    Journal:  Platelet-derived growth factor D expression in adrenal cells is modulated by corticosteroids: putative role in adrenal suppression. (Pubmed Central) -  May 15, 2022
    This builds on previous research that identified a SNP in PDGFD (rs591118) as an independent risk factor for adrenal suppression in adults and children with obstructive airway disease treated with inhaled corticosteroids. First in vitro experiments to support a link between the PDGF and cortisol production pathways, supporting the hypothesis that PDGFD variants can affect an individual's sensitivity to corticosteroid-induced adrenal suppression.

    Journal:  Effect of Periocular Triamcinolone Acetonide on Prevention of Postsurgical Intraretinal Cystoid Changes after Vitrectomy with Membrane Peeling. (Pubmed Central) -  May 15, 2022
    First in vitro experiments to support a link between the PDGF and cortisol production pathways, supporting the hypothesis that PDGFD variants can affect an individual's sensitivity to corticosteroid-induced adrenal suppression. Periocularly applied triamcinolone acetonide at the end of vitrectomy with membrane peeling was well tolerated and showed prevention of early transient macular edema but did not prevent the development of new postoperative intraretinal cystoid changes.

    Review:  Quantitative comparison of different inhaled corticosteroids in the treatment of asthma in children. (Pubmed Central) -  May 14, 2022
    This study analyzed the entire time-course of the drug efficacy of Inhaled corticosteroids in the treatment of asthma in children aged 5-12, which found that although the maximum efficacy of both ciclesonide and budesonide was the same, the onset speed of ciclesonide was faster than that of budesonide. The above information provides the necessary quantitative information for the implementation of medication guidelines for steps 2-3 asthma in children.

    Journal:  Lysine deacetylases (KDACs) regulate PKCõ phosphorylation in response to muscle atrophy. (Pubmed Central) -  May 14, 2022
    Using C2C12 myotubes stimulated with dexamethasone (Dex) as a model of muscle atrophy, we report that KDAC inhibition using Trichostatin A (TSA) restored myotube size and attenuated Dex-induced atrophy genes, Murf1 and Atrogin-1...Consistent with this hypothesis, we report that KDAC inhibition could not restore myotube size in response to Dex in the presence of a PKCδ inhibitor or when overexpressing a dominant negative PKCδ. Combined, these suggest a non-canonical role for KDACs in the regulation of muscle physiology outside of its role in chromatin condensation and gene regulation.

    Preclinical, Journal:  Physiological Effects of Binary Mixtures of Common Bath Salt Constituents: Studies with MDPV, Methylone, and Caffeine in Rats. (Pubmed Central) -  May 14, 2022
    These data demonstrate that intravenous administration of either MDPV, methylone, or caffeine alters temperature, heart rate, blood pressure, and locomotor activity in a dose-dependent manner, consistent with what has been reported in "bath salts" users in emergency room settings. To more deeply characterize the interactions between these constituents, future studies will determine the degree to which the reinforcing and physiological effects of these drug mixtures could be mediated by pharmacokinetic interactions between individual constituents.

    Journal:  Glucocorticoids Preferentially Influence Expression of Nucleoskeletal Actin Network and Cell Adhesive Proteins in Human Trabecular Meshwork Cells. (Pubmed Central) -  May 14, 2022
    To understand how proteins, including cytoskeletal and cell adhesion proteins that are recognized to shuttle between the cytosolic and nuclear regions, influence gene expression and other cellular activities, we used proteomic analysis to characterize the nuclear protein fraction of dexamethasone (Dex) treated human TM cells...Interestingly, deficiency of BRG1 expression blunted the Dex-induced increases in the levels of some of these proteins in TM cells. In summary, these findings indicate that the widely recognized changes in actin cytoskeletal and cell adhesive attributes of TM cells by glucocorticoids involve actin regulated BRG1 chromatin remodeling, nuclear MICAL2, and glypican-4 regulated Wnt signaling upstream of the serum response factor/myocardin controlled transcriptional activity.

    Clinical, Journal:  A Single Glucocorticoid Injection Accelerate Erythropoiesis but Does Not Improve Performance. (Pubmed Central) -  May 14, 2022
    In conclusion, a single intramuscular injection of triamcinolone acetonide was sufficient to accelerate erythropoiesis and increase Hb , but did not translate into improved aerobic exercise performance in the present study. Future research should confirm whether anti-doping authorities should consider glucocorticoid injections as a method of blood volume manipulation.

    Preclinical, Journal:  Early Life Stress Induced Hypothalamic-Pituitary-Adrenal (HPA) Axis Dysfunction Promotes Chronicity of Experimental Colitis in Mice. (Pubmed Central) -  May 14, 2022
    Furthermore, pharmacologic induction of HPA-axis dysfunction via early life exposure (PD 1-14) to the synthetic glucocorticoid, dexamethasone, mirrors the results we observed in the MSEW model of ELS - both prior to and subsequent to colitis induction. Our results indicate that early life exposure to prolonged stress leads to HPA-axis dysfunction and colonic corticosterone deficits which may subsequently promote persistent colonic inflammation in susceptible hosts.

    HIGH-THROUGHPUT EVALUATION OF THE POTENTIAL OF CANCER DRUGS TO ENHANCE NATURAL KILLER CELL IMMUNOTHERAPY IN CHRONIC MYELOID LEUKEMIA. (Hall Stolz 1-2) -  May 13, 2022 - Abstract #EHA2022EHA_2672;
    For example, dasatinib and imatinib have been suggested to enhance NK cell cytotoxicity through regulation of activating and inhibitory receptors...The SMAC mimetics birinapant and LCL-161 were the most potent enhancers of NK cell cytotoxicity and had no effect on target cells alone...On the contrary, cytotoxicity-inhibiting drugs included previously known immunosuppressive drugs dexamethasone and prednisolone, as well as novel candidates, such as sotrastaurin, a PKC inhibitor...Conclusion We discovered novel drug classes, which had both activating and inhibitory effects on the NK cell cytotoxicity against CML cells in vitro. Defining NK cell - drug - CML cell interactions in a high-throughput setting provides a framework for future combination immunotherapies to further improve treatment-free remission in CML.

    LENALIDOMIDE–BASED MAINTENANCE AFTER AUTOLOGOUS HEMATOPOIETIC STEM CELL TRANSPLANT FOR PATIENTS WITH HIGH RISK MULTIPLE MYELOMA () -  May 13, 2022 - Abstract #EHA2022EHA_2480;
    Len-combo regimens were either doublets (Len with either dexamethasone [dex: n=10], elotuzumab [n=28] or ixazomib [n=14]) or triplets (Len with either bortezomib/dex [n=10], ixazomib/dex [n=10] or carfilzomib/dex [n=6])...Busulfan + melphalan (Bu-Mel) conditioning was used in 12% vs. 18% in Len-only vs. Len-combo groups (p=0.32)...Conclusion In this single center retrospective analysis, intensification of post-transplant Len maintenance did not show an improvement in the outcomes for patients with HRMM. However, there was a trend towards improved PFS in patients with HR abnormalities other than 1q+.
  • ||||||||||  OUTPATIENT AUTOTRANSPLANTS FOR PERSONS WITH MULTIPLE MYELOMA () -  May 13, 2022 - Abstract #EHA2022EHA_2442;
    Background Patients younger than 70 years diagnosed with multiple myeloma (MM) and without associated comorbidities routinely receive consolidation treatment with hematopoietic Stem Cell Transplantation (SCT) conditioned with high-dose melphalan after several induction cycles...8 were in the first line of treatment (induction with bortezomib-lenalidomide-dexamethasone scheme)...The most frequent reason for readmission is fever, and they can be discharged with empirical intravenous antibiotic treatment in the first 24 hours in most cases. With our results, supported by the experience of other centers, we are considering the cancellation of the prophylactic administration of G-CSF to reduce the incidence of engrafment syndrome.

    SINGLE NUCLEOTIDE POLYMORPHISMS OF THE HIF1A GENE ARE ASSOCIATED WITH SENSITIVITY OF GLUCOCORTICOID TREATMENT IN PEDIATRIC ITP PATIENTS () -  May 13, 2022 - Abstract #EHA2022EHA_2243;
    Moreover, the CC genotype was a risk factor for insensitive to GT the OR (95% confidence interval) was 5.96(5.23-6.69) in standard prednisone (PDN) (P=.0069) and 6.35(5.33-7.37) in high-dose dexamethasone (HDD) (P=.04). Conclusion Although HIF1A gene polymorphisms were not associated with susceptibility to ITP, the CT genotype at rs11549465 was associated with the sensitivity to glucocorticoid-treatment of ITP patients, suggesting that the rs11549465 SNP may contribute to the sensitivity of glucocorticoid treatment in pediatric ITP patients.
