    Journal:  Tocolytic effect of the monoterpenic phenol isomer, carvacrol, on the pregnant rat uterus. (Pubmed Central) -  Nov 26, 2020
    The aim of this study was to evaluate the in vitro tocolytic effect of CAR on the contractility of isolated pregnant rat uterus in the presence of a calcium channel antagonist (nifedipine) and a cyclooxygenase inhibitor (indomethacin)...Therefore, the tocolytic effect induced by CAR was associated with the blockade of the calcium channels in the pregnant rat uterus. This property placed CAR as a potentially safe and effective adjuvant agent in cases of preterm labor, an area of pharmacological treatment that requires urgent improvement.
  • ||||||||||  Preclinical, Journal:  Nicotinic acid promotes sleep through prostaglandin synthesis in mice. (Pubmed Central) -  Nov 14, 2020
    Systemic administration of monomethyl fumarate, another agonist of the nicotinic acid receptor GPR109A, fully recapitulated the somnogenic and thermoregulatory effects of nicotinic acid suggesting that they are mediated by the GPR109A receptor. The cyclooxygenase inhibitor indomethacin completely abolished the effects of nicotinic acid indicating that prostaglandins play a key role in mediating the sleep and thermoregulatory responses of nicotinic acid.

    Preclinical, Journal:  Gastroprotective activity of hydroalcoholic extract of the leaves of Urera baccifera in rats. (Pubmed Central) -  Nov 7, 2020
    Together, the results confirmed that the gastroprotection elicited by HEU was due reduction in oxidative damage, neutrophil migration, and peptic activity. This work validates the popular use of U. baccifera to treat gastric disorders and supports important future research for the identification of gastroprotective molecules from this species.

    Preclinical, Journal:  Cyclooxygenase activity in bradykinin-induced dermal extravasation. A study in mice and humans. (Pubmed Central) -  Nov 7, 2020
    These data suggest that COX activity contributes to bradykinin-induced dermal extravasation in mice and humans. In addition, our findings may open new treatment options and point to a potential activity of drugs interfering with the release of the COX substrate arachidonic acid, e.g. glucocorticoids.

    Journal:  Topical MTII Therapy Suppresses Melanoma Through PTEN Upregulation and Cyclooxygenase II Inhibition. (Pubmed Central) -  Nov 3, 2020
    Melanotan II (MTII), a synthetic analogue of the alpha-melanocyte stimulating hormone (α-MSH), has been applied for skin tanning in humans...Together, these results indicate that MTII elicits PTEN upregulation via MC1R, thereby suppressing melanoma progression through downregulating COX-2/PGE2 signaling. Hence, topical MTII therapy may facilitate a novel therapeutic strategy against melanoma.

    Journal:  Short-term application of ibuprofen before ovulation. (Pubmed Central) -  Nov 1, 2020
    Logistic regression showed no significant association of ibuprofen intake, LH- level or reason for infertility on embryo quality. Based on our results, we suggest that, particularly within natural cycle IVF, ibuprofen does no harm around ovulation as analgesic treatment.

    Journal:  Sex differences in vascular reactivity of coronary artery bypass graft conduits. (Pubmed Central) -  Oct 28, 2020
    (1) Concentration response curves for PE and ET-1 were generated in an intact IMA and SV and endothelium denuded IMA segments, (2) in the presence of the nitric oxide synthase inhibitor (L-NAME) or the cyclooxygenase inhibitor (indomethacin) in an endothelium-intact IMA and (3) the activity state (abundance and phosphorylation) of the α-adrenergic receptor was investigated using Phos-tag™ western blot analysis...Female IMA and SV graft conduits are hypersensitive to α-adrenergic stimuli. This endothelial cyclooxygenase pathway-mediated hypersensitivity may produce excessive IMA and SV graft constriction in females administered catecholamines and could contribute to their poorer CABG outcomes.

    Journal:  Large health benefits can be achieved by better treatment of gout (Pubmed Central) -  Oct 27, 2020
    Allopurinol is the first line choice of ULT and should be started with low dose, which is increased until the treatment target is reached...NSAID/cox-inhibitors, colchicine and glucocorticoids are all valid short-term treatments of gout attacks...Recent RCTs of ULT treatment have demonstrated the importance of thorough and adequate information to the patient and regular follow-up until treatment targets are reached. Such a strategy improve both compliance and outcomes of ULT treatment.

  • ||||||||||  Journal:  NF-κB "decoy" inhibits COX-2 expression in epileptic rat brain. (Pubmed Central) -  Oct 24, 2020
    In the experimental group compared to the control group, the nuclear factor-kappa B expression in the hippocampal dentate gyrus, cerebral cortex, piriform cortex, and other brain regions was significantly lower (P < 0.01), with the accompanying decrease in cyclooxygenase-2 protein and mRNA expression (P < 0.01) in the hippocampus. In conclusion, κB-decoy can inhibit nuclear factor kappa B activation in epileptic rat brain and cyclooxygenase-2 overexpression.
  • ||||||||||  Journal:  Molecular Imaging of Inflammation in Osteoarthritis Using a Water-Soluble Fluorocoxib. (Pubmed Central) -  Oct 20, 2020
    COX-2-selectivity was verified either by blocking the enzyme's active site with celecoxib or by molecular imaging with nontargeted 5-carboxy-X-rhodamine dye. These data indicate that fluorocoxib D is an ideal candidate for early detection of inflammatory or neoplastic lesions expressing elevated levels of COX-2.
  • ||||||||||  [VIRTUAL] ANGINA AND ATRIAL FIBRILLATION IN MELOXICAM USE () -  Oct 12, 2020 - Abstract #CHEST2020CHEST_976;
    Given the negative work up and resolution following discontinuation of the agent, the patient’s angina and atrial fibrillation were likely associated with meloxicam use. Further prospective studies should be performed to assess association of non selective COX inhibitors in development of acute cardiac events and long term cardiovascular mortality.

    We constructed a novel supramolecular hydrogel by carrying out a coassembly of cisplatin and short naproxen-capped peptides. This procedure boosted the radiosensitization effect of cisplatin by increasing the number of Pt-DNA adducts, arresting the cell cycle, and inhibiting cyclooxygenase-2.
  • ||||||||||  pioglitazone / Generic mfg.
    Journal:  Suppressive role of PPARγ in the IgE-dependent activation of mast cells. (Pubmed Central) -  Oct 2, 2020
    Oral administration of pioglitazone (PPARγ agonist) significantly suppressed body temperature change of mice in passive systemic anaphylaxis, supporting the inhibitory functions of PPARγ in IgE/Ag-dependent activation of MCs in vivo...Knockdown of COX-1 and/or COX-2 by siRNA showed that suppression of IgE/Ag-mediated activation was mainly dependent on COX-1. Taken together, these results indicate that PPARγ suppresses IgE/Ag-induced transactivation of cytokine genes and the Ptgs2 gene in MCs in a manner distinguishable from that of PGs.
