    Journal:  The Impact of Alpha-1-Adrenergic Receptor Antagonists on the Progression of Parkinson's Disease. (Pubmed Central) -  Mar 18, 2024
    Patients prescribed a PGK1-activating AARA experienced fewer PD-related outcome events than patients prescribed tamsulosin. These results may indicate a role for terazosin and other PGK1 activators in slowing disease progression of PD; however, randomized controlled trials are needed.
  • ||||||||||  Innate Immune Effects of ?1-adrenoreceptor Antagonism in Sarcoidosis (TDP13) -  Mar 17, 2024 - Abstract #ATS2024ATS_9072;
    This pilot data provide proof of principle of a connection between NA, ?1-ARAs, and disease outcomes in sarcoidosis and suggest a possible link to mtDNA-mediated PRR activation. Further study of this neuroimmune mechanism may lead to novel therapies.
  • ||||||||||  Journal:  Blockade of sympathetic ganglia improves vascular dysfunction in septic shock. (Pubmed Central) -  Mar 8, 2024
    Both blockers normalized the in vivo response to vasoconstrictors, and reduced plasma IL-1 and NOx levels and NOS-2 expression in the aorta.Blockade of ganglionic sympathetic transmission reduced the vascular dysfunction in experimental sepsis. This beneficial effect seems to be, at least in part, due to the preservation of ?1 adrenergic receptor density and to reduced NOS-2 expression and may lead to adjuvant ways to treat human sepsis.
  • ||||||||||  Journal, Head-to-Head:  Effect of different alpha-receptor antagonists on metabolic parameters: a head-to-head comparison. (Pubmed Central) -  Mar 8, 2024
    This beneficial effect seems to be, at least in part, due to the preservation of ?1 adrenergic receptor density and to reduced NOS-2 expression and may lead to adjuvant ways to treat human sepsis. Although these results do not support previous findings that tamsulosin has adverse effects on metabolic parameters, they suggest that doxazosin treatment may be a reason for preference in patients with BPH or LUTS accompanied by metabolic disorder.

  • ||||||||||  Journal:  Broad-spectrum ginsentides are principal bioactives in unraveling the cure-all effects of ginseng. (Pubmed Central) -  Feb 7, 2024
    Orally administered ginsentides were effective in animal models to reduce ADP-induced platelet aggregation, to prevent collagen and adrenaline-induced pulmonary thrombosis as well as anti-stress behavior of tail suspension and forced swimming tests in mice. Together, these results strongly suggest that ginsentides are the principal panacea compounds of ginsengs because of their ability to target multiple extra- and intra-cellular proteins to reverse stress-induced damages.
  • ||||||||||  Journal:  Allergic manifestations in women with silicone breast implants. (Pubmed Central) -  Dec 28, 2023
    Allergic reactions associated with SBI are not uncommon. Imbalance of circulating levels of IgE autoantibodies against GPCRs of the autonomic nervous system might play a role not only in allergic reactions, but also in other enigmatic aspects of SBII such as autoimmune dysautonomia.

    Preclinical, Journal:  Localization and neurochemical identity of alpha1-adrenergic receptor-containing elements in the mouse locus coeruleus. (Pubmed Central) -  Nov 20, 2023
    Another study showed that during novelty-induced behavioral activation, the ?1AR antagonist prazosin reduced c-fos expression in brain regions known to contain motoric ?1ARs, except for the LC, where c-fos expression was enhanced...Double labeling immunohistochemistry found the ?1AR expressed in GFAP-labeled astrocytes, in both GABAergic and glutamatergic axon terminals, and in a portion of the ?1AR dendrites, colocalized with tyrosine hydroxylase (TH, a marker for noradrenergic neurons). This study sheds light on the neuroanatomical framework underlying the effects of NE and pharmaceuticals acting directly or indirectly on ?1ARs in the LC.
  • ||||||||||  Adrenergic gating of a rapid disconnection of fast-spiking cell circuits in the visual cortex (WCC Halls A-C) -  Nov 3, 2023 - Abstract #Neuroscience2023NEUROSCIENCE_11335;
    Also, ?1-adrenergic receptor activation, in-vivo and in slices, in conjunction with pre- and post-synaptic activation, enable "all-or-none" elimination of excitatory input onto PV cells. We propose that the ?1-adrenergic dependent Pyr?PV disconnection resembles the MD-evoked synapse elimination and mechanistically works according to the pull-push rule.
  • ||||||||||  Inhibition of noradrenaline-dependent synaptic transmission in the dorsal raphe nucleus by alpha2-adrenergic receptors (WCC Halls A-C) -  Nov 3, 2023 - Abstract #Neuroscience2023NEUROSCIENCE_11291;
    Taken together, these results demonstrate that alpha2-AR are presynaptic receptors located on noradrenergic projections from the LC to the DR. Activation of alpha2-AR in the DR is expected to reduce the excitability of serotonin neurons by 1) limiting noradrenaline release thereby decreasing alpha1-AR-mediated excitation and 2) direct hyperpolarization via activation of GIRK channels.

    Activation of alpha2-AR in the DR is expected to reduce the excitability of serotonin neurons by 1) limiting noradrenaline release thereby decreasing alpha1-AR-mediated excitation and 2) direct hyperpolarization via activation of GIRK channels. The stroma cells and the cell surrounding functional structures of the ovary such as the follicles, the corpus luteum, and cysts express ?1-adrenergic and

    Sensitivity of arterial baroreflex as a prognostic marker in determining the properties of vegetotropic compounds (WCC Halls A-C) -  Nov 3, 2023 - Abstract #Neuroscience2023NEUROSCIENCE_3858;
    SBR against the background of fobufol (0.5 mg/kg) was determined by extrapolation of the experimental data of the remaining 8 groups of animals. Using the same method, a model for predicting the membrane-stabilizing properties of ?-B was developed.It is proposed to take into account differences in the impact effect of known antihypertensive drugs on hypertensive and chronotropic reactions in the Oxford ABR testing in predictive models when studying the properties of new vegetotropic drugs.

    Preclinical, Journal:  Methyleugenol Has an Antidepressant Effect in a Neuroendocrine Model: In Silico and In Vivo Evidence. (Pubmed Central) -  Oct 28, 2023
    This study investigated the antidepressant activity of the phenylpropanoid methyleugenol (ME) in adult female mice exposed to a stress model induced by dexamethasone...After 165 and 180 min, they were treated with ME (25, 50 and 100 mg/kg intraperitoneally) or imipramine (10 mg/kg intraperitoneally) after 45 min and 30 min, respectively; they were then submitted to tests which were filmed...In the in vivo evaluation of the pathways of action, the antidepressant potential of ME (50 mg/kg) was reversed by SCH23390 (4 mg/kg intraperitoneally) dopaminergic D1 receptor, Prazosin (1 mg/kg intraperitoneally) ?1 adrenergic receptor, and PCPA (4 mg/kg intraperitoneally) tryptophan hydroxylase inhibitor. Our findings indicate that ME did not alter with the locomotor activity of the animals and shows antidepressant activity in female mice with the participation of the D1, ?1 and serotonergic systems.

    Preclinical, Journal:  Genetic background influences arterial vasomotor function in male and female mice. (Pubmed Central) -  Oct 3, 2023
    Vasorelaxation responses were assessed by examining relaxation to an endothelium-dependent vasodilator acetylcholine (ACh, 10 -10 ?M) and an endothelium-independent vasodilator sodium nitroprusside (SNP, 10 -10 ?M)...Results indicated some concentration-response heterogeneity in response to ACh and SNP between vessels from females and males, but no significant differences in responses to PE. Collectively, these findings indicate that vasomotor responses vary by genetic background, sex, and artery type.
  • ||||||||||  Preclinical, Journal:  In (Pubmed Central) -  Sep 7, 2023
    Estrus-specific output reduction coincides with prolonged aVCS activation of inhibitory interneurons. Accordingly, in estrus the AOB microcircuit sets the stage for coital stimulation to inhibit the output neurons, possibly via high frequency bursting-dependent enhancement of reciprocal synapse efficacy between inter- and output neurons.

    After fasting overnight, initial fasting blood glucose was measured, and mice were anesthetized with isoflurane for measurement of blood perfusion via laser speckle contrast analysis using a PeriMed PeriCam PSI NR system...In the CyB5R3 strain, mice housed on corncob bedding displayed a significant reduction in phenylephrine-mediated change in perfusion. In the Hba1 strain, phenylephrine-induced change in perfusion was not different in the corncob group.
  • ||||||||||  Allergic contact dermatitis resulting from phenylephrine in glaucoma eye drops () -  Jul 3, 2023 - Abstract #WCD2023WCD_5112;
    Alongside atropine, cyclopentolate, and tropicamide, it is widely used as a mydriatic eye drop, although it is the only one of the four medicines to not cause antimuscarinic effects...Patch testing was also negative with proparacaine hydrochloride, phenylephrine hydrochloride, pilocarpine hydrochloride, tropicamide, timolol maleate, and latanoprost...In patients suspected of having phenylephrine-induced ACD, it is important to perform patch testing to determine whether the causative agent was phenylephrine itself or a preservative agent. It is also important to test other ophthalmic solutions so that other options can be recommended to ophthalmology for future eye examinations
  • ||||||||||  Radial astrocytes in the developing retinotectal system are activated by norepinephrine and promote visually-driven escape behaviour (Montreal Bonaventure Hotel (CAN meeting headquarters)) -  Jun 10, 2023 - Abstract #CANACN2023CAN_ACN_697;
    Complementary electrophysiological recordings from postsynaptic tectal neurons suggest that norepinephrine reduces presynaptic input in the tectum. Additionally, we show that the targeted chemogenetic activation of tectal radial astrocytes in freely swimming animals nearly doubles loom-evoked escape rates suggesting that radial astrocytes may play an important role in mediating the probability of initiating escape behaviour during predation events.
