    Preclinical, Journal:  Does the in vitro egg hatch test predict the failure of benzimidazole treatment in Haemonchus contortus? (Pubmed Central) -  Aug 27, 2021   
    The results of the in vivo FECRT indicated that albendazole treatment reduced infections consisting of composite doses of 10, 20, 30, 40, 60, and 80% larvae of the resistant isolate by 91.3, 78.0, 63.3, 48.4, 36.5, and 41.4%, respectively...The EHT could thus accurately estimate the in vivo efficacy of the drug and percentage of the resistance allele in the population using hatching parameters in delineation doses. This finding was also supported by comparing the FECRT data to the hatching percentages in the EHT on 30 goat farms in Slovakia with natural mixed infections of gastrointestinal parasites.

    For the in vivo assay, CE-infected mice were divided into model group, albendazole (ABZ) group (200 mg/kg), low AS (AS-L) group (50 mg/kg), moderate AS (AS-M) group (100 mg/kg), and high AS (AS-H) group (200 mg/kg)...The content of HO in hydatid fluid treated by AS showed significant decrease compared with the model group (P < 0.01), while the T-SOD level showed significant elevation compared with model group (P < 0.01). In this study, we confirmed that the effects of AS on Echinococcus granulosus protoscoleces and metacestodes may be related to the DNA damages induced by oxidative stress, which provided solid information for the research and development of drugs for cystic echinococcosis.

    These genome-edited strains were exposed to both albendazole and fenbendazole to quantitatively measure animal responses to BZs...We performed competitive fitness assays and demonstrated that the E198V allele reduces animal health, supporting the hypothesis that this allele might be less fit in field populations. Overall, we present a powerful platform to quantitatively assess anthelmintic resistance and effects of specific resistance alleles on organismal fitness in the presence or absence of the drug.

    In this ex vivo study, S. cervi parasitoses were treated with Ivermectin (50 μM), Albendazole (200 μM) alone and Ivermectin + Albendazole (50 + 200 μM) at 37°C for 8 h and the motility and viability of the parasitoses were evaluated...Trafficking protein, acyl transferase, MATH involved in protein folding were also found to be downregulated. Thus, this study based on biochemical and proteomic approaches indicates that a combination of anti-filarial drugs Iver and Alb can alter calcium homeostasis in bovine filarial parasitosis leading to induction of ER stress culminating into apoptosis.

    Over the past few years we have initiated a collaborative drug discovery programme to develop new anthelmintic agents with activity against whipworm as a priority.2-5 An overview of our drug discovery pipeline will be provided, alongside the discovery and systematic investigation of the structure-activity relationship of a series of dihydrobenzoxazepinone (DHB) compounds that block motility of the murine intestinal nematode parasite, Trichuris muris ex vivo.2,5 Excitingly these compounds also show activity against other parasitic nematodes, including Brugia malayi and Heligmosomoides polygyrus and the trematode Schistosoma mansoni, a parasite that causes schistosomiasis, demonstrating their potential for optimisation as broad-spectrum anthelmintics.5 More recently we have discovered additional classes of small molecule which kill Trichuris muris in vitro and which significantly reduce the worm burden of infected mice in vivo following oral administration. Our lead compound retains similar levels of activity in a benzimidazole-resistant model parasite, showing that it acts through a distinct mechanism of action to front-line agents.

However, the resulting PK profiles for each combination of drug and ruminant species represents a new dataset that can be used to support the US FDA Center for Veterinary Medicine's Minor Use/Minor Species indexing process for drug approvals in minor species such as sheep and goats.

    ABZ itself had no effect on the bile acid profiles and the expression of relevant enzymes and transporters. A comparison of randomized trials is needed to compare the recurrence rates between splenectomy and spleen-preserving procedures in the treatment of giant splenic hydatid cysts.

    The experimental design-based and software-assisted method development proved to be a fast and reliable way of replacing a method with inadequate selectivity and long runtime with a robust UHPLC-based method, which offers baseline separation for all monitored impurities in 10 min. Results confirm that software-based chromatographic modelling can not only speed up the analytical method development process, but also improve the reliability of the developed method.

Sufficiently powered clinical and diagnostic studies are needed to overcome both the lacking evidence regarding salvage therapy and the diagnostic neglect of antimicrobial resistance.

    Treatment with 0.2 mg Albendazole/mouse/day temporarily reduces the number of affected organs in immunocompetent C57Bl/6 mice, but not in CD4 and CD8 mice...The E. cuniculi genotype III spore burden in individual organs reached up to 10 or 10 in immunocompetent or immunodeficient mice, respectively; however, these mice did not demonstrate any obvious clinical signs of microsporidiosis, and the immunodeficient mice survived longer. Our findings clearly show that the survival of mice does not correspond to spore burden, which provides new insight into latent microsporidiosis from an epidemiological point of view.
