Review, Journal:  Sumatriptan-naproxen sodium in migraine: A review. (Pubmed Central) -  Sep 25, 2024
Multi-mechanistic therapeutic agents offer an opportunity to optimise acute medications by targeting multiple mediators involved in the pathogenesis of migraine. Sumatriptan-naproxen sodium resulted in greater initial and sustained pain freedom, compared with either sumatriptan, naproxen-sodium and/or placebo, for the treatment of single or multiple attacks of migraine across both adult and adolescent study populations.

Journal:  Metformin inhibits OCT3-mediated serotonin transport in the placenta. (Pubmed Central) -  Sep 25, 2024
Our findings across all experimental models show inhibition of placental OCT3 by metformin, resulting in reduced serotonin uptake by the trophoblast. This sheds light on mechanisms that may underpin metformin-mediated effects on fetal development.

    Topography of Corrugator Supercilii Muscle Relative to the Eyebrow and Its Clinical Application for Blepharospasm (DEMAND) -  Sep 24, 2024 - Abstract #AAO2024AAO_2269;
Purpose This study aimed to map the corrugator supercilii muscle (CSM) location relative to the eyebrow to guide onabotulinumtoxinA (OBT-A) injections for blepharospasm treatment...Conclusion For effective targeting of the CSM, it is recommended to administer OBT-A injections precisely at the center of the medial end of the eyebrow and just above the midpupillary line. Considering the minimal presence of the CSM beyond the midpupillary line, injecting OBT-A into the CSM in this lateral region are typically considered unnecessary.

Journal:  ATG10S promotes IFNL1 expression and autophagic degradation of multiple viral proteins mediated by IFNL1. (Pubmed Central) -  Sep 24, 2024
These results clarified that ATG10S promoted autophagosome formation with the assistance of IFNL1 to ensure autophagy flux and autophagic degradation of multiple viral proteins and that ATG10S could also act as a novel transcription factor to promote IFNL1 gene expression. Importantly, this study further explored the antiviral mechanism of ATG10S interaction with type III interferon and provided a theoretical basis for the development of ATG10S into a new broad-spectrum antiviral protein drug.

Journal:  Pharmacological fingerprint of antipsychotic drugs at the serotonin 5-HT2A receptor. (Pubmed Central) -  Sep 24, 2024
In addition, G protein-selective partial agonism was found for aripiprazole and cariprazine...Computational analyses of the pharmacological and structural fingerprints support a mechanistically based clustering that recapitulate the clinical classification (typical/first generation, atypical/second generation, third generation) of the antipsychotic drugs. The study provides a new framework to functionally classify antipsychotics that should represent a useful tool for the identification of better and safer neuropsychiatric drugs and allows formulating hypotheses on the links between specific signaling cascades and in the clinical outcomes of the existing drugs.

Journal:  Genetic contribution to microglial activation in schizophrenia. (Pubmed Central) -  Sep 24, 2024
Overall, despite the increased expression of inflammatory genes, we observed no clear functional signs of hyperactivation in microglia from patients with SCZ. We conclude that microglia of the patients with SCZ have gene expression aberrations related to inflammation response and extracellular matrix without contributing to increased microglial activation.

Journal:  Bipolar Disorder in a female with CDKL5 Deficiency Disorder: A Case Report. (Pubmed Central) -  Sep 24, 2024
Conclusions : While sleep and behavioral disorders fall within the expected phenotype for CDD, this is the first report of bipolar disorder. Careful attention to patterns of sleep and behavior that may indicate mood cycling in this population is required, particularly in the setting of limited communication and functional abilities.

Journal, Adverse events:  Normocytic Anemia as a Side Effect of Clozapine: A Case Report. (Pubmed Central) -  Sep 24, 2024
This rare case underscores the importance of considering red blood cell parameters in patients on clozapine, offering insights that go beyond the commonly reported agranulocytosis side effect and potentially expanding awareness of adverse hematological effects associated with this antipsychotic medication. Further research is crucial to unravel the underlying mechanisms and establish definitive causality.

Journal:  The neurotrophin artemin and its receptor GFR?3 mediate migraine-like pain via the ion channel TRPM8. (Pubmed Central) -  Sep 24, 2024
Further, we show TRPM8 expression in the meninges and that direct supradural infusion of either a TRPM8-specific agonist or artemin itself produces mechanical allodynia, the latter dependent on TRPM8 and ameliorated by concurrent treatment with sumatriptan. These results indicate that neuroinflammatory events in the meninges can produce migraine-like pain in mice via artemin and GFR?3, likely acting upstream of TRPM8, providing a novel pathway that may contribute to migraine pathogenesis.

Journal:  Effect of antiemetics on zolbetuximab-induced gastric injury and emesis in ferrets. (Pubmed Central) -  Sep 23, 2024
Fosaprepitant showed suppressive effects on emesis, and use of dexamethasone or concomitant use of fosaprepitant with other antiemetics tended to alleviate gastric tissue damage. The onset of emesis in humans receiving zolbetuximab may be associated with damage in the gastric mucosa, and antiemetics may mitigate gastrointestinal adverse events.

Review, Journal:  The Impact of Antidepressants on COVID-19 and Post-Acute COVID-19 Syndrome: A Scoping-Review Update (Pubmed Central) -  Sep 23, 2024
In regions where neither vaccines nor antiviral agents currently approved for the prevention or treatment of COVID-19 are available, AD and in particular fluvoxamine would be a cost-effective alternative to protect against a severe course, even if this AD appears to have a smaller effect against COVID-19 than the currently approved antiviral agents, but with presumably better tolerability. A direct comparative clinical trial with approved antiviral agents is still pending and should be positive to further open the door for a guideline-based recommendation of fluvoxamine (or perhaps even AD) for COVID-19 or its aftermath.
