(Pubmed Central) - Oct 11, 2022 Tianhuang formula (TH) has been proved to have a lipid-lowering effect in which constituents of ginsenoside Rb1, ginsenoside Rg1, ginsenoside Rb, ginsenoside Re, and ginsenoside R1 from Panax notoginseng and berberine, palmatine, and coptisine from Coptis chinensis have low drug permeability, which results in poor intestinal absorption into the human body, and are thus able to come into contact with the gut microflora for a longer time...The traditional Chinese medicine TH improved NAFLD by regulating the gut microbiome, and its mechanism was related to the "Lactobacillus-5-MIAA-Nrf2" pathway. It provided a promising way for the intervention of NAFLD.

By studying the key protein expression levels of the Kelch-like ECH-associated protein 1 (Keap1)/NF-E2-related factor 2 (Nrf2) pathway, the results show that WGPs could activate Nrf2 and glutamate-cysteine ligase catalytic subunit (GCLC) up-regulation. For glutamate-cysteine ligase modifier (GCLM), WGP2 and WGP7 lead to its down-regulation, while WGP11 leads to its significant up-regulation.The present study found that peptides from wheat germ can effectively mitigate the cellular damage induced by the ɑ-gliadin peptide, which provides a new perspective for the prevention and treatment of CD.

The anti-colitic effects were significantly compromised by the co-administration of an HO-1 inhibitor.

These results elucidate a new function of SSH1 that plays a significant negative regulatory role in the Nrf2 antioxidant pathway relevant for AD and other tauopathies.

Functional genomics analysis demonstrated that the neuroprotective effects of Bach1 inhibition was due to upregulation of Bach1-targeted pathways that are associated with both Nrf2-dependent antioxidants response element (ARE) and Nrf2-independent non-ARE genes. Our findings suggest that HPPE treatment after the onset of AD pathology can reverse/slow further progression of AD-like pathology and that Bach1 inhibition is a promising therapeutic approach.

Our results suggest that IEGs may only be dysregulated immediately following exposure but could contribute to long-term detrimental effects on hippocampal neuroplasticity, which may be improved with induction of neuroprotective factors.

Journal, IO biomarker, Synthetic lethality: Novel NRF2-activated cancer treatments utilizing synthetic lethality. (Pubmed Central) - Oct 7, 2022 An important clinical feature of these tumours is their defiance to all current anti-cancer treatment regimens, highlighting the need for the development of new therapeutic strategies to target NRF2-activated cancers. In this review, we discuss the mechanisms through which acquired NRF2 hyperactivation can result in resistance of tumours to immune checkpoint inhibitor therapies in addition to classical chemotherapeutics, and propose with examples that using a synthetic lethal strategy mediated by NRF2-target gene-dependent bioactivation of prodrugs represents a promising strategy to specifically enhance toxicity to heretofore untreatable NRF2-hyperactivated human tumours.

(Pubmed Central) - Oct 5, 2022 Moreover, the modulation of Keap1 and Nrf2 as well as HO-1 upregulation by pyrogallol was reversed by pretreatment with N-acetylcysteine (NAC) and a P38 kinase inhibitor (SB203580)...Compared to untreated hUCMSCs, Sprague-Dawley (SD) rats with pyrogallol-primed hUCMSCs transplantation showed enhanced improvements in LPS-mediated lung pathological alterations, the increased lung index (lung/body ratio), apoptosis of epithelial cells, the activation of TLR4/NF-κB signaling as well as the release of pro-inflammatory mediators. Together, these results suggested that hUCMSCs with pyrogallol pretreatment enhanced the therapeutic efficacy of hUCMSCs, which may provide a promising therapeutic strategy to maximize the therapeutic efficacy of hUCMSC-based therapy for treating LPS-associated ALI.

In sum, Forsythiaside protected H9c2 cells from oxidative stress and apoptosis induced by HO by activating the Nrf2/HO-1 pathway.

Taken together, we found that ASX can protect against AILI by activating the Nrf2/HO-1 pathway, which mainly affects oxidative stress, autophagy, and ferroptosis processes, and the HMSN@ASX nanosystem can target the liver to enhance the treatment efficiency of AILI.
