  • ||||||||||  Journal:  Stapled Phd Peptides Inhibit Doc Toxin Induced Growth Arrest in Salmonella. (Pubmed Central) -  Dec 19, 2023   
    After making a series of substitutions to improve bacterial uptake, we identified a lead stapled Phd peptide that is able to counteract Doc toxicity in Salmonella. This provides an exciting starting point for the further development of therapeutic peptides capable of reducing antibiotic persistence in pathogenic bacteria.

    Besides exhibiting a preference for double-stranded RNA, we find that the RhsP2 activity is highly promiscuous with identified cellular targets including numerous non-coding RNAs, including the entire tRNA pool and the essential RNA-processing ribozyme, RNase P. Further characterization of RhsP2-intoxicated cells indicates that target cell death arises from the simultaneous inhibition of protein translation and disruption of RNA processing. Overall, our data demonstrate a new mechanism of bacterial antagonism and uncover an unprecedented enzymatic activity catalyzed by the ART family of enzymes.

    Therefore, this study might provide a new target against S. aureus infections. Galloylated catechins modify protein toxin structure, inhibiting the toxin from binding to the requisite molecules on the host cell surface.

    The ADP-ribosylation of eukaryotic elongation factor-2 by the bacterial toxin inhibits the ribosomal translation of protein, and the trastuzumab-PE24 conjugate exhibited the cytotoxicity toward Her2-overexpressing cell lines. The PEDIP reaction can also be applied for many other types of cargo with slight modifications of the method.
