  • ||||||||||  Journal:  Human cytomegalovirus degrades DMXL1 to inhibit autophagy, lysosomal acidification, and viral assembly. (Pubmed Central) -  Apr 15, 2024
    Formation of the virion assembly compartment, which requires lysosomes, occurs significantly later with US33A-expressing virus infection, with reduced viral replication. These data thus identify a viral strategy for cellular remodeling, with the potential to employ US33A in therapies for viral infection or rheumatic conditions, in which inhibition of lysosome acidification can attenuate disease.

    Journal:  Celastrol enhances the viability of random-pattern skin flaps by regulating autophagy through the AMPK-mTOR-TFEB axis. (Pubmed Central) -  Apr 11, 2024
    Additionally, CEL stimulated angiogenesis, suppressed apoptosis, and lowered oxidative stress levels immediately after elevated autophagy in ischemic regions; These effects can be reversed using the autophagy inhibitor chloroquine (CQ)...This investigation suggests that CEL benefits the survival of RPSF by augmenting angiogenesis and impeding oxidative stress and apoptosis. The results are credited to increased autophagy, made possible by the AMPK-mTOR-TFEB axis activation.

    Unfortunately, effective strategies to target these cell survival pathways have not yet been successfully developed. Thus, there is an urgent need for the continued interrogation of autophagy and "reversible" senescence in clinically relevant breast cancer models with the long-term goal of identifying new molecular targets for improved treatment of ER+ breast cancer.
  • ||||||||||  Biomarker, Journal, Tumor microenvironment, IO biomarker:  CDKL3 shapes immunosuppressive tumor microenvironment and initiates autophagy in esophageal cancer. (Pubmed Central) -  Apr 6, 2024
    And, radiation could downregulate CDKL3 expression and autophagy induction, while ESCC patients with high CDKL3 expression had a significantly lower response rate after nCRT than those with low CDKL3 expression. CDKL3 may play an important role in anti-tumor immunity by regulating autophagy to promote the formation of immunosuppressive TME, thus playing a critical role in the prognosis of ESCA.
  • ||||||||||  Review, Journal:  Mitophagy and spermatogenesis: Role and mechanisms. (Pubmed Central) -  Apr 5, 2024
    Furthermore, studies have shown that inhibited autophagy-infected spermatozoa had reduced motility and increased amounts of phosphorylated PINK1, TOM20, caspase 3/7, and AMPK. Therefore, in terms of reproductive physiology, mitophagy is the removal of mitochondria derived from sperm and the following preservation of mitochondria that are exclusively maternal.

    Animal experiments also confirmed the results of cell experiments and attenuated the effects of overexpression of Sestrin2 after injection of the ferroptosis activators erastin or 3-MA. Our study revealed that Sestrin2 inhibits ferroptosis by inhibiting STAT3 phosphorylation and ER stress and promoting autophagy levels, thereby alleviating DR.
  • ||||||||||  Review, Journal:  Crosstalk between m6A modification and autophagy in cancer. (Pubmed Central) -  Apr 4, 2024
    m6A modification can either directly inhibit autophagy or promote its initiation, but the complex relationship between m6A modification, autophagy, and cancer remains poorly understood. Therefore, this paper aims to review the dual role of m6A and autophagy in cancer, explore the impact of m6A modification on autophagy regulation, and discuss the crucial role of the m6A modification-autophagy axis in cancer progression and treatment resistance.

    Calcitriol modulates intestinal AMPK/SIRT1 signaling towards alleviating compromised barrier integrity in MASH: a potential autophagy-dependent role (Poster Area) -  Apr 2, 2024 - Abstract #EASLILC2024EASL_ILC_1102;
    In this study, we utilized a diet-induced MASH model in male C57BL/6 mice randomly allocated into four experimental groups: (I) a normal standard chow diet group, (II) a group fed with a high-fat diet (HFD), (III) a group treated with CAL at 5 ng/gm/day i.p. twice a week, and (IV) a group that received both CAL and hydroxychloroquine (HQ; an autophagy inhibitor), at a dosage of 60 mg/kg/day, p.o. Interventions were initiated after 12 weeks of MASH induction and mice were sacrificed at the end of week 16... The present study findings suggest the capacity of CAL to improve barrier function and curb intestinal inflammation in MASH partly through augmenting AMPK/SIRT1 signaling in an autophagy dependent manner.
