Journal: Down-regulated microRNA-223 or elevated ZIC1 inhibits the development of pancreatic cancer via inhibiting PI3K/Akt/mTOR signaling pathway activation. (Pubmed Central) - Dec 16, 2021 Screened cells were transfected with miR-223- or ZIC1-related oligonucleotides or plasmids, or AZD8055, the dual inhibitor of the PI3K/Akt/mTOR signaling pathway to test the functions of miR-223, ZIC1 or PI3K/Akt/mTOR signaling pathway in the biological functions of PC cells...MiR-223 up-regulation or ZIC1 down-regulation induced opposite results on PC cells. This study highlights that down-regulated miR-223 or elevated ZIC1 inhibits the development of PC via restricting activation of the PI3K/Akt/mTOR pathway, which has important meanings for exploring the mechanism of PC.

Journal: Oleate ameliorates palmitate-induced impairment of differentiative capacity in C2C12 myoblast cells. (Pubmed Central) - Dec 16, 2021 The positive effect of OA on myoblast differentiative capacity disappeared if PI3K inhibitor LY294002 was added. In conclusion, our study showed that PA could destroy differentiative capacity of C2C12 myoblasts by affecting the expression of Pax7, MyoD and MyoG, and OA could improve this impairment through PI3K /Akt signaling pathway.

Journal, PD(L)-1 Biomarker, IO biomarker: Morphine-3-glucuronide upregulates PD-L1 expression via TLR4 and promotes the immune escape of non-small cell lung cancer. (Pubmed Central) - Dec 16, 2021 Repeated M3G intraperitoneal injections promoted LLC tumor growth and lung metastasis through the upregulation of tumor expressed PD-L1 and the reduction of CTL in the tumor microenvironment. M3G specifically activated TLR4 in NSCLC cells and upregulated PD-L1 expression through the PI3K signaling pathway, thereby inhibiting CTL cytotoxicity and finally promoting tumor immune escape.

Journal: FK228 sensitizes radioresistant small cell lung cancer cells to radiation. (Pubmed Central) - Dec 16, 2021 The study uncovered that FK228 sensitized human radioresistant SCLC cells to radiation mainly through induction of chromatin decondensation and suppression of DNA damage signaling and repair. Our study provides a rational basis for a further clinical study to test the potential of FK228 as a radiosensitizing agent to increase the radiation-induced tumor cell kill in LS-SCLC patients.

Journal: Kinome-wide analysis of the effect of statins in colorectal cancer. (Pubmed Central) - Dec 16, 2021 Taken together, GM1 may be a potential preventive treatment for the neurotoxicity caused by continuous exposure to ketamine. Statins induce BMP-specific activation of PTEN and inhibition of PI3K/Akt/mTOR signalling in CRC.

Clinical, Journal, IO biomarker: Synergistic efficacy of dual PI3K-d/g inhibitor Duvelisib with Bcl2 inhibitor Venetoclax in Richter's Syndrome PDX models. (Pubmed Central) - Dec 16, 2021 P1 This work provides, for the first time, a proof-of-concept of the efficacy of dual targeting of PI3K-d/g and Bcl-2 in RS, opening for a Duv/Ven combination for these patients. Clinical studies in aggressive lymphomas, including RS, are underway (NCT03892044).
- |||||||||| Journal: PI3K, mTOR AND GSK3 MODULATE CYTOKINES' PRODUCTION IN PERIPHERAL LEUKOCYTE IN TEMPORAL LOBE EPILEPSY. (Pubmed Central) - Dec 16, 2021
Herein we demonstrated that the production of cytokines by immune cells from patients with TLE differs from non-epileptic patients. This differential regulation may be associated with the altered activity and responsiveness of intracellular molecules, such as PI3K, mTOR and GSK-3, which, in turn, might contribute to the inflammatory state that exists in epilepsy and its pathogenesis.
- |||||||||| Review, Journal: Cross-talk between non-coding RNAs and PI3K/AKT/mTOR pathway in colorectal cancer. (Pubmed Central) - Dec 16, 2021
PI3K/AKT/mTOR pathway contributes to many cellular processes, and dysregulation of this pathway frequently occurs in cancers. In this review, the authors have mostly focused on the significant non-coding RNAs regulators of the PI3K/AKT/mTOR pathway and their contribution to the development or inhibition of CRC and their potential as diagnostic or therapeutic targets in CRC treatment.
- |||||||||| Preclinical, Journal: Angelica gigas root ameliorates ischaemic stroke-induced brain injury in mice by activating the PI3K/AKT/mTOR and MAPK pathways. (Pubmed Central) - Dec 16, 2021
The proteins related to cell growth, differentiation, and death were upregulated by this treatment, and the major recovery mechanisms appeared to involve the attenuation of the mitochondrial function of Bcl-2/Bax and activation of the PI3K/AKT/mTOR and MAPK signalling pathways in ischaemic neurons. This study provides evidence supporting the use of A. gigas root against ischaemic stroke and suggests a novel developmental starting point for the treatment of ischaemic stroke.
