- |||||||||| Review, Journal: Update in Molecular Aspects and Diagnosis of Autoimmune Gastritis. (Pubmed Central) - Jul 28, 2023
The current focus lies on individuals demonstrating atypical presentations of the disease, including those diagnosed in childhood, those yielding negative results for autoantibodies, and those lacking the typical endoscopic characteristics of mucosal atrophy. Here, we discuss the recent developments in this field, focusing on genetic predisposition, epigenetic modifications, lymphocytes, cytokines, oxidative stress, infectious agents, proteins, microRNAs, autoantibodies, serum pepsinogen, gastrin, esophagogastroduodenoscopy and microscopic findings, and the risk of gastric neoplasm.

Furthermore, the semi-supervised model could also successfully learn and predict sample types, achieving a 0.986 AUC score on the EV dataset, even when 10% samples in the five GEO datasets were labeled. In conclusion, three hub genes (ATP4A, ATP4B, and ESRRG) closely related to gastric cancer were mined, based on which the ML diagnostic model of gastric cancer was conducted.
- |||||||||| Gastric Transcriptional Alterations in Response to Helicobacter Pylori Infection (Exhibit and Poster Hall) - Jun 4, 2022 - Abstract #ASMMicrobe2022ASM_Microbe_2528;
The reduced transcript levels of these markers were correlated with a significant loss of chief and parietal cells detected by histologic analysis. Collectively, these results suggest that H. pylori causes a wide range of alterations in the gastric transcriptome, including changes in multiple gastric cell lineages, and provide a new approach for identifying biomarkers of gastric atrophy and premalignant changes.

PPI ADMINISTRATION INDUCED COLONIC TIGHT JUNCTION BARRIER DYSFUNCTION IS ASSOCIATED WITH INCREASE IN PH, MLCK AND ENETROCOCCUS FECALIS. (24 - San Diego Convention Center) - Apr 25, 2022 - Abstract #DDW2022DDW_2554; Methods : 8 to 12-week-old male C57BL/6 mice were divided into two groups (controls and PPI treatment, Omeprazole, 20mg/kg B.W. for 30 days)...Both PPI administration in WT mice and Atp4a deletion induced an increase in colonic luminal pH. Conclusion : Our Results suggest that the gastric H+K+ATPase inhibition by PPI or genetic KO increases mouse colonic TJ barrier permeability, associated with altered pH, MLCK activation and overgrowth of a pathobiont EF.
- |||||||||| Journal: Bioinformatic analysis of potential hub genes in gastric adenocarcinoma. (Pubmed Central) - Apr 2, 2022
The Kaplan-Meier online plotter tool showed associations of all hub genes except for CA9 with prognosis in gastric adenocarcinoma patients; CXCL8 and ALDH1A1 were positively correlated with survival, and the other genes were negatively correlated with survival. These 10 hub genes may be involved in important processes in gastric adenocarcinoma development, providing new directions for research to clarify the role of these genes and offer insight for improved treatment.

PN also increased the expression of ATP4a, cytochrome P450 family 21 subfamily a member 2, and hydroxy-delta-5-steroid dehydrogenase, 3 beta and steroid delta-isomerase 2 proteins in the mitochondria, and decreased the content of mtROS in cmECs. PN alleviated the pH in the colonic lumen and hypoxia-induced glycolysis in cmECs by reducing the hypoxia-induced glycolysis caused by the downregulation of ATP4a protein, thereby promoting the repair of colonic mucosal injuries in colitis.

Journal: Maternal antibiotic treatment affects offspring gastric sensing for umami taste and ghrelin regulation in the pig. (Pubmed Central) - Mar 19, 2021 This study systematically explored the key genes and pathways involved in PGC and AGC, providing insights into therapeutic individualized management. Data show that sensing for umami taste and ghrelin regulation can be affected by maternal environment, but the development of acid secretion, orexigenic signaling and taste perception in the stomach are mostly developmentally controlled.

Chronic achlorhydria and hypergastrinemia in aged Atp4a-/- mice produced progressive hyperplasia and mucolytic and intestinal metaplasia and activated the Warburg effect via PI3K/AKT/mTOR signaling. BGN expression levels appear to be an independent prognostic factor for predicting the survival times of GC patients.
- |||||||||| Clinical, Journal: H+/K+ATPase Expression in the Larynx of Laryngopharyngeal Reflux and Laryngeal Cancer Patients. (Pubmed Central) - Jan 23, 2021
Gastric proton pump subunits are expressed in the larynx of LPR and LSCC patients. Mitochondrial damage and changes in gene expression observed in cells expressing the full proton pump, absent in those expressing a single subunit, suggest that acid secretion by functional proton pumps expressed in upper airway mucosa may elicit local cell and molecular changes associated with inflammation and cancer.

Review, Journal: Proton Pump Inhibitor Use, Hypergastrinemia, and Gastric Carcinoids-What Is the Relationship? (Pubmed Central) - Oct 10, 2020 Overall, the risk of developing a g-NET appears greatest in patients who are more than 10 years on drug and on higher doses: those affected by chronic H. pylori gastritis and/or consequent gastric atrophy may also be at increased risk. While the overall risk of g-NETs induced by PPI therapy is undoubtedly low, it is real: this necessitates caution in using PPI therapy for long periods of time, particularly when initiated in young subjects.

IN HUMAN ESOPHAGEAL SMOOTH MUSCLE CELLS, IL-4 STIMULATES EOTAXIN-3 SECRETION THAT IS BLOCKED ONLY MINIMALLY BY A PPI OR A P-CAB. () - Jan 28, 2020 - Abstract #DDW2020DDW_2978; Unlike esophageal epithelial cells, however, Th2 cytokine-stimulated eotaxin-3 secretion by ESM cells is decreased only minimally, albeit significantly, by omeprazole or SCH-28080. These findings suggest that PPIs and P-CABs might have limited effect on eosinophils infiltrating the esophageal muscularis propria in EoE patients, which could explain why their esophageal motility abnormalities persist despite PPI treatment that eliminates mucosal eosinophilia.
