used prophylactic antibacterial antibiotic during intensive chemotherapy and allogeneic hematopoietic cell transplantation (allo-HCT), on the gut microbiota in 2 cohorts of patients, 1 cohort comprising 20 patients with acute leukemia receiving intensive chemotherapy and the other cohort comprising 20 allo-HCT recipients...Other taxa and microbiota diversity were unaffected. Overall, the effect of LEVO on the gut microbiota in these cohorts was mild.

    Journal:  Fluoroquinolone Prophylaxis in Autologous Stem Cell Transplantation: Worthy of a Second Look. (Pubmed Central) -  Jun 24, 2021
    In accordance with institutional policy, patients undergoing ASCT for multiple myeloma routinely receive levofloxacin prophylaxis during their period of neutropenia, whereas patients undergoing the ASCT for lymphoma do not...Fluoroquinolone prophylaxis reduces the rate BSI in patients undergoing ASCT but increase the incidence of resistant organisms. We recommend routine antimicrobial prophylaxis in patients undergoing ASCT to reduce the risk of BSI with systematic and regular review of outcomes.

    Clinical, Journal:  Which Nursing Home Residents With Pneumonia Are Managed On-Site and Which Are Hospitalized? Results from 2 Years' Surveillance in 14 US Homes. (Pubmed Central) -  Jun 24, 2021
    Respiratory rate is associated with hospitalization but was not documented for more than a quarter of residents, suggesting the clinical benefit of more consistently conducting this assessment. Differential hospitalization rates for persons with dementia and on hospice suggest that care is being tailored to individuals' wishes, but this assumption merits study, as does use of fluoroquinolones (due to side effects) and treatment duration (due to potential contribution to antibiotic resistance).

    Clinical, Journal:  Characteristic antimicrobial resistance of clinical isolated strain Stenotrophomonas maltophilia CYZ via complete genome sequence. (Pubmed Central) -  Jun 24, 2021
    Differential hospitalization rates for persons with dementia and on hospice suggest that care is being tailored to individuals' wishes, but this assumption merits study, as does use of fluoroquinolones (due to side effects) and treatment duration (due to potential contribution to antibiotic resistance). The current results give us a better understanding of the genetic characteristics of the antimicrobial resistance of S. maltophilia CYZ, and provide well genetic basis for further studying on the phenotype.

    Preclinical, Journal:  A mouse model of rapidly progressive fatal hemorrhagic pneumonia caused by Stenotrophomonas maltophilia. (Pubmed Central) -  Jun 24, 2021
    We established a mouse model of rapidly progressive fatal hemorrhagic pneumonia caused by Stenotrophomonas maltophilia which is useful for determining the therapeutic effect of various agents. We believe that this model will prove useful to further elucidate the mechanisms underlying hemorrhagic pneumonia, as well as in the design and development of novel therapeutic modalities and targets.

    Journal:  Antibiotic Therapy and the Gut Microbiome: Investigating the Effect of Delivery Route on Gut Pathogens. (Pubmed Central) -  Jun 24, 2021
    The degree of dysbiosis was significantly reduced toward the end of the sampling period, as bacterial populations began to return to pretreatment composition. Richness and diversity levels appeared to return to pretreatment levels more quickly in intravenous groups, suggesting convenient parenteral delivery systems may have a role to play in reducing longer term gut dysbiosis in the treatment of infection.

    Journal:  Drug Release from Thermo-Responsive Polymer Brush Coatings to Control Bacterial Colonization and Biofilm Growth on Titanium Implants. (Pubmed Central) -  Jun 23, 2021
    In particular, thermo-responsive poly(di(ethylene glycol) methyl ether methacrylate) (PDEGMA) brushes on titanium implants synthesized via a surface-initiated atom transfer radical polymerization with activators regenerated through the electron transfer technique (ARGET ATRP) allows for a controlled and thermally triggered release of the antibiotic levofloxacin at the wound site...At temperatures of the infection site >37 °C the lower critical solution temperature behavior of the brushes afforded the triggered release. Hence, in addition to the known antifouling effects, the PDEGMA coating ensured enhanced bactericidal effects, as demonstrated in initial in vivo tests with rodents infected with Staphylococcus aureus.
  • ||||||||||  OPC-167832 / Otsuka
    Journal:  OPC-167832, a novel carbostyril derivative with potent anti-tuberculosis activity as a DprE1 inhibitor. (Pubmed Central) -  Jun 22, 2021
    Finally, 3-4 drug regimens comprised of delamanid and OPC-167832 as the core along with bedaquiline, moxifloxacin, or linezolid showed superior efficacy in reducing bacterial burden and preventing relapse compared to the standard treatment regimen. In summary, these results suggest that OPC-167832 is a novel and potent anti-TB agent and regimens containing OPC-167832 and new or repurposed anti-TB drugs may have the potential to shorten the duration of treatment for TB.
