In summary, overexpression of LMP1 and LMP2A inhibits COX-2, which is mediated by a decrease of TRAF2, and p-ERK is involved in the inhibition of COX-2 by LMP1 in gastric cancer.

(Pubmed Central) - Jun 4, 2020 The venoarteriolar reflex was assessed at the following forearm skin sites: Experiment 1 (n = 11): 1) lactated Ringer solution (Control), 2) 10 mM N-nitro--arginine (NOS inhibitor), 3) 10 mM ketorolac (COX inhibitor), and 4) combined NOS + COX inhibition; Experiment 2 (n = 15): 1) lactated Ringer solution (Control), 2) 50 mM tetraethylammonium (K channel blocker), 3) 5 mM glybenclamide (K channel blocker), and 4) 10 mM 4-aminopyridine (K channel blocker)...We showed that K and K channels modulate the venoarteriolar reflex with minimum roles of NOS, COX, and K channels in human non-glabrous forearm skin in vivo. Thus, cutaneous venoarteriolar reflex changes could reflect altered K channel function.

The lipidomics results suggest two potential treatment targets: blocking the formation of prostaglandins by inhibiting COX and stabilizing the anti-inflammatory lipid mediators containing epoxides by inhibiting the enzyme soluble epoxide hydrolase (sEH).

However, more studies are needed to comprehensively understand antiplatelet effects on the PRP secretome.

Indomethacin did not alter DCVC or DCVC/DSO to hypoxia.These findings indicate: 1) COX contributes, in a largely uniform fashion, to cerebrovascular tone during normoxia, and 2) COX is not obligatory for hypoxic vasodilation in any regions supplied by large extracranial or intracranial arteries.

In summary, these novel pyrazolo[3,4-d]pyrimidine analogues showed interesting anti-inflammatory activity and could act as a starting point for future drugs.
