    Using a Cre-LoxP recombination system to excise the mutated PolgA to stop de novo mtDNA mutations, we are studying the impact of voluntary exercise on pre-existing mtDNA mutations across different tissues. Ongoing studies utilizing methods, such as next generation sequencing, western blot, qPCR, and immunohistochemistry aim to further elucidate the role that exercise plays in counteracting the mitochondrial impairment associated with the aging process.

    To explore this hypothesis, we began by evaluating the effect of a single dose of lorcaserin, a highly selective 5HT2c agonist, on neural excitability in 26-month-old male and female mice...Overall, our data suggests that 5HT2c agonism is a promising therapeutic approach for treating age-related neural hypoexcitability and weakness. Importantly, 5HT2c agonism may be an effective strategy for treating weakness and physical frailty in the older adults, greatly improving quality of life and healthspan.

    Journal:  A class of chemical compounds enhances clustering of muscle nicotinic acetylcholine receptor in cultured myogenic cells. (Pubmed Central) -  Aug 22, 2024   
    The effects were higher than that of disulfiram, a clinically available aldehyde dehydrogenase inhibitor, which we identified to be the most competent preapproved drug to enhance ATF2-luciferase activity in the same assay system...Extensive docking simulation and binding structure prediction of the four compounds with all available human proteins using AutoDock Vina and DiffDock showed that the four compounds were unlikely to directly bind to MuSK or Dok7, and the exact target remained unknown. The identified compounds are expected to serve as a seed to develop a novel therapeutic agent to treat defective NMJ signal transmission.

    In addition, aging, low BMI, low calf circumference, smoking, depression, osteoporosis, malnutrition and malnutrition risk are risk factors for the development of sarcopenia in the older adult in China. For these high-risk populations, early identification, prevention, and intervention can be carried out to delay the occurrence and progression of sarcopenia.

    Sarcopenia prevalence was highest in individuals over 65 (62.1%) and was significantly associated with African American race (16.8%) and Asian/Pacific Islanders (3.3%) ( Table 1). Clinically, sarcopenia was associated with higher rates of hepatic encephalopathy (63.5%), ascites (2.9%), and hepatocellular carcinoma (2.1%).

    Multivariate regression analysis of patients with cirrhosis showed sarcopenic patients had higher odds of spontaneous bacterial peritonitis (Odds Ratio (OR) 1.66, 95% CI 1.31-2.10), portal vein thrombosis (OR 1.94, 95% CI 1.54-2.44), hepatocellular carcinoma (OR 1.38, 95% CI 1.06-1.78), ascites (OR 1.8, 95% CI 1.54-2.10), transjugular intrahepatic portosystemic shunt (TIPS) placement (OR 3.87, 95% CI 2.56-5.84), and liver failure (OR 2.39, 95% CI 2.01-2.84) compared to non-sarcopenic patients. There was non-significant association with encephalopathy (OR 0.79, 95% CI 0.55-1.15) and variceal bleeding (OR 1.08, 95% CI 0.83-1.40).
