(Pubmed Central) - Jul 8, 2020 Introducing a methyl substituent at the 2-methyl position of the CyHQ core enabled the bypass of the photorearrangement and significantly improved the aniline release efficiency. We successfully applied the strategy to the photoactivation of mifepristone (RU-486), an antiprogestin drug that is also used to induce the LexPR gene expression system in zebrafish and the gene-switch regulatory system based on the pGL-VP chimeric regulator in mammals.

(Pubmed Central) - Jul 7, 2020 However, mifepristone administered to females caused the decrease of the percentage of normal developed plutei larvae compared to the control, while it did not alter sperm motility parameters and fertilization success in males. The immunohistological localization of progesterone receptor-like immunoreactivity on the plasma membrane of oocytes and ova and the molecular weight of a progesterone receptor-like immunoband identified by western blot, are in agreement with a membrane progesterone receptor deducted from the genome sequence of the sea urchin Strongylocentrotus purpuratus and suggest that in Paracentrotus lividus mifepristone actions may be mediated by a progesterone receptor.

- |||||||||| [VIRTUAL] Non-Genomic Effects of Glucocorticoids Differentially Modulate Glucocorticoid Receptor Site-Specific Phosphorylation (ATS 2020 Virtual) - Jul 6, 2020 - Abstract #ATSI2020ATS-I_3745;
The role of protein synthesis as well as GR was also examined using cycloheximide and RU486, respectively...While SB203580 and LY294002 had little effect on CB-induced pGR-ser226, PD98059 and SP600125 dramatically reduced pGR-ser226...Conclusion Together, our findings show that the non-genomic effects of GC i) differentially affect GR phosphorylation by upregulating pGR-ser226, but not pGR-ser211, in a manner that is GR-independent but ERK/JNK dependent and ii) rapidly activate MAPK pathways. Further studies are still needed to determine the role of GR-independent early events such as nonspecific interactions with the cell membrane in regulating later GC genomic activities.

Overall, our study found that dexamethasone can promote embryonic stem cell-derived cardiomyocyte maturation through Parkin-mediated mitophagy.

[VIRTUAL] NALCN Expression Is Regulated by Progesterone and Estrogen in Human Myometrial Smooth Muscle Cells () - Jun 20, 2020 - Abstract #ENDOI2020ENDO-I_1853; Conversely, treatment with the ER antagonist, ICI 182,780, significantly increased NALCN mRNA expression, while treatment with the PR antagonist RU486 significantly decreased NALCN mRNA expression suggesting E2 and P4 work through their respective receptors to regulate NALCN...This data reveals a new mechanism by which NALCN is regulated in the myometrium and may suggest a novel role for NALCN during pregnancy. Further investigation into these novel roles can provide an insight into potential targets to modulate uterine quiescence and contractility.

Journal: Compound A inhibits urothelial tumorigenesis via both the androgen receptor and glucocorticoid receptor signaling pathways. (Pubmed Central) - Jun 9, 2020 Finally, CpdA was found to reduce AR transactivation and selectively induce GR transrepression (i.e. suppression of NF-κB transactivation and expression of its regulated genes), but not GR transactivation (i.e. activation of glucocorticoid-response element-mediated transcription and expression of its targets) in SVHUC cells. These findings suggest that CpdA suppresses urothelial tumorigenesis via both the AR and GR pathways, which may consequently provide an effective option of chemoprevention for bladder cancer, especially in patients with superficial disease following transurethral surgery.

Effect of Roscovitine and mifepristone in luminal breast cancer cells (Virtual Meeting II: E-Posters) - May 16, 2020 - Abstract #AACRII2020AACR-II_3870; Whereas both T47D-YA and -YB cells were similarly inhibited by Roscovitine, only in T47D-YA cells the combination of the antiprogestin mifepristone (MFP; 10 nM) and Roscovitine 1 µM in the presence of FGF2 induced a higher inhibitory effect than MFP or Roscovitine single treatments (p<0.01). We conclude that CDK2 inhibitors may be an option for luminal breast cancer treatment independent of the levels of cyclin A, and that they might be used in combination with antiprogestins only in breast cancer cells overexpressing PRA isoform.
