Journal:  Remote Reproductive Rights. (Pubmed Central) -  Jan 31, 2023
In July 2020, a federal district court lifted the U.S. Food & Drug Administration's ("FDA") restriction requiring patients to pick up the first drug of a medication abortion-mifepristone-at a healthcare facility...Those residing in states hostile to abortion will have to seek cross-border care, carry pregnancies to term, or find other avenues to end pregnancies. But the portability of abortion pills, when mailed by prescribers or dispensed by certified pharmacies, will test how closely states officials (or anyone else) can police or impede access to medication abortion.

Preclinical, Journal:  Disturbed relationship between glucocorticoid receptor and 5-HT1AR/5-HT2AR in ADHD rats: A correlation study. (Pubmed Central) -  Jan 27, 2023
The GR agonist can upregulate the expression of 5-HT and 5-HT2AR and downregulate the expression of 5-HT1AR in the prefrontal cortex of SHR as well as reduce the hyperactivity and attention deficit behavior in SHR, while the opposite was true for the GR inhibitor. It is suggested that the dysfunction of the 5-HT system in ADHD rats is closely related to glucocorticoid receptor activity.

Review, Journal:  No-Test Medication Abortion: A Systematic Review. (Pubmed Central) -  Jan 27, 2023
It is suggested that the dysfunction of the 5-HT system in ADHD rats is closely related to glucocorticoid receptor activity. Medication abortion performed without prior pelvic examination or ultrasonogram is a safe and effective option for pregnancy termination.

Journal:  Iris Pallida Extract Alleviates Cortisol-Induced Decrease in Type 1 Collagen and Hyaluronic Acid Syntheses in Human Skin Cells. (Pubmed Central) -  Jan 21, 2023
Moreover, IPE blocked the cortisol-induced translocation of the glucocorticoid receptor (GR) from the cytoplasm to the nucleus as effectively as the GR inhibitor mifepristone...Our results suggest that IPE can act as an inhibitor of cortisol in human skin cells, preventing cortisol-induced collagen and HA degradation by blocking the nuclear translocation of the GR. Therefore, IPE may be used as a cosmetic material or herbal medicine to treat stress-related skin changes.

Journal:  Mechanoactivation as a Tool to Assess the Autoxidation Propensity of Amorphous Drugs. (Pubmed Central) -  Jan 19, 2023
Additionally, the impact of adding an antioxidant (e.g., butylated hydroxytoluene) to the mixture during comilling was also examined. This study can be helpful in evaluating the stability of amorphous solids stored in accelerated (non-hermetic) conditions, in screening solid-state autoxidation propensity of drugs, and for the rational selection of antioxidants.

Journal:  The role of progesterone and the progesterone receptor in cancer: progress in the last 5 years. (Pubmed Central) -  Jan 18, 2023
Mifepristone may be less effective for cancers positive for the nPR because the nPR may be protective and blocking it may have detrimental effects. Based on this hypothetical model, the development of other potential treatment options to provide even greater efficacy for treating cancer are discussed.

Journal:  MYB and ELF3 differentially modulate labor-inducing gene expression in myometrial cells. (Pubmed Central) -  Jan 4, 2023
Furthermore, both MYB and ELF3 proteins affect labor-driving gene promoter activity, although in surprisingly opposing ways: Gja1 and Fos promoter activation increases in the presence of MYB and decreases in the presence of ELF3. Collectively, our study adds to the current understanding of the transcription factor network that defines the transcriptomes of SMCs during late gestation and implicates two new players in the control of labor timing.

Journal:  Long-term use of ultra-low-dose mifepristone for uterine leiomyoma control and safety discussion: A case report. (Pubmed Central) -  Jan 4, 2023
Collectively, our study adds to the current understanding of the transcription factor network that defines the transcriptomes of SMCs during late gestation and implicates two new players in the control of labor timing. Judging from our case, long-term, ultra-low-dose mifepristone for uterine fibroids treatment was safe and more effective than conventional therapy.

Preclinical, Journal:  Early life stress induces visual dysfunction and retinal structural alterations in adult mice. (Pubmed Central) -  Dec 31, 2022
A treatment with mifepristone, injected every 3 days between PNDs 4-16, prevented functional and structural alterations induced by MSEW. These results suggest that retinal alterations might be included among the childhood adversity-induced threats to life quality, and that an early intervention with mifepristone avoided ELS-induced retinal disturbances.

Journal:  The impact of criminalisation on abortion-related outcomes: a synthesis of legal and health evidence. (Pubmed Central) -  Dec 30, 2022
International human rights bodies have recognised that criminalisation results in the provision of poor-quality healthcare goods and services, is associated with lack of registration and unavailability of essential medicines including mifepristone and misoprostol, obstructs the provision of abortion information, obstructs training for abortion provision, is associated with delayed and unsafe abortion, and does not achieve its apparent aims of ether protecting abortion seekers from unsafe abortion or preventing abortion...It provides a further underpinning from empirical evidence of the harms of criminalisation that have already been identified by human rights bodies. It also provides additional evidence to support the WHO's recommendation for full decriminalisation of abortion.

Journal:  Mifepristone, preemption, and public health federalism. (Pubmed Central) -  Dec 27, 2022
States have long regulated drugs in ways that diverge from FDA, arguably increasingly so in recent years. Accordingly, the article investigates the implications that preemption challenges in the abortion context may have for other areas of state drug regulation, making the case that the benefits of public health federalism need not be undermined by successful preemption challenges in the abortion arena.

Journal:  Social inequalities in utilization of a feminist telehealth abortion service in Brazil: A multilevel analysis. (Pubmed Central) -  Dec 24, 2022
Independently of this, service utilization was higher in states with higher income and education access, with lower proportions of racialized women, and located in the South, Southeast and Central-West regions. We concluded that while feminist telehealth abortion initiatives provide a life-saving service for some abortion seekers, they are not fully equipped to overcome entrenched social inequalities in their utilization, both at individual and contextual levels.

Journal:  Glucocorticoid-induced microRNA-378 signaling mediates the progression of pancreatic cancer by enhancing autophagy. (Pubmed Central) -  Dec 20, 2022
The GC receptor (GR) antagonist mifepristone (RU486) counteracted DEX-induced autophagy features, suggesting that the GC-GR complex is involved in the induction of autophagy...The construction of a molecular docking model revealed stable binding of miR-378 to the DEX-GR complex, suggesting direct regulation. These substantial, novel, in-depth data reveal that GCs favor autophagy-mediated cancer progression by inducing miR-378 and GR binding and implicate GR and miR-378 as new therapeutic targets.

Journal:  Transcriptomic profiling of canine decidualization and effects of antigestagens on decidualized dog uterine stromal cells. (Pubmed Central) -  Dec 20, 2022
Maternal-stroma derived decidual cells, the only cell population in the canine placenta expressing the nuclear progesterone (P4) receptor (PGR), are crucial for the maintenance of canine pregnancy...The antigestagens aglepristone and mifepristone ablate the expression of decidualization markers in DUS cells (e.g., PGR, PRLR, IGF1 or PTGES)...Comparison between antigestagen-mediated effects and transcriptional changes in the canine placenta at term allowed the identification of 191 DEGs associated with diminished cell proliferation and adhesion, and vascular and immune modulation. This study emphasizes the importance of P4/PGR signaling for decidual cell function, providing new insights into the maintenance of canine pregnancy.

Journal:  Fasting influences aquaporin expression, water transport and adipocyte metabolism in the peritoneal membrane. (Pubmed Central) -  Dec 16, 2022
Fasting induces a coordinated regulation of lipolytic and lipogenic factors and aqua(glycero)porins in the peritoneum, driving structural and functional changes. These data yield novel information on the specific roles of aquaporins in the peritoneal membrane and indicate that fasting improves fluid removal in a mouse model of PD.

Journal:  Medical abortion in India - An imperative need for task sharing. (Pubmed Central) -  Dec 13, 2022
Majority (101; 90%) took two drugs (Mifepristone and Misoprostol), 28 (25%) used correct dosage...Incorrect dosage and lack of gestational age calculation were two most common errors in the risk assessment protocol. Expanding provider base, by training midlevel providers, can overcome these and unmask the full potential of MA to make abortion safer.

Journal:  Comparing preference for and use of medication abortion in Texas after policy changes in 2014 and 2018. (Pubmed Central) -  Dec 7, 2022
State policies can affect people's ability to obtain their preferred abortion method. Efforts to provide both abortion options whenever possible, and inform people where each can be obtained, remains an important component of person-centered care despite increasing state abortion restrictions and bans following the reversal of Roe v Wade.

Journal:  How much will it hurt? Factors associated with pain experience in women undergoing medication abortion during the first trimester. (Pubmed Central) -  Dec 6, 2022
Efforts to provide both abortion options whenever possible, and inform people where each can be obtained, remains an important component of person-centered care despite increasing state abortion restrictions and bans following the reversal of Roe v Wade. The identification of women at risk for severe pain based on clinical and historical factors as well as the definition of an adequate analgesic regimen may help to improve women's care and pain management during medication abortion.
  • ||||||||||  relacorilant (CORT125134) / Corcept Therap
Journal:  Peripheral glucocorticoid receptor antagonism by relacorilant with modest HPA axis disinhibition. (Pubmed Central) -  Nov 30, 2022
In human HEK-293 cells, relacorilant potently antagonized dexamethasone- and cortisol-induced GR signaling, and in human peripheral blood mononuclear cells relacorilant largely prevented the anti-inflammatory effects of dexamethasone...This contrast between relacorilant and mifepristone is possibly due to the distinct transcriptional coregulator recruitment by the GR. In conclusion, relacorilant is thus an efficacious peripheral GR antagonist in mice with only modest disinhibition of the HPA axis, and the distinct properties of relacorilant endorse the potential of selective GR antagonist treatment for endogenous Cushing's syndrome.

Journal:  A screen of small molecule and genetic modulators of life span in female Drosophila identifies etomoxir, RH5849 and unanticipated temperature effects. (Pubmed Central) -  Nov 23, 2022
Mifepristone increases life span in female Drosophila melanogaster, and its molecular target(s) remain unclear...Pioglitazone is reported to activate both mammalian PPARγ and its Drosophila homolog Eip75B...Even when applied after mating was complete, a shift to 29°C temperature reduced mating-induced midgut hypertrophy by half, and the life span effects of mating by 4.8-fold. Taken together, these results identify promising small molecules for further analysis, and inform the design of experiments involving the GAL4/GAL80ts system.

Clinical, Retrospective data, Review:  The efficacy of mifepristone combined with methotrexate for the treatment of ectopic pregnancy: a systematic review and meta-analysis. (Pubmed Central) -  Nov 18, 2022
The combination of mifepristone and methotrexate can improve the efficacy of ectopic pregnancy without amplifying the toxic side effects. Larger scale and better design of the randomized controlled trials are needed.KEY MESSAGESIn recent years, the increase in ectopic pregnancies and their impacts on female fertility makes physicians have to find an effective medical treatment as soon as possible that can avoid surgery. The mifepristone combined with methotrexate therapy for EP has better curative effects on improving the cure rate, lowering β-HCG level, reducing the mass, and alleviating symptoms of abdominal pain and bleeding, without amplifying the toxic side effects.Literature with high quality is lacking, and well-designed, large-scale and high-quality multicenter randomized controlled trials are needed.
