    The data suggest a molecular circuit involving the MALAT1-mascRNA system, interactions between MALAT1 and NEAT1, and key immune effector molecules, cumulatively impacting upon the development of atherosclerosis. It appears reasonable to look for therapeutic targets in this circuit and to screen for anomalies in the NEAT1-MALAT1 region in humans, too, as possible novel disease risk factors.

    Journal:  Sorafenib attenuated the function of natural killer cells infiltrated in HCC through inhibiting ERK1/2. (Pubmed Central) -  Mar 4, 2020   
    This phenotype made us study the terminal function of NK cells, revealing that Sorafenib could decrease the production of effector molecules and cytokines, such as perforin, granzyme B, TNF-α, IFN-γ, etc. Besides, p-ERK1/2 in NK cells was inhibited after treatment with Sorafenib, and a similar tendency of NK cells could be achieved using ERK1/2 inhibitor. Collectively, our data suggested that Sorafenib functioned as a critical inhibitor that controlled the number and function of NK cells through inhibiting ERK1/2.

    We prospectively enrolled Ugandans presenting with first episode cryptococcal meningitis from March 2015 to May 2017, as part of a larger study focused on drug treatment with amphotericin + fluconazole +/- sertraline to improve neurological outcomes...These findings demonstrate a crucial role for cytotoxic cell populations and naive T-cell stimulation in human cryptococcal outcomes. Further research efforts should include characterizing the role and activating stimuli of cytotoxic cells in the clearance of Cryptococcus as well as T-cell function in activation of the adaptive immune response in humans with cryptococcosis.

    We demonstrate that airway-derived HIV-specific CD8+T cells poorly express cytolytic molecules (Granzyme B and Perforin) and possess markers consistent with high immune regulation (PD1 and Eomes). The poor cytolytic potential and highly regulated phenotype of airway HIV-specific CD8+T cells could promote the persistence of HIV-infected cells in the airway in individuals on long term ART.

Therefore, our results demonstrate that WASP may be involved in regulating cytotoxic function and apoptosis in CD8 T cells following MCMV infection, which is supported by the distribution and memory compartment of MCMV-specific T cells in MCMV infected WAS mice.

    We conclude that nano2/4 enhances the systemic T cell immunity that improves the host's ability to control chronic parasite persistence and Chagas cardiomyopathy. While overall inflammation may be increased in HPV+ patients with recurrent disease, based on the current study, the tumor associated inflammation may be contributing to an enhanced immunosuppressive environment.

    Mechanistically, we demonstrated that simultaneous targeting of Tim-3, PD-1, and Lag-3 cooperatively increased the levels of granzyme B and tumor-specific cytolytic activities of CD8 TIL. Our data indicate that multiple checkpoint molecules are coordinately upregulated to inhibit the function of hyperactivated T cells in the TME and requirement for the simultaneous blockade of PD-1, Tim-3 and Lag3 for cancer treatment.

    Colocalization of GSDMD with granzyme B was observed in the proximity of immune synapses, and GSDMD deficiency reduced the cytolytic capacity of CD8 T cells. Overall, our study highlights a function, to our knowledge previously unknown, for GSDMD in CTLs and demonstrated that GSDMD is required for an optimal CTL response to cancer cells.

    Epitope mapping confirmed that vaccination elicited responses against predicted T-cell epitopes, but also allowed to identify a set of 8 new HIV-1 HLA-DR-restricted CD4+ T-cell epitopes. These results may help to better design future DC therapeutic vaccines and underscore the role of vaccine-elicited CD4+ T-cell responses to achieve control of HIV replication.

    While T-bet ablation restricted interferon-γ (IFN-γ) production, loss of Blimp-1 prevented GzmB expression in response to IL-2, suggesting two independent programs required for polyfunctionality of tumor-reactive CD4 T cells. Our findings underscore the role of Treg cells, IL-2, and Blimp-1 in controlling the differentiation of cytotoxic CD4 T cells and offer a pathway to enhancement of anti-tumor activity through their manipulation.

PD-1 pathway blockade increased the numbers of CD101Tim3 CD8 T cells, suggesting that these newly generated transitional cells play a critical role in PD-1-based immunotherapy.

PD-1 pathway blockade increased the numbers of CD101Tim3 CD8 T cells, suggesting that these newly generated transitional cells play a critical role in PD-1-based immunotherapy. These results suggest that C. albicans may exert a regulatory role in the defense response of oral mucosal epithelial cells by altering secretory levels of defense effector molecules.
