Preclinical, Review, Journal: Antifibrotic action of Yifei Sanjie formula enhanced autophagy via PI3K-AKT-mTOR signaling pathway in mouse model of pulmonary fibrosis. (Pubmed Central) - Feb 14, 2020 In addition, YFSJF-induced autophagy is mediated by the PI3K-AKT-mTOR pathway, and knockdown of PI3K by siRNA up-regulated the expression of autophagy-related genes and down-regulated the expression of collagen in human lung fibroblasts (HLF). Our findings provide a detailed understanding that YFSJF-antifibrotic effects are mainly mediated by triggering autophagy, and suppressing phosphorylation of the PI3K-AKT-mTOR pathway is required for YFSJF-curative effect.
- |||||||||| Review, Journal: Interplay Among PI3K/AKT, PTEN/FOXO and AR Signaling in Prostate Cancer. (Pubmed Central) - Feb 14, 2020
Indeed, the cross talk between PI3K/AKT and other pathways, such as AR, WNT, and ERK signaling pathways is known to play essential roles in disease progression and drug resistance in prostate cancer. Therefore, co-targeting the PI3K/AKT signaling pathway and its cooperating pathways may be critical for improving the anti-cancer efficacy of PI3K/AKT inhibitors in the clinic.

Preclinical, Journal: Catalpol induces cell activity to promote axonal regeneration via the PI3K/AKT/mTOR pathway in vivo and in vitro stroke model. (Pubmed Central) - Feb 13, 2020 Moreover, rapamycin (Rapa) was used to inhibit the mTOR pathway to observe the catalpol mechanism on neuronal cell activity to promote axonal growth, and the proteins related with PI3K/AKT/mTOR pathway were detected by Western blot assay...Catalpol can also reversed proteins reduced by Rapa related with PI3K/AKT/mTOR pathway. These results suggested that catalpol might contribute to internal neuronal cell activity and axonal regeneration by regulating PI3K/AKT/mTOR pathway.

Pretargeted delivery of PI3K/mTOR small-molecule inhibitor-loaded nanoparticles for treatment of non-Hodgkin lymphoma (Freedom Ballroom E/F, Philadelphia 201 Hotel) - Feb 13, 2020 - Abstract #ACSSp2020ACS_Sp_17389; Comprehensive in vivo studies using Namalwa and Raji mouse models confirm that the dual Ab pretargeted system effectively inhibits tumors growth, improves survival, and reduces the systemic side effects of BEZ235. Our data demonstrate that a novel NP-enabled pretargeted system consolidates the use of kinase inhibition with Ab-directed targeted therapy and improves the therapeutic window of small molecule inhibitors.

From Bench to Bedside and Back Again: The Journey to the Discovery of GDC-0077 (Grand Ballroom B, Pennsylvania Convention Center) - Feb 13, 2020 - Abstract #ACSSp2020ACS_Sp_11428; While many of these inhibitors have struggled to balance clinical responses with adequate safety profiles, several conclusions have been drawn from the clinical and preclinical data to guide the discovery of improved next generation molecules. Genentech's history and learnings from research with PI3K inhibitors will be summarized and focus particularly on the data and observations that contributed to the design and discovery of GDC-0077, a highly selective PI3Ka inhibitor that selectively degrades p110a and is currently in clinical trials.

Protective effect of piceatannol against oxidative stress-induced apoptosis in ARPE-19 cells and its mechanism (102B, Pennsylvania Convention Center) - Feb 13, 2020 - Abstract #ACSSp2020ACS_Sp_10101; LY294002 (PI3K inhibitor) dramatically blocked piceatannol-mediated increasing of Nrf2 nuclear translocation, HO-1 expression, and cytoprotective activity, indicating the involvement of PI3K/Akt pathway in the cytoprotective effect of piceatannol. The results from this suggest the potential of piceatannol in reducing the risk of age-related macular degeneration.

- |||||||||| Preclinical, Journal: Nitric Oxide Synthase Is Involved in Follicular Development via the PI3K/AKT/FoxO3a Pathway in Neonatal and Immature Rats. (Pubmed Central) - Feb 10, 2020
Meanwhile, the ovarian expression in the L-Arg group in terms of p-AKT, p-FoxO3a, and LC3-II on PD19 were significantly (p < 0.05) upregulated, while the expressions of PTEN and cleaved Caspase-3 were (p < 0.05) downregulated as a result of NOS/NO generation, respectively. Therefore, the results suggest that NOS is possibly involved in the maturation of follicular development to puberty via the PI3K/ AKT/FoxO3a pathway, through follicular autophagia and apoptosis mechanisms.

Journal: MicroRNA‑212 facilitates the motility and invasiveness of esophageal squamous carcinoma cells. (Pubmed Central) - Feb 8, 2020 These data suggest that miR‑212 activates multiple signaling cascades and facilitates ESCC cell motility and invasion by promoting the epithelial‑mesenchymal transition and degrading the ECM. Berberine may be a potential therapeutic agent against metastasis in patients with ESCC, who express high levels of miR‑212.

Such combined approaches could potentially be used as neoadjuvant therapies, limiting the development of resistance to treatments or sensitizing cells to other therapeutics. To determine which drugs should be combined with PIM inhibitors for each patient, it will be key to develop companion diagnostics that predict response to each co-targeted option, hopefully providing a personalized medicine pathway for subsets of prostate cancer patients in the future.

PI3Kγ dysfunction led to degeneration of the intestinal microbial community and might alter the synthesis of some antibiotics, bile acids, and thiamine. The usage of PI3Kγ inhibitors for inflammation and cardiovascular diseases might lead to knock-on effect on our organism through intestinal microbiota.

Journal: Orthosilicic Acid Accelerates Bone Formation in Human Osteoblast-Like Cells Through the PI3K-Akt-mTOR Pathway. (Pubmed Central) - Feb 7, 2020 However, with the addition of PI3K-Akt-mTOR pathway-specific inhibitor LY294002, the expression of PI3K, P-Akt, P-mTOR, RUNX2, COL1, ALP, OCN, and P1NP decreased. The results indicated that the PI3K-Akt-mTOR pathway played a positive regulatory role in the process of orthosilicic acid-mediated osteogenesis in vitro.
- |||||||||| Journal: Inhibition of IGF-1-PI3K-Akt-mTORC2 in lipid rafts increases neuronal vulnerability in a genetic lysosomal glycosphingolipidosis. (Pubmed Central) - Feb 7, 2020
Our results show that LR disruption by psychosine de-sensitizes cells to extracellular growth factors by inhibiting signal transmission from the plasma membrane to intracellular compartments. This mechanism serves also as a mechanistic model to understand how alterations of the membrane architecture by the progressive accumulation of lipids undermines cell function, with potential implications in other genetic sphingolipidoses and adult neurodegenerative conditions.

Journal: Effective Treatment of Metastatic Melanoma by Combining MAPK and PI3K Signaling Pathway Inhibitors. (Pubmed Central) - Feb 7, 2020 Combined treatment also more effectively increased apoptosis, and inhibited tumor growth in vivo. This suggests a clinical potential of combined treatment to overcome ceased treatment activity which is often seen after monotherapies, and strongly encourages the evaluation of the treatment strategy on melanoma patients with brain metastases.
