Retrospective data, Review, Journal:  Intravenous lidocaine in spine surgery: A meta-analysis of randomized controlled trials. (Pubmed Central) -  Feb 11, 2022
Cumulative morphine consumption at 48 h was not statistically significant between lidocaine and control groups...Our results indicated that current literature does not support the use of intravenous lidocaine as an adjunctive measure of pain management after spine surgery. Given the relatively few numbers of studies in this field, further randomized controlled trials are needed to make a definitive conclusion on the effectiveness of lidocaine in spine surgery patients.

Preclinical, Journal:  Contrasting effects of the α7 nicotinic receptor antagonist methyllycaconitine in different rat models of heroin reinstatement. (Pubmed Central) -  Feb 10, 2022
We showed previously that the α7 nicotinic acetylcholine receptor antagonist, methyllycaconitine, specifically attenuated morphine-primed reinstatement of conditioned place preference in rodents and this effect was mediated in the ventral hippocampus...The α7 nicotinic acetylcholine receptor antagonist, methyllycaconitine, prevented reinstatement of the opioid conditioned place preference, consistent with a role for α7 nicotinic acetylcholine receptors in the retrieval of associative memories of drug liking. The lack of effect of methyllycaconitine in heroin-dependent rats in two intravenous self-administration models suggests that α7 nicotinic acetylcholine receptors do not play a role in later stages of heroin abuse.

Journal:  The role of circTmeff-1 in incubation of context-induced morphine craving. (Pubmed Central) -  Feb 10, 2022
These findings suggested that circTmeff-1, a novel circRNA, promotes incubation of context-induced morphine craving by sponging miR-541/miR-6934 in the NAc core. Thus, circTmeff-1 represents a potential therapeutic target for context-induced opioid craving, following prolonged abstinence.

Preclinical, Journal:  Myelopeptides Reduce Morphine Tolerance in C57BL/6j Mice. (Pubmed Central) -  Feb 10, 2022
In the hot plate test, only three myelopeptides (MP2, MP5, and MP6) were found to reduce the formation of morphine tolerance. MP1 significantly reduced the analgesic effect of morphine on days 1-3 of administration, but contributed to the preservation of the analgesic effect during the period of tolerance development.

Review, Journal:  Endogenous Opiates and Behavior: 2019. (Pubmed Central) -  Feb 9, 2022
This paper is the forty-second consecutive installment of the annual anthological review of research concerning the endogenous opioid system, summarizing articles published during 2019 that studied the behavioral effects of molecular, pharmacological and genetic manipulation of opioid peptides and receptors as well as effects of opioid/opiate agonists and antagonists. The review is subdivided into the following specific topics: molecular-biochemical effects and neurochemical localization studies of endogenous opioids and their receptors (1), the roles of these opioid peptides and receptors in pain and analgesia in animals (2) and humans (3), opioid-sensitive and opioid-insensitive effects of nonopioid analgesics (4), opioid peptide and receptor involvement in tolerance and dependence (5), stress and social status (6), learning and memory (7), eating and drinking (8), drug abuse and alcohol (9), sexual activity and hormones, pregnancy, development and endocrinology (10), mental illness and mood (11), seizures and neurologic disorders (12), electrical-related activity and neurophysiology (13), general activity and locomotion (14), gastrointestinal, renal and hepatic functions (15), cardiovascular responses (16), respiration and thermoregulation (17), and immunological responses (18).

Preclinical, Journal:  Treatment with dopamine β-hydroxylase (DBH) inhibitors prevents morphine use and relapse-like behavior in rats. (Pubmed Central) -  Feb 9, 2022
These results seem to point to the significance  of DBH inhibition as a potential pharmacotherapy against morphine use disorders.

Preclinical, Journal:  Effects of the selective dopamine D receptor antagonist PG01037 on morphine-induced hyperactivity and antinociception in mice. (Pubmed Central) -  Feb 9, 2022
These results suggest that attenuation of acute opioid-induced hyperactivity may be a behavioral effect shared among DR-selective antagonists, thus supporting continued investigations into their use as potential treatments for opioid use disorder. However, PG01037 is unlike newer, highly-selective DR antagonists in its capacity to reduce opioid-induced antinociception, indicating that modulation of opioid analgesia may vary across different DR antagonists.

Clinical, Retrospective data, Journal:  Next day discharge after the Nuss procedure using intercostal nerve cryoablation, intercostal nerve blocks, and a perioperative ERAS pain protocol. (Pubmed Central) -  Feb 9, 2022
However, PG01037 is unlike newer, highly-selective DR antagonists in its capacity to reduce opioid-induced antinociception, indicating that modulation of opioid analgesia may vary across different DR antagonists. INC combined with bupivacaine intercostal nerve blocks and a pre- and post-hospital analgesia protocol facilitated discharge one day after the Nuss procedure, achieved excellent pain control, and eliminated the need for intravenous opioids.

Journal:  Benefits of the enhanced recovery after surgery pathway for orthognathic surgery. (Pubmed Central) -  Feb 8, 2022
The patient charts were analyzed for postoperative opioid consumption (measured in morphine milligram equivalents, MME) and PONV...The ERAS group also had a lower incidence of PONV, with 1.2 episodes of PONV compared to 2.4 episodes in the non-ERAS group (P= 0.008). This study demonstrates that the ERAS protocol is effective in decreasing postoperative opioid consumption and PONV.

Preclinical, Journal:  Experimental Study on the Central Mechanism of Penehyclidine Hydrochloride against Relapse Behavior in Morphine-Dependent Rats. (Pubmed Central) -  Feb 8, 2022
Each group was randomly divided into 5 subgroups (n = 10): 4 h after administration, 7 h after administration, 13 h after administration, 25 h after administration (K48, LP48, MP48, and HP48), and 37 h after administration, and then, Morris water maze experiment and immunohistochemical detection of the rat brain hippocampus were carried out. 4 and 7 hours after administration, compared with group 1, the TchE activity increased and Ach level decreased in groups 2, 3, and 4 and the difference was significant (P < 0.05), so the principle of penehyclidine hydrochloride against morphine-dependent rats is that penehyclidine hydrochloride causes cognitive impairment in the brain of mice, thereby achieving antimorphine effects.
