Journal: An inhalable nanoparticle enabling virulence factor elimination and antibiotics delivery for pneumococcal pneumonia therapy. (Pubmed Central) - Oct 17, 2024 Here we report an inhalable catalase (CAT)-tannic acid (TA) nanoassembly for local antibiotic (levofloxacin) delivery and simultaneously neutralizing the secreted H2O2 virulence factors to treat pneumococcal pneumonia...In synergy with antibiotic LVX-mediated S. pneumoniae elimination, CT@LVX significantly decreases lung injury companied with reduced inflammatory, resulting in 100?% survival of mice with pneumonia. In a clinically isolated S. pneumoniae strain-induced pneumonia mouse model, CT@LVX also shows superior outcomes compared to the traditional antibiotic treatment, highlighting its potential clinical application prospects.

Preclinical, Journal: Curcumin and Vitamin D Reduce HMGB-1 mRNA Levels in Mice Infected with Salmonella typhi. (Pubmed Central) - Oct 17, 2024 Mice in group A were treated with the antibiotic levofloxacin (1.95 mg/kg once daily) as the positive control; group B mice were administered curcumin at a dose of 200 mg/kg body weight; group C mice were treated with a curcumin dose of 200 mg/kg BW and vitamin D; and group D mice received distilled water (placebo) as the negative control...HMGB-1 mRNA level in group C decreased significantly by 5.76-fold (95% confidence interval: 2.55, 8.98). In contrast, HMGB-1 mRNA levels did not decrease significantly in group B. These results suggest that the combination of curcumin and vitamin D reduced HMGB-1 mRNA levels in infected mice, highlighting the potential of this combination as an antimicrobial and anti-inflammatory agent.

Antibiotic resistance of Acinetobacter baumannii against ciprofloxacin, levofloxacin, and piperacillin-tazobactam were statistically significantly different between the wards...In Ward 3, a significantly moderately positive association was observed for ceftazidime consumption and A. baumannii resistance (r = 0.61), positive correlation between piperacillin-tazobactam and E. coli resistance (r = 0.65), and a strong positive correlation in Ward 3 between levofloxacin and Pseudomonas aeruginosa resistance (r = 0.71)...This study has supported the involvement of pharmacists in the AMS team by conducting auditing activities that promote safe compliance of restricted antibiotic use among patients. Pharmacists can greatly participate on either prospective or retrospective review of antibiotic utilization and analyze trends of antibiotic consumption data to provide feedback to prescribing physicians on prescribing patterns and possible correlation with occurrence of antibiotic resistance.

In this report, we present the case of a 76-year-old male with compensated cirrhosis who presented with cellulitis complicated by bacteremia secondary to M. odoratus and was managed successfully with levofloxacin.
- |||||||||| A Rare Case of Stenotrophomonas maltophilia-Refractory Peritonitis in a Patient on Automated Peritoneal Dialysis () - Oct 12, 2024 - Abstract #KIDNEYWEEK2024KIDNEY_WEEK_5128;
Treated with empiric intra-peritoneal vancomycin and cefepime followed by cefpodoxime for 4-weeks...Antibiotics were changed to trimethoprim/sulfamethoxazole and levofloxacin, and PD catheter was removed...International guidelines recommend dual antibiotic coverage for environmental gram-negative bacteria such as Pseudomonas and Stenotrophomonas; special attention was paid to treatment of Stenotrophomonas in most recent guideline update. This case highlights the importance of considering this organism in patients with PD peritonitis regardless of their immunocompetence, the importance of hand hygiene, and also spotlights the resistant nature of the organism prompting us to be more vigilant when presented with such case..

- |||||||||| Review, Journal: Interaction of drug molecules with surfactants below (Benesi-Hildebrand equation) and above the critical micelle concentration (Kawamura equation). (Pubmed Central) - Oct 9, 2024
Various surfactants were analyzed with diphenhydramine hydrochloride, levofloxacin, phenothiazine, moxifloxacin, and chlorpromazine hydrochloride to determine monomeric binding constants, while sulfathiazole, sodium valproate, cefotaxime, losartan, and metformin hydrochloride were assessed for partitioning coefficient values. Errors in Benesi-Hildebrand plots may arise from considering surfactant concentrations above the CMC, while mistakes in Kawamura plots may stem from neglecting to determine the CMC in the presence of drug molecules, which can alter the surfactant's behavior.
